The XylC-mediated aromatic metabolic network modulates the metabolic response of Pseudomonas aeruginosa to deltamethrin.

The widespread emergence of pyrethroid resistance represents a major challenge in pest and vector management. This study examined the function of the xylC-encoded benzaldehyde dehydrogenase in Pseudomonas aeruginosa under deltamethrin stress. The metabolic response of a ΔxylC mutant cultured with 50 mg/L deltamethrin was compared to the wild-type strain, and functional complementation was achieved by adding purified XylC protein. Integrated metabolomic and proteomic analyses were used to characterize the associated metabolic adaptations. Deltamethrin exposure inhibited the production of 3-(2-c

RNAi-Mediated Knockdown of the Sodium Channel Auxiliary Subunit TipE Increases Pyrethroid-Associated Mortality and Reveals Metabolic Transcriptional Responses in Megalurothrips usitatus.

Background: The bean flower thrips, Megalurothrips usitatus, is a destructive agricultural pest with increasing resistance to pyrethroid insecticides. Most mechanistic studies emphasize mutations in the pore-forming voltage-gated sodium channels α-subunit, whereas the contribution of insect-specific sodium-channel auxiliary subunits remains unclear. Methods: We used dsRNA feeding, pyrethroid bioassays, RNA sequencing, and RT-qPCR validation to evaluate the role of temperature-induced paralytic E (TipE) in M. usitatus. Results: The optimized RNAi condition was 400 μg/mL of dsTipE for 48 h, whic

Elevated Temperature Decreases the Insecticidal Efficacy of Yahe LN Insecticide-Treated Nets: Implications for Transport and Storage.

A recent durability study has revealed that Yahe LN® insecticide-treated nets (ITNs; Fujian Yamei Industry & Trade Co., LTD, Fuzhou, China) distributed in Papua New Guinea (PNG) in 2021 did not retain their insecticidal efficacy after several months post-distribution. Insecticide-treated nets are frequently shipped and stored in containers where they are exposed to elevated temperatures. When used in the tropics, ITNs may continue to be exposed to high ambient temperatures. The aim for the present study is to better understand the potential impact of elevated temperature during transport and s

Predicting slug injury in corn and the role of carabid natural enemies in regulating slug populations.

While integrated pest management and biological control may be valuable approaches to managing slugs in field corn, their use is complicated by unclear thresholds and extensive use of preventative insecticides, as well as confusion regarding what predatory species contribute to biocontrol. To better understand slug injury and biocontrol in field corn, we conducted a comparison between neonicotinoid seed treated, in-furrow pyrethroid (IFP) treated, and untreated plots at 3 farms in Maryland for 3 years, measuring slug activity-abundance, seedling injury, yield, the predator community, and preda

A commercial insecticide-induced neurotoxicity and snake venom nerve growth factor-inspired peptides-mediated neuroprotection in Caenorhabditis elegans: Mechanistic, safety, and pharmacokinetic (in vivo imaging) evaluation of peptides.

Agriculture workers are especially at risk from insecticide (INSECT) exposure, which is associated with neurological disorders, such as Parkinson's disease (PD). The present study evaluated the mechanism of neurotoxicity induction in a commercial insect containing chlorpyrifos (50%) and cypermethrin (5%) in Caenorhabditis elegans models, as well as the neuroprotective efficacy of two custom peptides (CPs), HNP (heptadeca-neuropeptide) and TNP (trideca-neuropeptide), derived from snake venom nerve growth factor. CPs significantly mitigated INSECT-induced neurotoxicity by preventing chemosensory

A complete genome sequence enables efficient genetic mapping of adaptive traits in a phytoseiid predatory mite (Amblyseius andersoni).

Phytoseiid mites are important biological control agents in Integrated Pest Management (IPM), yet the genetic architecture underlying their adaptive traits remains largely unknown due to limited genomic and genetic resources. Here, we establish an integrated genetic framework for Amblyseius andersoni, generating a genome sequence and developing a high-resolution genetic mapping approach. The genome was assembled using long-read sequencing, yielding a highly contiguous assembly of approximately 202 Mb with 18,923 predicted protein-coding genes and high completeness. We then dissected resistance

Insecticidal efficacy of residual spraying with deltamethrin-clothianidin (Fludora® Fusion) in Papua New Guinea.

BACKGROUND: In Papua New Guinea (PNG), insecticide-treated nets leave considerable gaps in protection from Anopheles mosquito bites and, as a result, malaria cases are increasing. Residual spraying (RS) may be an effective complementary malaria control strategy. To understand the potential impact of RS and guide RS implementation, it is important to quantify the duration of insecticidal efficacy of RS formulations. The materials used to construct houses in rural PNG differ significantly from the African study sites where most such data have been generated. This study investigated determinants

Environmental contaminants drive insecticide resistance in Anopheles mosquitoes in Ghana.

Insecticide-based interventions such as IRS and LLINs have significantly reduced malaria transmission globally. However, their sustainability is increasingly threatened by insecticide resistance. While insecticide and pesticide use are known resistance drivers, the role of environmental contaminants remains underexplored. This study investigated the impact of environmental contaminants on insecticide resistance by sampling Anopheles larvae from six sites in Ghana, including petroleum spill sites, mining areas and industrial zones. WHO bioassays revealed resistance to clothianidin (Mortality ra

Biochemical and molecular disruptions in hook snout carp induced by co-exposure to lambda-cyhalothrin and difenoconazole.

Pesticides are recognized as prominent toxicants in aqueous ecosystems, which frequently impact aquatic life. The pyrethroid insecticide lambda-cyhalothrin (LDC) and the triazole fungicide difenoconazole (DFC) are frequently co-detected in many water sources. However, the joint harmful effects of these chemicals on fish are still poorly understood. In this study, a 30-day co-exposure experiment was conducted using LDC and DFC on the hook snout carp (Opsariichthys bidens) to assess their toxic impacts on hepatic tissues. Biochemical assays demonstrated a significant increase in catalase (CAT) a

A noninferiority cluster randomised evaluation of a broflanilide indoor residual spraying insecticide, VECTRON T500, for malaria vector control in Tanzania.

Effective malaria vector control is being undermined by the rapid spread of insecticide resistance. VECTRON T500, a new indoor residual spraying (IRS) product containing the active ingredient broflanilide as a 50% wettable powder (WP), was previously shown to be efficacious in experimental hut trials. A two-arm non-inferiority cluster randomized controlled community trial was conducted in Muheza District, Tanga Region, Tanzania. VECTRON T500 was compared to the IRS product Fludora Fusion (clothianidin 50% + deltamethrin 6.25% WP-SB). Sixteen village clusters were pair-matched on baseline vecto

Developmental toxicity of formulated insecticide mixture containing imidacloprid and beta-cyfluthrin in fish: Insights using zebrafish.

Insecticides are critical in controlling pests and disease vectors. However, there is still a lack of ecotoxicological studies using commercial formulations of insecticides containing active ingredients. The study aimed to evaluate the developmental toxicity of a commercial insecticide mixture (imidacloprid [IMI] + beta-cyfluthrin [β-CYF]). Mortality, hatching rate, spontaneous contraction, heartbeat, morphological changes, reactive oxygen species (ROS), skeletal development, and locomotor behavior of zebrafish were analyzed. Embryos were exposed to imidacloprid (IMI) and β-cyfluthrin (β-CYF)

Knockdown-resistant mutations in head lice (Pediculus humanus capitis) collected from schoolchildren in Riyadh, Saudi Arabia.

Head lice infestation remains one of the most common child problems. This problem is not only attributed to the ability of head lice to spread rapidly but also because of the head lice resistance that develops from incomplete or improper treatment. Pyrethroids are a group of medications that have been widely used for the treatment of head lice. However, in recent years, several countries reported knockdown resistance (kdr). The aim of this study is to evaluate the frequency of pyrethroid-resistant mutations in Riyadh City, Saudi Arabia. To do that, Sanger sequencing was employed to find the fr

Characterizing the escalation of pyrethroid resistance and its impact on bed nets efficacy alongside molecular basis in Anopheles funestus from Cameroon.

BACKGROUND: Malaria prevention still relies greatly on vector control interventions. However, increasing levels of resistance to pyrethroids across Africa have significantly reduced the efficacy of pyrethroid-based interventions leading to an increase of malaria burden. Consequently, it is imperative to characterize the extent and molecular basis of this resistance. METHODS: This study was conducted from 2020 to 2021 in a South-North transect across Cameroon. WHO tube assay was used to assess the susceptibility profile of Anopheles funestus to the four main classes of insecticides. The effica

Impact of pyriproxyfen on the reproduction of pyrethroid-resistant Anopheles gambiae s.l. populations in benin: what implications for vector control?

BACKGROUND: The efficacy of LLINs is compromised by the emergence of pyrethroid resistance in malaria vectors. New insecticides that target other sites of action in the mosquito, including pyriproxyfen (PPF), are needed to maintain this efficacy. METHODS: Various populations of An. gambiae s.l. collected from April to November 2022 in 4 Beninese communes and the susceptible strain (Kisumu) were used to assess their susceptibility to PPF via CDC bottle tests and to assess the bioefficacy of Royal guard® (RG) alpha-cypermethrin + pyriproxyfen-impregnated nets via WHO cone tests. Characteristics of malaria vector populations and transmission before a randomised controlled trial assessing the efficacy of next-generation insecticide-treated nets in Côte d'Ivoire.

BACKGROUND: The World Health Organization (WHO) recommends mass distribution of insecticide-treated nets (ITNs) to prevent malaria transmission. Unfortunately, resistance to pyrethroids affects the efficacy of standard ITNs. To overcome this resistance and continue to protect the population, the WHO has recommended new types of ITNs that combine a pyrethroid insecticide with either a synergist (PBO) or a second insecticide, such as chlorfenapyr. This study examines the baseline characteristics of malaria vectors prior to the distribution of three types of insecticide-treated nets as part of a

Esterase-Mediated Pyrethroid Resistance in Populations of an Invasive Malaria Vector Anopheles stephensi from Ethiopia.

BACKGROUND: The swift expansion of the invasive malaria vector Anopheles stephensi throughout Africa presents a major challenge to malaria control initiatives. Unlike the native African vectors, An. stephensi thrives in urban settings and has developed resistance to multiple classes of insecticides, including pyrethroids, organophosphates, and carbamates. METHODS: Insecticide susceptibility tests were performed on field-collected An. stephensi mosquitoes from Awash Sebac Kilo, Ethiopia, to assess insecticide resistance levels. Illumina RNA-seq analysis was then employed to compare the transcr
