In situ photocrosslinking ROS-adaptive caffeoyl chitosan/boronic acid-grafted gelatin hydrogels for treatment of combined radiation-burn injury.

Combined radiation-burn injury (CRBI) is a serious wound that is difficult to treat and typically results from radiation therapy, nuclear explosions, or nuclear accidents, where ionizing radiation and thermal burns usually occur simultaneously or sequentially. Excessive expression of reactive oxygen species (ROS) contributes to CRBI. Caffeic acid (CA) is a common natural antioxidant polyphenol whose clinical application is limited by its poor solubility and low stability. Here, we develop an in situ photocrosslinking caffeoyl chitosan/boronic acid-grafted gelatin hydrogel to treat CRBI. Caffeo

Ultrasound-based monitoring of early mesenchymal stem cell therapy mitigates severe radiation-induced soft tissue injury in minipigs.

PURPOSE: Cutaneous radiation syndrome (CRS) arises from high dose-ionizing radiation exposure and remains a major clinical challenge owing to limited effective therapies. This study investigated the time-dependent therapeutic potential of human umbilical cord blood-derived mesenchymal stem cells (hUCB-MSCs) for treating radiation-induced soft tissue injury in a Göttingen minipig model. MATERIALS AND METHODS: Six minipigs were locally irradiated with 100 Gy gamma radiation on each thigh. Two animals served as irradiated group (IR group), whereas four received local intradermal hUCB-MSC injecti

Inhalable ROS-Responsive Nanospray Activates PPAR-γ to Restore Macrophage Mitochondrial Homeostasis and Attenuate Radiation-Induced Lung Injury.

Radiation-induced lung injury (RILI) is a major complication of nuclear radiation exposure and thoracic radiotherapy, driven in part by a vicious cycle of oxidative stress and mitochondrial dysfunction. Peroxisome proliferator-activated receptor gamma (PPAR-γ), a key regulator of mitochondrial homeostasis and inflammatory resolution, therefore represents a potential therapeutic target, yet multi-omics analyses revealed that this immunometabolic checkpoint remains functionally constrained in irradiated macrophages. To reactivate this pathway, we developed an inhalable, ROS-responsive nanospray

Association of neurite orientation dispersion and density imaging with cognitive impairment in nasopharyngeal carcinoma patients with radiation-induced temporal lobe injury.

BACKGROUND: To evaluate whether neurite orientation dispersion and density imaging (NODDI) can reflect cognitive impairment (CI) in nasopharyngeal carcinoma (NPC) patients with late-delayed radiation-induced temporal lobe injury (RTLI). METHODS: In this prospective study, 90 NPC patients with RTLI underwent structural MRI, conventional diffusion-weighted imaging, and NODDI. Cognitive function was assessed using the Montreal Cognitive Assessment (MoCA; score < 26 indicated CI). NODDI-derived parameters-intra-neurite volume fraction (ICVF), isotropic volume fraction (ISOVF), and orientation dis

Carboxymethyl chitosan crosslinked self-assembled collagen hydrogel protects against radiation-induced skin injury by modulating iron homeostasis.

Radiation-induced skin injury (RSI) is a critical complication of radiotherapy, affecting over 95% of patients and causing severe morbidity with treatment interruptions. Clinical therapies fail to address its multifactorial pathogenesis, including vascular dysfunction, oxidative stress, and chronic inflammation. Current clinical chitosan-based wound care materials face challenges like rapid degradation and insufficient mechanical stability. To tackle these, we developed a self-assembled carboxymethyl chitosan-crosslinked collagen hydrogel (Col+CMC) using traceable type I collagen from shad fis

Loss of Mist1 alters the characteristics of Paneth cells and impacts the function of intestinal stem cells in physiological conditions and after radiation injury.

Intestinal stem cells (ISCs) and Paneth cells (PCs) reside at the bottom of the crypts of Lieberkühn in the small intestine. Recent studies have shown that the transcription factor Mist1, also named BHLHA15, plays an important role in the maturation of PCs. Since there is an intimate interaction between PCs and ISCs, we speculated that the loss of Mist1 could impact these two neighboring cell types. Here, we report that mice lacking Mist1 had fewer but larger PCs with shrunken secretory granules, accompanied by an increase in goblet cells and tuft cells. Mist1 loss significantly decreased the

A multi-omics approach exploring the gut-liver axis following combined radiation exposure and burn injury in a Sinclair minipig model.

While radiation and burn injury have distinct local and systemic effects, they both negatively impact intestinal permeability/function. In a nuclear attack, combined thermal burns and radiation exposure (e.g., combined injury (CI)) would be a dominant injury pattern. Despite this, how burns affect gastrointestinal acute radiation syndrome, and vice versa, has been largely unstudied. Next-generation sequencing has revealed a strong bidirectional link between the liver and gut. Here, we used a porcine model to evaluate the impact of burn, radiation, and CI on the gut microbiota and liver transcr

Discovery the Mechanism of Qingdi Mixture for Radiation-Induced Lung Injury Based on Network Pharmacology, Clinical Retrospective Analysis and Experimental Validation.

Radiation-induced lung injury (RILI) can be caused by thoracic tumor radiotherapy. Qingdi mixture (QDM) has been routinely used in preventing RILI during tumor radiotherapy. However, the molecular mechanisms of QDM remain to be fully elucidated. Initially, we employed network pharmacology to identify potential therapeutic targets and their related signaling pathways. Secondly, molecular docking was applied to validate the interactions between the QDM components and hub targets. Furthermore, we retrospectively collected clinical data from RILI patients who received basic therapy combined with Q

Reduced symptoms of late radiation tissue injury of the vagina after treatment with hyperbaric oxygen therapy: A retrospective analysis of 19 patients.

INTRODUCTION: Hyperbaric oxygen therapy (HBOT) is a well-established treatment for late radiation tissue injury (LRTI) of the pelvis, such as radiation-cystitis and -proctitis, but not for LRTI of the vagina. This study aims to describe the outcomes of patients with vaginal symptoms after HBOT. METHODS: The records of all patients with LRTI of the vagina, referred for HBOT from a tertiary hospital, between 2010 and 2020 were retrospectively analyzed. Patients with a non-vaginal primary complaint, fistulas, or incomplete HBOT treatment (<20 sessions) were excluded. Outcomes included patient- a

Radiation-induced brain injury in non-human primates: A dual tracer PET study with [11C]MPC-6827 and [11C]PiB.

Radiation-induced brain injury (RIBI) and Alzheimer's disease (AD) share key pathological features, including β-amyloid (Aβ) plaque formation and microtubule (MT) destabilization, both contributing to neurodegeneration. This pilot study assessed Aβ deposition and MT stability in non-human primates (NHPs) exposed to ionizing radiation, utilizing [11C]PiB and [11C]MPC-6827 PET imaging to explore neurodegenerative mechanisms. Fourteen rhesus macaques, seven irradiated and seven controls underwent PET imaging. Tracers were synthesized and brain regions (ex. cingulate, hippocampus, and occipital lo

Fetal radiation during acetabular and pelvic ring fixation: Which injury patterns dictate dangerous levels of fluoroscopic cumulative dose.

BACKGROUND: Gravid females with pelvic fractures are rarely encountered by the orthopaedic trauma surgeon. The initial injury can be detrimental to the pregnant patient, but an unnecessary "second hit" from surgery could also contribute to the outcome of the fetus. Understanding the surgical risks for this unique patient population requires knowledge about the negative effects of anesthesia, surgical exposures, and radiation. Radiation levels below 5 rad are negligible to the fetus, while doses above 15 rad significantly increase the harmful fetal effects. The purpose of this study was to iden

Small extracellular vesicles derived from umbilical cord mesenchymal stem cells alleviate radiation-induced cardiac organoid injury.

BACKGROUND: Radiation-induced heart disease (RIHD) is one of the most serious complications of radiation therapy (RT) for thoracic tumors, and new interventions are needed for its prevention and treatment. Small extracellular vesicles (sEVs) from stem cells have attracted much attention due to their ability to repair injury. However, the role of umbilical cord mesenchymal stem cell (UCMSC)-derived sEVs in protecting cardiac organoids from radiation-induced injury and the underlying mechanisms are largely unknown. METHODS: A radiation-induced cardiac organoid injury model was established by us

Predictive accuracy of machine learning for radiation-induced temporal lobe injury in nasopharyngeal carcinoma patients: a systematic review and meta-analysis.

BACKGROUND: Radiotherapy is a common treatment for nasopharyngeal carcinoma (NPC) but can cause radiation-induced temporal lobe injury (RTLI), resulting in irreversible damage. Predicting RTLI at the early stage may help with that issue by personalized adjustment of radiation dose based on the predicted risk. Machine learning (ML) models have recently been used to predict RTLI but their predictive accuracy remains unclear because the reported concordance index (C-index) varied widely from around 0.31 to 0.97. Therefore, a meta-analysis was needed. METHODS: The PubMed, Web of Science, Embase,

Colon-Targeted Release of Gel Microspheres Loaded with Antioxidative Fullerenol for Relieving Radiation-Induced Colon Injury and Regulating Intestinal Flora.

Radiation-induced colitis is a serious clinical problem worldwide. However, the current treatment options for this condition have limited efficacy and can cause side effects. To address this issue, colon-targeted fullerenol@pectin@chitosan gel microspheres (FPCGMs) are developed, which can aggregate on colon tissue for a long time, scavenge free radicals generated in the process of radiation, and regulate intestinal flora to mitigate damage to colonic tissue. First, FPCGMs exhibit acid resistance and colon-targeted release properties, which reduce gastrointestinal exposure and extend the local
