Hypertrophic cardiomyopathy (HCM) variants in genes encoding the myosin heavy chain (MHC) (MYH7), myosin light chains (MYL2 and MYL3), and cardiac myosin binding protein-C (cMyBP-C, MYBPC3) lead to cardiac hypertrophy, with abnormal contractility, relaxation, and energy consumption. Here, we defined the structural consequences of pathogenic and benign missense variants in these genes by mapping 233 variants (MYH7, n = 175; MYBPC3, n = 41; MYL2, n = 12; MYL3, n = 5) onto a cryo-EM-based atomic model of the human cardiac thick filament. We identified HCM variants residing in 30 molecular interfa

Toxoplasma gondii, an intracellular parasite with a global distribution, causes toxoplasmosis, resulting in significant economic losses. Drug resistance and vaccine deficiency have spurred interest in identifying novel intervention targets, including autophagy and autophagy-related proteins (ATGs). ATG5 is an important member of ATGs that plays a crucial role in disrupting the T. gondii parasitophorous vacuole membrane (PVM). However, its effects on the parasite's intracellular lifecycle warrant further investigation. In this study, the transgenic Vero cells were constructed using a lentivirus

PURPOSE: Clinical fieldwork in genetic counseling education is constrained by limited training sites, shortages of clinical supervisors, and administrative burdens. Simulation-based learning, including role-play, has therefore been used to support development of counseling competencies. This study described the development, implementation, operational structure, and feasibility of a role-play-based practicum in a graduate genetic counseling program.
METHODS: This retrospective descriptive report was based on Genetic Counseling Practicum, a spring 2024 course for 11 third-semester students in

Ran is a small GTPase of the Ras superfamily that governs nucleocytoplasmic transport, including that of miR-126, a microRNA that supports the homeostasis and expansion of leukemia stem cells (LSCs). Ran binds to Exportin 5 to facilitate the transport of precursor (pre)-miR-126 across the nuclear membrane for its maturation. Our goal is to inhibit Ran to prevent transport of pre-miR-126 to the cytoplasm. Like other Ras family proteins, targeting Ran with small molecules is challenging due to its relatively flat surface and lack of binding cavities. Ran's activity is regulated by a long and dis

Vascular leakage (VL) is a severe pathology occurring in a broad range of scenarios, e.g., during sepsis, cytokine storms, or as side effect of immunotherapies. Its severity is underlined by the high lethality rate of 20-30% for the systemic capillary leakage syndrome. While many compounds are reported to affect endothelial cell (EC)-activation, exact mechanisms behind VL remain unclear. We analyzed activation, viability, cytokine secretion, and relative permeability of human umbilical vein endothelial cells (HUVECs) upon treatment with 16 different stimuli. Relative HUVEC-permeability was ass

Feline coronaviruses (FCoVs) are divided into two groups: feline infectious peritonitis virus (FIPV) and feline enteric coronavirus (FECV). FIPV is responsible for the severe disease known as feline infectious peritonitis, while FECV typically causes mild symptoms, such as diarrhea, and often does not lead to any disease at all. Currently, it is not possible to distinguish between FIPV and FECV at the molecular level. Therefore, there is an urgent need to understand the molecular features of FIPV. Here, we generated a recombinant virus by replacing the ORF1ab region and the coding sequence for

Pneumonia caused by Staphylococcus aureus infection has consistently been a significant cause of morbidity and mortality worldwide. Extensive research to date indicates that N6-methyladenosine (m6A) modification plays a crucial role in the development and progression of various diseases. However, it remains unknown whether the m6A modification affects the progression of bacterial pneumonia. To explore this question, we assessed the levels of m6A as well as the expression of methyltransferases (METTL3 and METTL14), demethylase fat mass and obesity-related protein (FTO), and methylation reader p

Hair cells (HCs) within the inner ear cochlea are specialized mechanoreceptors required for hearing. Cochlear HCs are not regenerated in mammals, and their loss is a leading cause of deafness in humans. Cochlear supporting cells (SCs) in newborn mice have the capacity to regenerate HCs, but persistent Notch signaling, presumably activated by SC-specific Notch ligand Jagged1 (JAG1), prevents SCs from converting into HCs. Here, employing an organoid platform, we show that while JAG1 participates in HC-fate repression, JAG1's primary function is to preserve the "progenitor-like characteristics" o

Identifying risk factors that pre-dispose people on peritoneal dialysis (PD) to develop exit site infections (ESIs) may help improve prevention and treatment. Given the differences anatomically, hormonally and of the microbiota profile between males and females, we hypothesised that there is a difference in ESI incidence, outcomes, and the epidemiology of organism-specific ESI between males and females. This study was a retrospective case note review of all PD patients at our centre between 2012 and 2024. Of the 486 patients on PD, 202 (42%) were female and 273 positive swabs from 151 patients

NR6A1 is a member of the nuclear receptor superfamily of ligand-activated transcription factors and can bind to conserved DNA sequences, acting as a transcriptional repressor. NR6A1 has been reported to promote prostate cancer, gastric cancer, and testicular germ cell tumor progression. In the present study, we confirmed the oncogenic role of NR6A1 in several types of cancer cells, including HeLa, TFK1, and A549 cells, and revealed that NR6A1 knockdown led to a decrease in lung adenocarcinoma cell proliferation, a reduction in glucose consumption, a reduction in lactic acid production, and dec

Identification of host factors that play a key role in viral replication is of great importance for antiviral development. Metabotropic glutamate receptor subtype 2 (mGluR2) is the receptor to trigger clathrin-mediated endocytosis (CME), the major pathway by which influenza virus enters cells. However, other host factors almost certainly involved in the influenza virus CME are largely unknown. Here, we found that the four-transmembrane protein claudin-11 plays an integral part in influenza virus CME. Claudin-11 promotes the dissociation of KCa1.1 (potassium calcium-activated channel subfamily

INTRODUCTION: The extent, activity, and effects of renal involvement in systemic lupus erythematosus are conditions difficult to diagnose and a renal biopsy is usually required for such purpose. Non-invasive methods are then of high interest to better understand lupus nephritis (LN) features and prognosis. We have evaluated VEGF and suPAR levels as non-invasive biomarkers of disease activity in LN.
METHODS: This cross-sectional study enrolled patients with LN aged 18 years or older of both genders who were assisted at the Outpatient Clinic of Glomerular Diseases of the Division of Nephrology

The T-box transcription factor TBX1 is expressed in the cardio-pharyngeal mesoderm. The correct cell fate decisions of cardio-pharyngeal mesoderm cells are critical, as any defect in this process can alter second heart field morphogenesis and lead to cardiac outflow tract and pharyngeal apparatus defects. The second heart field plays a crucial role in cardiac development by incorporating cardiac progenitors into the heart. It is also the major gene implicated in 22q11.2 deletion (or DiGeorge) syndrome, a primary genetic cause of congenital heart defects associated with hypoplasia of the cardia

BACKGROUND: Hepatic fibrosis increases with aging, but its physiological progression and underlying mechanisms remain poorly defined. Given that CEACAM1 repression causes metabolic dysfunction and liver injury, the current studies investigated whether it mediates age-related hepatic fibrosis.
MATERIALS AND METHODS: The metabolic phenotype, histological, immunochemical and Western blot analyses were performed in male C57BL6/J wild-type and LCC1 mice with liver-specific CEACAM1 overexpression at 2-17 months of age.
RESULTS: Progression of metabolic dysfunction during physiological aging began

BACKGROUND: Cryptosporidium parvum is a common protozoan pathogen responsible for moderate to severe diarrhea in humans and animals. Parasite invasion and egress cause damage to intestinal epithelial cells, which is mediated by a variety of secretory proteins from several unique organelles, such as micronemes. Previous spatial proteomic analysis has identified insulinase-like protease 6 (INS6) as a putative microneme protein in C. parvum. However, the functional contribution of INS6 to Cryptosporidium pathogenicity remains poorly characterized. In this study, we used genetic manipulation techn

BACKGROUND: Decidualization is a critical step in establishing pregnancy in mammals. Successful decidualization depends on intricate gland-stromal crosstalk. Clusterin (Clu) is a ubiquitously secreted protein in physiological fluids that is involved in numerous physiological functions. However, the role of Clu in decidualization is not fully understood.
RESULTS: In this study, we examined the expression pattern of Clu during early pregnancy in mice and explored its potential function in decidualization. Our results revealed that Clu was expressed in the uterine glands on Days 1-2 of early pre

Fatty liver disease, a condition characterized by fatty degeneration of the liver, mainly classified as non-alcoholic fatty liver disease (NAFLD) and alcoholic liver disease (ALD), has become a leading cause of cirrhosis, liver cancer and death. The gut-liver axis is the bidirectional relationship between the gut and its microbiota and its liver. The liver can communicate with the gut through the bile ducts, while the portal vein transports the products of the gut flora to the liver. The intestinal flora and its metabolites directly and indirectly regulate hepatic gene expression, leading to a

OBJECTIVE: Social participation is an important aspect associated with health-related outcomes in chronic diseases. However, little is known about the factors that may affect participation in patients with rheumatoid arthritis (RA). We aimed to examine whether pain, fatigue, anxiety, depression, and illness perception are associated with social participation in patients with RA when controlled for clinical and sociodemographic variables. We also analysed the mediating role of illness perception in the association between physical and psychological variables on social participation.
METHOD: We

Dendrites are injured in a variety of clinical conditions such as traumatic brain and spinal cord injuries and stroke. How neurons detect injury directly to their dendrites to initiate a pro-regenerative response has not yet been thoroughly investigated. Calcium plays a critical role in the early stages of axonal injury detection and is also indispensable for regeneration of the severed axon. Here, we report cell and neurite type-specific differences in laser injury-induced elevations of intracellular calcium levels. Using a human KCNJ2 transgene, we demonstrate that hyperpolarizing neurons on
