Synergistic effect of 5-HT4 receptor activation and phosphodiesterase type 7 inhibition on object recognition and working memory performances in adult mice.

CONTEXT: Low-dose multitarget pharmacological strategies are viewed as promising approaches for managing cognitive impairment in aging. Here … ecognition test and the spontaneous alternation test under conditions of spontaneous forgetting and scopolamine-induced amnesia. RESULTS: Dose-response studies revealed that RS67333 (0.5 and/or 1 mg/kg) and BRL50481 (2.5 and 5 mg/kg) significantly prolonged recognition memory trace and reversed scopolamine-induced deficits in both recognition and working mem …

COL1A1 regulates the progression of dry eye disease through metabolic reprogramming and inflammatory responses via the HIF1A/HK2 pathway.

BACKGROUND: Dry eye disease (DED) is a prevalent ocular surface disorder with complex pathogenesis involving glycolytic reprogramming and in … ssion and COL1A1-HIF1A co-overexpression were performed. In vivo, a DED mouse model was induced via scopolamine injection and low humidity, with adeno-associated virus (AAV)-mediated Col1a1/Hif1a overexpression. Assessments included qRT-PCR, Western blotting, ELISA, lactate assay, extracellular acidification rate (ECAR) measurement, transepithelial electrical …

μOPTO: A microfluidic paper-based optoelectronic tongue as presumptive tests for the discrimination of alkaloid drugs for forensic purposes.

Natural and synthetic alkaloids are widely used for several applications, ranging from clinical purposes to criminal activities. Presumptive … icators in lieu of one specific test and capable of discriminating 8 different alkaloid drugs (i.e. scopolamine, atropine, cocaine, morphine, ephedrine, caffeine, dipyrone and alprazolam) used for recreational, criminal and medical purposes. Neurobehavioral effects of liraglutide and sitagliptin in experimental models.

Glucagon-like peptide (GLP-1) receptor agonists and dipeptidyl peptidase 4 (DPP-4) inhibitors are two currently approved therapies for type … in EPM and Morris water maze (MWM) following memory deficit induced by pentylenetetrazole (PTZ) or scopolamine. In tail-flick test sitagliptin (6 mg/kg) produced transient nociceptive effect. Liraglutide (200 µg/kg) reduced peripheral square crossings by rats in open field test as well as reduced closed arm entries in the EPM, indicating a decline in explora …

Transepidermal water loss with and without sweat gland inactivation.

The influence of eccrine sweating on transepidermal water loss (TEWL) was investigated. TEWL was simultaneously measured on both forearms, with and without topical inactivation of the eccrine sweat glands by 0.3 ml of 0.5% aqueous scopolamine hydrobromide (HBr), applied under 1 h occlusive patches. The degree of sweat inhibition, after exercise, was measured at 2, 3 and 4 h after patch removal. In 42 out of 44 subjects, complete sweat inhibition (on exercise) was achieved only at 4 h after removal. After a 15-min rest in a room at 20 degrees C, the pre-exercise TEWL values (at 4 h) on the trea…

Effects of Anticholinergics on Death Rattle: A Systematic Review and Network Meta-Analysis.

Background: Anticholinergics have been used to treat death rattle (DR) in dying patients with palliative care. However, the effect of anticholinergics is still controversial. No quantitative summary of their effects is reported. Objective: This study aimed to systematically review and quantitatively synthesize the effect of anticholinergics on DR treatment and prophylaxis. Design: A systematic search was performed in the electronic databases (PubMed, Embase®, and Cumulative Index to Nursing and Allied Health Literature [CINAHL]) from inception to October 2021. Studies conducted to determine th

Effects of hyoscine and meciozine on vigilance and short-term memory.

An investigation was made of the effects of 1 mg. hyoscine hydrobromide and of 50 mg. meclozine hydrochloride on the performance of tasks of vigilance and short-term memory. In further trials the effects of these drugs were measured after ingestion of 32 g. ethyl alcohol. Efficiency was impaired by hyoscine taken alone, but meclozine alone had no significant effect. The effect of hyoscine was substantially increased by the presence of alcohol and under these conditions the effect of meclozine was equally great. The vigilance task appeared to be more sensitive to the effects of the drugs than t

Modulatory effects of α7-nicotinic cholinergic receptors on perceptual sensitivity in a visual signal detection task.

Network pharmacology, molecular docking and In vivo validation reveal the neuroprotective effects of TT-TeMac™ against cholinergic dysfunction and hippocampal lesions.

AD is a neurodegenerative disorder marked by progressive cognitive decline, particularly memory impairment, largely driven by cholinergic dy … analyses and confirmed by molecular docking using MOE. The in vivo validation used a rat model with scopolamine-induced cholinergic dysfunction (1 mg/kg bw/day, ip) for 7 days. Subsequently, behavioral (NOR and MWM), biochemical (AchE and BuhE) and histological (H&E and CV) analyses were performed. Seven compounds were identified in TT-TeMac™ (terminolic acid …

Orexin-A attenuated motion sickness through modulating neural activity in hypothalamus nuclei.

BACKGROUND AND PURPOSE: We evaluated the hypothesis that central orexin application could counteract motion sickness responses through regul … B-334867 and TCS-OX2-29 (OX2 antagonist). The efficacy of intranasal application of orexin-A versus scopolamine on motion sickness symptoms in cats was also investigated. KEY RESULTS: Orexin-A (i.c.v.) dose-dependently attenuated motion sickness-related behavioural responses and hypothermia. Fos expression was inhibited in the ventral part of the dorsomedial …

Investigation of Antistress and Antidepressant Activities of Synthetic Curcumin Analogues: Behavioral and Biomarker Approach.

Depression is a serious psychiatric disorder that affects millions of individuals all over the world, thus demanding special attention from … suspension tests (TSTs) as depression mice models. Biochemical examinations were performed after a scopolamine-induced stress model by decapitating the mice, and brain tissues were isolated for biochemical assessment of catalase (CAT), superoxide dismutase (SOD), glutathione (GSH), and malondialdehyde (MDA). The curcumin analogue C2 exhibited higher DPPH (2, …

In Vivo Pharmacological Comparison of TAK-071, a Positive Allosteric Modulator of Muscarinic M1 Receptor, and Xanomeline, an Agonist of Muscarinic M1/M4 Receptor, in Rodents.

Activation of the M1 muscarinic acetylcholine receptor (M1R) may be an effective therapeutic approach for Alzheimer's disease (AD), dementia … uppressed only MK-801-induced hyperlocomotion. In a previous study, we showed that TAK-071 improved scopolamine-induced cognitive deficits in a rat novel object recognition task (NORT) with 33-fold margins versus cholinergic side effects (diarrhea). Xanomeline also improved scopolamine-induced cognitive impairments in a NORT; however, it had no margin versus …

Neuropharmacological profile of aqueous extract of Anaphe venata larva (Notondotidae) in rats.

Consumption of Anaphe larva had been reported to cause seasonal ataxia and impaired consciousness. Therefore this study examined the neuropharmacological and mechanism(s) of action of aqueous extract of Anaphe venata in rats. Behavioural effects namely rearing, stretching, sniffing and ataxia were determined after the intraperitoneal administration of aqueous extract of Anaphe larva in rats. Animals were divided into groups and graded doses (100, 200 and 400 mg/kg, i.p.) of extract were administered. The control group was administered normal saline (vehicle). The effects of scopolamine (3 mg/k…
