Granulocyte changes in infectious mononucleosis.

Various changes in the granulocytes have been investigated in a large series of patients with infectious mononucleosis. A high proportion of cases regularly show a neutropenia with a shift to the left during the first three to four weeks of the disease; these changes, though transient, may be profound. The band cells present during the acute phase of the disease also show certain alterations in cytochemical staining and low alkaline phosphatase scores are common at this time. The maturation and release of leucocytes from the bone marrow is apparently normal. Theoretical mechanisms for the neut

Ultrastructural immuno-localization of tropoelastin in the chick eye.

Immuno-gold labeling at the electron-microscopy level was used to investigate the distribution of tropoelastin in the chick eye. Intense staining was found in the amorphous part of mature elastic fibers in different regions of the organ. In elaunin fibers, both the amorphous core and the surrounding microfibrils were clearly labeled. In addition, reactive sites were detected in the oxitalan fibers of the stroma- of the cornea and in Descemet's membrane, which showed a gradient of reactive sites increasing from the center toward the periphery. Oxitalan fibers of the stroma often fused with Desc

Autosomal dominant polycystic kidney disease in Persian and Persian-cross cats.

A form of autosomal dominant polycystic kidney disease (ADPKD) similar in clinical features to human ADPKD occurs in the Persian cat. We characterized the morphologic and immunohistochemical features of this disease in a colony of affected cats. Complete postmortem examinations were performed on 11 normal and 22 affected cats ranging in age from 3 months to 10 years. Kidneys were evaluated by gross and histologic examinations, ultrastructure, lectin staining, bromodeoxyuridine immunochemistry for labeling index and immunochemistry for distribution of Na/K ATPase. Feline ADPKD was characterized

Atypical micromorphology and uncommon location of cryptococcosis: a histopathologic study using special histochemical techniques (one case report).

Here we report an unusual case of disseminated cryptococcosis in a patient with AIDS. Although typical Cryptococcus neoformans micromorphology was observed in tongue biopsy, cervical lymph node examination revealed atypical histopathologic findings. These included pseudohyphae, chains of budding yeasts and structures resembling germ tubes. Cryptococcus neoformans infection in supraclavicular lymph nodes was also confirmed by culture. The importance of using special histochemical techniques-Mayer's mucicarmine stain for mucicarminophilic capsule and Grocott's silver stain-in the diagnosis of cr

Conjunctival oculosporidiosis in east Africa caused by Rhinosporidium seeberi.

BACKGROUND AND OBJECTIVE: Conjunctival rhinosporidiosis is an infectious disease rarely recorded outside the Indian subcontinent. The disease is caused by Rhinosporidium seeberi, an endosporulating microorganism of uncertain taxonomic classification. We report a series of cases manifesting this infection. MATERIAL AND METHODS: The demographic, clinical, and histopathologic data of 14 cases of conjunctival rhinosporidiosis on record at our Ophthalmic Pathology Laboratory were reviewed. RESULTS: All cases were from East Africa; 10 were from Malawi and 4 from Kenya. Ten specimens were from male

WAFMGAN: A frequency-aware CycleGAN for structure-consistent virtual staining of transmission electron microscopy images.

Conventional transmission electron microscopy (TEM) staining improves image contrast but requires additional sample preparation, consumes reagents and tissue, and may introduce deformation, local mismatch, or tissue loss. When strictly paired stained and unstained sections are difficult to obtain, unpaired virtual staining provides a practical route for improving the readability of unstained TEM images while retaining correspondence with source-image structural cues. This paper presents WAFMGAN, a frequency-aware CycleGAN framework that combines Wavelet Asymmetric Frequency Mamba (WAFM) blocks

A click-compatible BmTyr platform for biotin-free proximity labeling and subcellular proteome profiling in primary T cells.

Proximity labeling has revolutionized the study of dynamic subcellular proteomes by enabling the capture of transient protein interactions within living cells, yet the application of existing platforms to hard-to-transfect primary cells remains challenging. Here, we leverage a bioorthogonal proximity labeling platform based on the copper-dependent tyrosinase BmTyr to profile subcellular proteomes in primary T cells. This system catalyzes the subcellular incorporation of an alkyne-phenol probe, enabling subsequent click-compatible conjugation to versatile azide-bearing tags for fluorescence ima

Gram-negative-staining Bacillaceae with thick cell wall and monoderm architecture uncover evolutionary diversity and challenge Gram-based classification.

Gram staining has guided microbiology for over a century by coloring cells purple or pink, a read-out thought to distinguish monoderms (single membrane, thick peptidoglycan) from diderms (inner and outer membranes with thin wall). Here we show this rule fails repeatedly across Bacillaceae lineages historically deemed "Gram-positive". By combining light and transmission-electron microscopy, antibiotic-sensitivity assays and comparative genomics across 57 strains, we identify "Gram-negative-staining monoderms" lacking outer membrane yet retaining thick peptidoglycan walls. These bacteria lack li

Detecting Callose Deposition in Soybean Lateral Roots During Fungal Infections.

Callose is a β-1,3-glucan polysaccharide deposited at the plant cell wall interface. It is involved in numerous plant physiological processes and responses to both biotic and abiotic stresses. Callose deposition under biotic stress conditions is typically associated with pattern-triggered immunity, which is activated upon recognition of pathogen-associated molecular patterns or damage-associated molecular patterns at the cell wall interface. These depositions reinforce compromised and damaged cell walls caused by pathogen invasion. The standard method for visualizing callose deposition in vari
