Role of cytochrome c as a stimulator of alpha-synuclein aggregation in Lewy body disease.

alpha-Synuclein is a major component of aggregates forming amyloid-like fibrils in diseases with Lewy bodies and other neurodegenerative disorders, yet the mechanism by which alpha-synuclein is intracellularly aggregated during neurodegeneration is poorly understood. Recent studies suggest that oxidative stress reactions might contribute to abnormal aggregation of this molecule. In this context, the main objective of the present study was to determine the potential role of the heme protein cytochrome c in alpha-synuclein aggregation. When recombinant alpha-synuclein was coincubated with cytoch

Dopamine and Rotenone Modulate α-Synuclein Phase Separation and Liquid to Solid Transition.

Liquid-liquid phase separation (LLPS) of α-Synuclein (α-Syn) is recognized as an early biophysical event driving pathological aggregation in Parkinson's disease (PD). Although 10%-15% of PD cases are due to familial mutations, the remaining cases are sporadic, often linked to various factors, like pesticides and metals. For example, rotenone and dopamine are known to be involved in PD pathology and are suggested to cause changes in α-Syn protein homeostasis, although the mechanism by which they influence α-Syn and cellular toxicity is largely unknown. In this work, we demonstrate that both dop

The NLRP3-α-Synuclein Circuit: A Core Driver and Therapeutic Target in Parkinson's Disease.

The neuroinflammatory mechanisms underlying Parkinson's disease (PD) have evolved from a view of localized central events to a systemic network perspective involving "brain-periphery" interactions. The NLRP3 inflammasome, as a core component of the innate immune system, plays a pivotal role within this network. This review aims to systematically elucidate NLRP3's multifaceted roles in PD: First, it delves into the "self-amplifying" vicious cycle between α-synuclein pathology and NLRP3 activation, supports the hypothesis that this cycle may serve as a core engine. Second, it transcends central

α-Synuclein Forms Distinct Micelle-Like Assemblies at Low Ionic Strengths.

Intrinsically disordered proteins (IDPs) often associate into biomolecular condensates. While micron-sized condensates are observed above a well-defined saturation concentration, the formation of nanoscale clusters also occurs at subsaturated concentrations. The diversity and physical nature of such nanoscale clusters remain underexplored with respect to the well-established condensate state. Here we show that the Parkinson's disease-associated protein α-synuclein forms highly monodisperse assemblies at low ionic strength, distinct from the larger polydisperse assemblies previously observed at

Clinical and biomarker predictors of α-synuclein seeding in cognitive impairment and motor parkinsonism.

Dopamine transporter (DAT) imaging and α-synuclein seed amplification assay (SAA) are biomarkers for Lewy body disease (LBD). We aimed to identify clinical and biomarker factors associated with SAA positivity in patients with cognitive impairment and frequent motor parkinsonism. Dual-phase DAT positron emission tomography (PET) scans, cerebrospinal fluid (CSF) phosphorylated-tau/β-amyloid ratio (PTAR) measurement, and SAA were performed in 108 patients, including 99 with motor parkinsonism. Diagnoses, which were not mutually exclusive, included normal pressure hydrocephalus (NPH, N = 60), Alzh

Human microglia clear intraneuronal α-synuclein aggregates by GPNMB-mediated trogocytosis.

Microglia are the primary immune cells of the brain, but their role in Parkinson's disease is not fully understood. Chronic microglial activation is toxic to neurons, but in the early stages of pathology, microglia also exert beneficial functions. We used induced pluripotent stem cell (iPSC)-derived coculture models to investigate how human microglia respond to α-synuclein aggregates that form de novo inside human dopaminergic neurons with α-synuclein gene triplication or are triggered by fibrils. We found that microglia cleared Ser129-phosphorylated α-synuclein aggregates through a contact-de

Multiomic Analysis Reveals Molecular Pathways Associated with Intestinal Aggregation of α-Synuclein.

Aggregates of the protein α-synuclein may initially form in the gut before propagating to the brain in Parkinson's disease (PD). Indeed, our prior work supports that enteroendocrine cells, specialized intestinal epithelial cells, could play a key role in the development of this disease. Enteroendocrine cells natively express α-synuclein and form synapses with enteric neurons as well as the vagus nerve. Severing the vagus nerve reduces the load of α-synuclein aggregates in the brain, suggesting that this nerve is a conduit for gut-to-brain spread. Enteroendocrine cells line the gut lumen; as su

Cholesterol differentially regulates α-synuclein binding across membrane packing regimes.

α-Synuclein (αSyn) is an intrinsically disordered protein that preferentially binds anionic membranes with lipid packing defects. Cholesterol is an abundant membrane component that regulates packing and organization within membranes, yet its effect on αSyn binding remains unclear as prior studies report both cholesterol-mediated enhancement and suppression. Here, we investigated whether these conflicting effects reflect differences in the intrinsic packing state of the phospholipid bilayer. Using a quantitative fluorescence microscopy-based binding assay, we measured αSyn binding preferences a

α-synuclein monoclonal antibodies in Parkinson's disease: A failed promise or unmet potential?

BACKGROUND: Monoclonal antibodies (mAbs) have shown disappointing results in targeting α-synuclein (α-Syn) aggregates in PD, leading us to question whether this therapeutic approach is fundamentally flawed or unrealized potential awaiting better application. This review aims to assess the current evidence, identify key challenges hindering this therapeutic approach, and highlight priorities for future research. METHODS: A narrative review of the literature was conducted focusing on available evidence with respect to safety, efficacy, pharmacokinetics, CNS exposure, peripheral and central targ

Membrane interfacial potential governs surface condensation and fibrillation of α-Synuclein in neurons.

Biomolecular condensates formed via liquid-liquid phase separation (LLPS) are essential for cellular organization. α-Synuclein, an amyloidogenic protein linked to Parkinson's Disease (PD), undergoes phase separation at high concentrations, but the influence of lipid membranes on this process remains unclear. Here, combining in vitro reconstitution, cell biology, and simulations, we show that membranous interfaces promote α-Synuclein condensation at physiologically relevant sub-critical concentrations ( ~ 10 nM) without crowding agents. Notably, condensation occurs only on membranes with a spec

The Influence of Gut Microbiome on Alpha-Synuclein Aggregation: Implications for Parkinson's Disease Pathogenesis.

Parkinson's disease (PD) is a progressive neurodegenerative disorder traditionally characterized by dopaminergic neuronal loss in the substantia nigra and the accumulation of misfolded α-synuclein (α-syn) aggregates. While genetic susceptibility and environmental exposures are well-recognized contributors to PD, growing evidence indicates that disease initiation and progression may also involve peripheral mechanisms originating in the gastrointestinal (GI) tract. Early non-motor symptoms such as constipation, along with the presence of α-syn pathology in the enteric nervous system, have led to

Quantifying the Acute Toxicity of Alpha-Synuclein Membrane-Accumulation in a Cell-Based Assay.

Parkinson's disease (PD) is a progressive neurodegenerative disorder that affects over 12 million people worldwide. A central pathological feature is the accumulation of aggregated alpha-synuclein (αS) in Lewy bodies and Lewy neurites. Engineered αS variants such as 3K (E35K+E46K+E61K) and KLK (KTKEGV→KLKEGV in 6 repeats) have been shown to enhance membrane binding and aggregation propensity, contributing to cellular toxicity. To further investigate the impact of 3K and KLK on αS biology, we developed a sensitive assay in a human neuroblastoma model to assess expression levels and cytotoxicity

α-Synuclein in Lewy Body Diseases: Progress, Remaining Challenges and Future Perspectives.

Lewy bodies (LBs) are the main pathological feature of the neurodegenerative diseases Parkinson's disease and Dementia with LBs. Since their discovery over 100 years ago, it is only in the last three decades, that, a wealth of genetic, pathological and pre-clinical evidence puts the spotlight on accumulated α-Synuclein (α-Syn) as the main component of LBs and implicated as a driver of these diseases. This has catapulted clinical trials for these diseases focussing on strategies to remove, reduce, disaggregate and prevent propagation of α-Syn. Advances in technical approaches have started to bu

PPM1H loss alters intrasynaptic alpha-synuclein localization in a LRRK2-dependent manner.

Mutations in the LRRK2 gene are a major genetic cause of Parkinson's disease (PD) and induce hyperphosphorylation of RAB GTPases through increased LRRK2 kinase activity. The phosphatase PPM1H counteracts this activity by selectively dephosphorylating LRRK2-phosphorylated RAB proteins. Here, we used primary PPM1H knockout neurons to investigate how loss of PPM1H and the resulting hyperphosphorylation of RAB proteins affect the distribution of alpha-synuclein (aSyn), a key protein in PD pathology, at presynaptic terminals. We found that PPM1H deficiency significantly increased the distance betwe

A Structure-Activity Relationship Study of Alpha Synuclein PET Radiotracer M503-1619.

A previous study identified [11C]M503-1619 as a lead compound for positron emission tomography (PET) radiotracer development. The goal of the current study was to conduct a structure-activity relationship (SAR) study on M503-1619 to improve its in vitro binding affinity for alpha synuclein (aSyn), as well as to identify potential radiotracers that could be labeled with fluorine-18. The results of the SAR study identified strict SARs regarding the introduction of a fluorine into the N-aryl piperazine and benzamide moieties. The most promising compound was the corresponding 2-fluoroethyl analog

Engineered neuronal exosomes mediate α-synuclein clearance to ameliorate Parkinson's disease.

Parkinson's disease (PD) is the second most common neurodegenerative disorder after Alzheimer's disease. A hallmark pathological feature of PD is the abnormal aggregation of α-synuclein (αSyn) into insoluble Lewy bodies. Consequently, developing strategies to inhibit αSyn aggregation in the brain has been a major research focus for PD treatment. This study developed a therapeutic approach using engineered neuronal exosomes. These exosomes were modified to extend their blood circulation half-life to 3.8 h and enhance targeting, with a 2.15 ± 0.09% brain signal proportion (vs. 0.78 ± 0.07% for f

microRNA-7-5p and α-Synuclein SAA Predict Parkinson's Disease Phenoconversion.

OBJECTIVE: Corroborate blood neuron-derived extracellular vesicle (NDEV) alpha-synuclein (αSyn), the CSF αSyn seed amplification assay (αSyn-SAA), and blood microRNA-7-5p (miR-7-5p) as markers for Parkinson's disease (PD) phenoconversion and determine if combining these markers would help select subjects who would be more likely to phenoconvert. METHODS: Using Parkinson Progression Marker Initiative data, time to PD phenoconversion was determined by Hoehn and Yahr scores. Log-rank analysis was used to find cutpoints. Cox regression analysis was used with log[miR-7-5p], log[NDEV αSyn], and the

Tyr39 Phosphorylation of α-Synuclein Accelerates Heterotypic Aggregation and Drives Toxic Amplification.

c-Abl-mediated Tyr39 phosphorylation of α-synuclein correlates strongly with Parkinson's disease (PD) progression, yet the mechanistic basis of Tyr39-phosphorylated α-synuclein (pY39-α-syn) in synucleinopathies remains elusive. Here, we show that a minor fraction of pY39-α-syn markedly accelerates the aggregation of wild-type α-synuclein (WT-α-syn), overriding the inhibitory effect of Hsc70. Kinetics, continuous-wave electron paramagnetic resonance (EPR), cryo-transmission electron microscopy (cryo-TEM), and [15N,1H]-NMR spectral analyses reveal that pY39-α-syn participates in primary nucleati

A Biological Framework for Parkinson's Disease: Advances in α-Synuclein-Centered Biomarkers and Staging.

Parkinson's disease (PD) is a neurodegenerative disorder marked by substantial heterogeneity in pathological distribution, disease trajectories, and coexisting neuropathologies. Despite this biological complexity, current diagnosis and staging of PD remain largely anchored in clinical manifestations. Neuronal aggregation of misfolded α-synuclein and degeneration of the nigrostriatal dopaminergic system precede the onset of motor symptoms; this phase is termed the prodromal stage of PD. Therefore, clinically based frameworks inherently limit early detection and stratification. In contrast, Alzh
