Acute anosmia in the mouse: behavioral discrimination among the four basic taste substances.

The importance of taste and smell in discrimination of tastes was examined in normal and anosmic mice. We studied the influence of olfaction on taste sensation using behavioral and electrophysiological methods in both normal animals and animals made anosmic mice by destroying their olfactory receptor cells with zinc sulfate (ZnSO(4)) solution. Electrophysiological responses from chorda tympani nerves showed that peripheral taste receptor cells transmitted taste signals normally to the central nervous system, even when the olfactory senses were abnormal. Behavioral observations showed that mice…

Antimicrobial drug suspensions: a blind comparison of taste of fourteen common pediatric drugs.

Children of preschool age most often receive medications in liquid form, and smell and taste are major determinants in achieving compliance. We compared smell, taste and other characteristics of 14 commonly prescribed antimicrobial suspensions in a blind test in 30 adult volunteers to determine whether there was a difference in their acceptability. A significant difference was observed with cephalosporins ranking highest and penicillins ranking lowest. Our findings support anecdotal observations and claims often made by parents that cephalosporin antimicrobial suspensions taste good and are re…

Evidence that taste aversion learning induced by l-5-hydroxytryptophan is mediated peripherally.

Rats learned to avoid a saccharin solution if their initial consumption of it was followed by intraperitoneal (IP) administration of 25 mg/kg l-5-hydroxytryptophan (l-5-HTP); this taste aversion learning did not occur in rats pretreated with 50 mg/kg (IP) of the aromatic l-amino acid decarboxylase inhibitor RO 4-4602 (benserazide). RO 4-4602 antagonized the l-5-HTP-induced elevation of 5-hydroxytryptamine (5-HT) in the mesentery but significantly increased the l-5-HTP-induced elevation of 5-HT in the brain. These results indicate that l-5-HTP-induced taste aversion is correlated with periphera…

TasTY: Development and evaluation of a taste test for quantifying taste sensitivity among preschool children-test-retest reliability, application, and comparison between children with and without prior SARS-CoV-2 infection.

UNLABELLED: Taste dysfunction gained attention during the coronavirus disease 2019 (COVID-19) pandemic; however, validated objective tests for preschool children remain limited. We aimed to develop a feasible, reliable test quantifying taste sensitivity among children aged 3-6 years and to compare performance between children with and without prior SARS-CoV-2 infection. We included 68 symptom-free children (median age 5.1 years; interquartile range (IQR) 3.9-5.9): 36 without known SARS-CoV-2 infection (control group, CG) and 32 with confirmed prior infection (SARS-CoV-2 infection group, C19-G)…

Preparation of orally disintegrating tablets with taste-masking function: masking effect in granules prepared with correctives using the dry granulation method and evaluation of tablets prepared using the taste-masked granules.

We investigated several methods of taste masking in the preparation of orally disintegrating tablets (ODTs), using furosemide (FU) as a model drug. Four types of FU preparations were prepared: granules with maltitol (MA), granules with yogurt powder (YO), a physical mixture of FU and MA, and a physical mixture of FU and YO. All taste-masking granules were prepared using the dry granulation method. The taste of each type of preparation was evaluated. All four preparations markedly improved the taste of the FU tablets, but the mixing ratios of the correctives did not affect the masking effect. N…

From Waste to Taste: Dynamic Interaction of Grape Stems with Wine Off-Odors.

Within the circular economy framework, grape stems, a major winemaking by-product, are increasingly recognized for their potential to modulate wine composition despite some criticalities. This study aimed to investigate fresh and withered stems as both sources of compounds and adsorbents of off-odors. Corvina and Cabernet Sauvignon stems were tested under three conditions: fresh, and 20% and 40% weight loss. Over 14 days of maceration in red wine, the release kinetics of key enological parameters, including pH, ethanol, total phenolics, methoxypyrazines, and C6 alcohols, were investigated. Con

Zinc deficiency enhances salt preference through altered peripheral and central taste processing mechanisms.

OBJECTIVES: Zinc deficiency alters salt preference, however, the underlying mechanisms remain unclear. We hypothesized that altered salt preference, during zinc deficiency, is associated with brainstem and hypothalamic nuclei modified activity which is involved in the gustatory processing and fluid balance regulation. METHODS: This study elucidated the abnormal intake of high-concentrated sodium chloride solution caused by zinc deficiency in taste reception, taste nerve, and brain. Initially, we performed taste behavioral tests including the chorda tympani (CT) nerve transection (CTx) and the…

Bitter and Sweet Diets Alter Taste Response and Alcohol Consumption Behavior in Mice.

BACKGROUND/OBJECTIVES: Taste guides the consumption of food and alcohol for both humans and rodents. Given that chronic dietary exposure to bitter and sweet foods are purported to alter the perception of bitter and sweet tastes respectively, we hypothesized that dietary habits may shape how the taste properties of ethanol are perceived and thus how it is consumed. METHODS: Using C57BL/6 mice as a model, we contrasted taste behavior, morphology, and expression after a 4-week diet featuring consistent bitter, sweet, or neutral (water) stimuli. RESULTS: Our results demonstrated that a 4-week bi…

Functional bitter taste signaling and stimulus intensity influence diet-induced salivary protein upregulation.

Exposure to tannic acid- and quinine-containing diets upregulates subsets of salivary proteins (SPs), which in turn, results in increased acceptance of the "bitter" diet. However, it is unclear what aspects of the diet consumption influence SP upregulation. It has been thought that taste receptor activation is necessary for upregulation of SPs since sensory stimulation precedes changes in SPs, but no studies have confirmed the contribution of taste to SP upregulation. To test this hypothesis, we used TRPM5 KO and TRPM4/5 DKO mice that have impaired bitter taste signaling. These mice are able t…

A Novel Food-Derived Particle Enhances Sweet and Salty Taste Responses in Mice.

BACKGROUND/OBJECTIVES: Reducing the dietary intake of sugar and salt is considered a key strategy for preventing the onset and progression of lifestyle-related diseases. However, these dietary interventions often compromise the taste of foods, which can reduce patient satisfaction. To address this challenge, we focused on novel food-derived particles (NFPs; patent number P7383867) consisting of lipid, α-cyclodextrin, and xanthan gum formulated as an emulsion with excellent retention and diffusion properties. METHODS: Here, we investigated the effects of NFPs on the taste responses of mice. R…
