A phase II study of docetaxel and carboplatin with concurrent radiation therapy for locally advanced head and neck cancer.

OBJECTIVE: In this study we evaluate the clinical response and safety profile of a regimen of docetaxel+carboplatin concurrent with radiotherapy (RT) in locally advanced squamous cell carcinoma of head and neck (HNSCC). METHODS: Between January 2006 and December 2008, we enrolled 38 patients (stage IVA: 29 patients; stage III: 9 patients). Fourteen had oral cavity cancer (tongue 10, buccal mucosa 2, alveolar ridge 1, floor of mouth 1), 10 had oropharyngeal cancer (base of tongue 5, tonsil 5), 13 had laryngeal cancer, and 1 had maxillary sinus cancer. Patients received concurrent docetaxel 15

Is risk of central nervous system (CNS) relapse related to adjuvant taxane treatment in node-positive breast cancer? Results of the CNS substudy in the intergroup Phase III BIG 02-98 Trial.

BACKGROUND: Breast cancer central nervous system (CNS) metastases are an increasingly important problem because of high CNS relapse rates in patients treated with trastuzumab and/or taxanes. PATIENTS AND METHODS: We evaluated data from 2887 node-positive breast cancer patients randomised in the BIG 02-98 trial comparing anthracycline-based adjuvant chemotherapy (control arms) to anthracycline-docetaxel-based sequential or concurrent chemotherapy (experimental arms). After a median follow-up of 5 years, 403 patients had died and detailed information on CNS relapse was collected for these patie

[A case of advanced gastric cancer with pulmonary carcinomatous lymphangitis responding remarkably to combination chemotherapy of docetaxel, CDDP and S-1].

A 31-year-old woman suffering from stomach pain was admitted to our hospital, and diagnosed with unresectable advanced gastric cancer. She was initially treated with combination therapy of S-1 and CDDP, and a partial response was achieved. After two courses of the chemotherapy, however, she complained of dyspnea, and pulmonary carcinomatous lymphangitis was confirmed by computed tomography. As second-line chemo-therapy, we attempted combination therapy with docetaxel, CDDP and S-1(DCS). After one course of the combination therapy, a remarkable response in the pulmonary carcinomatous lymphangit

Phase II trial of docetaxel and carboplatin in recurrent platinum-sensitive ovarian, peritoneal and tubal cancer.

OBJECTIVE: Docetaxel and carboplatin are active in relapsed ovarian, peritoneal and tubal cancer. Recently, two prospective-randomized trials showed an advantage of carboplatin combination regimen with paclitaxel or gemcitabine over carboplatinum alone in platinum-sensitive cases. The question on the most effective combination with the best tolerable side effects still needs to be answered. METHODS: Eligible patients had recurrent ovarian, peritoneal or tubal cancer (platinum-free interval >6 months), performance status 0-2 and normal bone marrow, renal and hepatic function. 25 patients (age

Mediastinal follicular dendritic cell sarcoma involving bone marrow: a case report and review of the literature.

We report a rare case of mediastinal follicular dendritic cell (FDC) sarcoma involving the bone marrow. The patient, a 46-year-old woman, had a clinically aggressive tumor in the anterior mediastinum that was initially diagnosed as a diffuse B-cell lymphoma. She received chemotherapy but showed no significant improvement. One year later, the patient presented at our institution with pelvic bone metastases. Biopsy specimens of the sacrum lesion and bone marrow were obtained. The diagnosis of FDC sarcoma was made based on histological examination and immunohistochemical findings, including stron

Thymic carcinoma successfully resected with superior vena cava after chemoradiotherapy.

A 57-year-old woman hospitalized for thymic cancer invading the superior vena cava and left brachiocephalic vein evidenced both pleural and pericardial effusion. After chemotherapy with cisplatin and docetaxel and concurrent radiotherapy, the entire tumor was successfully resected along with the pericardium, superior vena cava, and left brachiocephalic vein, followed by vascular reconstruction. Pathologically, viable tumor cells were identified only in the center of the tumor as anaplastic cell carcinoma. Induction chemoradiotherapy thus appears useful in enabling complete resection of advance

[Discovery and development of novel anticancer drug capecitabine].

Capecitabine (N4-pentyloxycarbonyl-5'-deoxy-5-fluorocytidine) is a novel oral fluoropyrimidine carbamate, which was designed to be sequentially converted to 5-fluorouracil (5-FU) by three enzymes located in the liver and in tumors. N4-alkoxycarbonyl-5'-deoxy-5-fluorocytidine derivatives including capecitabine pass intact through the intestinal tract and are sequentially converted to 5-FU by a cascade of the three enzymes. The first step is the conversion to 5'-deoxy-5-fluorocytidine (5'-DFCR) by carboxylesterase located in the liver, then to 5'-deoxy-5-fluorouridine (5'-DFUR) by cytidine deami

Hyaluronic Acid-Modified Nanoparticles Loaded with Cabazitaxel: A Promising Nanomedicine for Malignant Prostate Tumors.

INTRODUCTION: Prostate cancer is the most common cancer among men globally. The firstline drug, cabazitaxel (CTX), has significant side effects such as neutropenia and anemia. To address this, we aimed to develop hyaluronic acid (HA)-modified human serum albumin (HSA)-loaded CTX nanoparticles (HA-CTX NPs) to target prostate tumors with enhanced efficacy and reduced toxicity. METHODS: HA-CTX NPs were synthesized via a self-assembly method and optimized using unidirectional and response surface analyses. The NPs were characterized by particle size, zeta potential, and morphology. In vitro exper

Real-world clinical outcomes and economic burden of discontinuation of taxane therapy among patients with metastatic castration-resistant prostate cancer.

BACKGROUND: Metastatic castration-resistant prostate cancer (mCRPC) treatment commonly includes first-line (1L) androgen receptor pathway inhibitors (ARPIs) followed by second-line (2L) taxane-based chemotherapy. Adverse events (AEs) may lead to early taxane discontinuation, which can negatively affect outcomes and costs. This retrospective observational study assessed real-world outcomes, AEs, and costs of 2L taxane therapy by number of cycles among patients with mCRPC previously treated with a 1L ARPI; outcomes were compared with patients receiving 2L ARPI. METHODS: Overall survival (OS; ov

Cabazitaxel and camptothecin co-loaded in pH-sensitive transferrin-targeted, PEGylated cholesterol-based poly(amidoamine) dendrimers as a synergistic combination for prostate cancer.

The clinical utility of combining cabazitaxel (CBZ) and camptothecin (CPT) for advanced prostate cancer is hindered by emerging drug resistance and the absence of optimized nanocarriers for co-delivery. We investigated the potential of novel Tf-targeted nanocarriers to encapsulate CBZ and CPT, aiming to selectively augment their synergistic anti-cancer activity within in vitro prostate cancer models. Our findings demonstrate a robust synergistic interaction between CBZ and CPT (with a growth inhibition percentage of 87.42% and a combination index of 0.12) in PC3-Luc cells at CBZ:CPT molar rati

Impact of prior nivolumab on treatment outcomes of taxane plus ramucirumab in advanced gastric cancer.

BACKGROUND: Several studies have indicated that exposure to immune checkpoint inhibitors (ICIs) can enhance the efficacy of subsequent chemotherapy. Although combination chemotherapy with ICI has become a standard first-line treatment for human epidermal growth factor receptor 2 (HER2)-negative advanced gastric cancer (AGC), its impact on the efficacy of taxane plus ramucirumab (RAM) remains uncertain. METHODS: We retrospectively analyzed data from consecutive patients with HER2-negative AGC who had previously received oxaliplatin-based chemotherapy, with or without nivolumab, followed by tax

Risk Factors and Clinical Significance of Grade ≥3 Neutropenia During the First Cycle of Cabazitaxel Therapy With Primary Pegfilgrastim Prophylaxis in Metastatic Castration-resistant Prostate Cancer.

BACKGROUND/AIM: Cabazitaxel is an established treatment for metastatic castration-resistant prostate cancer (mCRPC) previously treated with a docetaxel-containing regimen; however, it is frequently associated with severe neutropenia. Although primary prophylaxis with pegfilgrastim is widely used, severe neutropenia still occurs. The present study aimed to identify risk factors for grade ≥3 neutropenia during the first cabazitaxel cycle (Cycle 1) under universal pegfilgrastim prophylaxis and to evaluate its clinical significance. PATIENTS AND METHODS: This retrospective study analyzed 40 patie

Comparing niraparib versus platinum-taxane doublet chemotherapy as neoadjuvant treatment in patients with newly diagnosed homologous recombination-deficient stage III/IV ovarian cancer: Findings from cohort C of the OPAL phase 2 trial.

BACKGROUND: Despite therapeutic improvements in advanced ovarian cancer (aOC), prognosis remains poor for patients with residual disease after cytoreductive surgery. Reported here are results from cohort C of the phase 1b/2 OPAL study of a neoadjuvant poly(ADP-ribose) polymerase (PARP) inhibitor for aOC. METHODS: Eligible adults with newly diagnosed stage III/IV homologous recombination-deficient (HRd) aOC and measurable disease per Response Evaluation Criteria in Solid Tumors (RECIST) v1.1 were randomized 1:1 to receive neoadjuvant platinum-taxane chemotherapy (platinum-taxane) or niraparib.

Clinical Trial Protocol for LuCAB: A Phase I-II Trial Evaluating Cabazitaxel in Combination with [177Lu]Lu-PSMA-617 in Patients with Metastatic Castration-Resistant Prostate Cancer.

[177Lu]Lu-prostate-specific membrane antigen (PSMA)-617 is a standard treatment for patients with metastatic castration-resistant prostate cancer (mCRPC) previously treated with docetaxel and an androgen receptor pathway inhibitor. However, for many, responses are short and progression is inevitable. Contributing factors to treatment resistance include molecular heterogeneity with variable PSMA expression, micrometastases that may not absorb sufficient radiation from 177Lu to result in cell death, and inherent or acquired radioresistance because of genomic alterations or the tumor microenviron

Docetaxel-induced liver and kidney toxicity in rats can be alleviated by suppressing oxidative stress, endoplasmic reticulum stress, inflammation, apoptosis and autophagy signaling pathways after Silymarin treatment.

Approximately 20 million new cancer cases have occurred worldwide, and dose limitation occurs because of the liver and kidney toxicity of chemotherapeutic agents. Inflammation/apoptosis/ROS pathways appear to be activated in the liver and kidney toxicity of chemotherapeutic agents. This study was conducted to investigate the potential effects of silymarin (SLY) use against docetaxel (DTX)-induced liver and kidney damage in rats. For this purpose, 30 mg/kg DTX was administered intraperitoneally to Sprague Dawley rats on the first day of the study, followed by SLY (25 or 50 mg/kg/day) orally for
