Neuroanatomical profiling of the rainbow trout brain parenchyma and meninges reveals specialized immune niches and region-specific hubs for bacterial immune surveillance.

Several studies have described immune responses in the teleost brain and meninges during infection, however, fundamental studies that systematically dissect how different regions of the brain maintain immune homeostasis in teleosts are missing. Here we present an in-depth investigation of the immune status of the brain parenchyma and meninges of juvenile rainbow trout (Oncorhynchus mykiss) at the steady state. We dissected four parenchymal brain regions including olfactory bulbs (OB), telencephalon (Tel), optic tectum (OT) and cerebellum (Cer) and its corresponding dorsal meninges. Gene expres…

Enhanced toxicity of bisphenol F compared to its predecessor bisphenol A in rainbow trout (Oncorhynchus mykiss) during a six-week feeding trial.

This study aimed to assess the toxicity of bisphenol A (BPA) and bisphenol F (BPF) in rainbow trout following six weeks of dietary exposure. Fish were exposed to BPA, BPF, or their combination at nominal concentrations of 10 and 1000 µg/kg (i.e., BPAlow, BPAhigh, BPFlow, BPFhigh, BPA+BPFlow). Haematological analysis revealed a significant reduction (p < 0.05) in leukocytes and lymphocytes, but only in the BPFhigh group. Plasma biochemical markers showed significant increases (p < 0.05) in creatinine in BPAlow and elevated amylase activity in both BPF groups. Surprisingly, markers of endocrine …

Effects of Chronic Stress from High Stocking Density in Mariculture: Evaluations of Growth Performance and Lipid Metabolism of Rainbow Trout (Oncorhychus mykiss).

(1) Background: In aquaculture, chronic stress due to high stocking density impairs animals' welfare and results in declined fishery production with low protein quality. However, most previous studies evaluated the effects of high stocking density on trout in freshwater rather than seawater. (2) Methods: Juvenile trout were reared for 84 days in circular tanks under three stocking densities, including low density ("LD", 9.15 kg/m3), moderate density ("MD", 13.65 kg/m3), and high density ("HD", 27.31 kg/m3) in seawater. The final densities of LD, MD, and HD were 22.00, 32.05 and 52.24 kg/m3, re…

Short communication: Tissue-specific transcript expression of P-glycoprotein isoforms abcb1a and abcb1b in rainbow trout (Oncorhynchus mykiss) following induction with clotrimazole.

P-glycoprotein (P-gp) plays a pivotal role in cellular defense, aimed at reducing xenobiotic accumulation. As a member of the ABC family of proteins, expression of this protein confers the multixenobiotic resistant (MXR) phenotype in aquatic organisms, including fish. To identify tissues protected by or contributing to the elimination of xenobiotics via P-gp, tissue-specific P-gp isoforms abcb1a and abcb1b transcript expression were measured in rainbow trout (Oncorhynchus mykiss). Tissues investigated included the proximal and distal intestines, liver, head kidney, gills, gonads, and 5 regions…

Proliferative Kidney Disease and Proliferative Darkening Syndrome are Linked with Brown Trout (Salmo trutta fario) Mortalities in the Pre-Alpine Isar River.

For many years, brown trout (Salmo trutta fario) mortalities within the pre-alpine Isar River in Germany were reported by the Bavarian Fisheries Association (Landesfischereiverband Bayern e.V.) and local recreational anglers during August and September. Moribund fish seemed to be affected by proliferative darkening syndrome (PDS). In addition, proliferative kidney disease (PKD) caused by Tetracapsuloides bryosalmonae was discussed. To investigate this phenomenon, the present field study monitored brown trout mortalities by daily river inspection in 2017 and 2018. Moribund brown trout (n = 31) …

A chimeric recombinant infectious hematopoietic necrosis virus induces protective immune responses against infectious hematopoietic necrosis and infectious pancreatic necrosis in rainbow trout.

Infectious pancreatic necrosis virus (IPNV) and infectious hematopoietic necrosis virus (IHNV) are two common viral pathogens that cause severe economic losses in all salmonid species in culture, but especially in rainbow trout. Although vaccines against both diseases have been commercialized in some countries, no such vaccines are available for them in China. In this study, a recombinant virus was constructed using the IHNV U genogroup Blk94 virus as a backbone vector to express the antigenic gene, VP2, from IPNV via the reverse genetics system. The resulting recombinant virus (rBlk94-VP2) sh…

Plerocercoids of the cestode Diphyllobothrium ditremum in brown trout Salmo trutta: substantial increase in infection after establishment of European minnow Phoxinus phoxinus.

This study focuses on plerocercoids of the cestode Diphyllobothrium ditremum in brown trout Salmo trutta from the subalpine lake Øvre Heimdalsvatn in south-central Norway. Salmo trutta was the only fish species in this lake until European minnow Phoxinus phoxinus was registered in 1969. The P. phoxinus population increased substantially in the following years. In contrast with the 1969-1972 period, when plerocercoids of D. ditremum were practically absent in S. trutta, there was a high prevalence and intensity of infection in the 2013 S. trutta samples. Because the life cycle of D. ditremum in…

Sex reversal of brook trout (Salvelinus fontinalis) by 17α-methyltestosterone exposure: A serial experimental approach to determine optimal timing and delivery regimes.

Commercial culture of Brook trout (Salvelinus fontinalis) in Tasmania was partly abandoned due to sexual maturation of male fish early on during the estuarine rearing phase. Maturation adversely affects body mass, flesh quality and immunocompetency effectively. Sex reversal techniques such as the in-feed addition of a synthetic androgen have proven difficult to adapt in brook trout. An appropriate timing, duration and delivery vehicle for administration of 17α-methyltestosterone (MT) to produce phenotypic males (neomales) from genotypically female brook trout required further investigation. In…

[Detection of viral hemorrhagic septicemia virus (VHSV) in rainbow trout (Oncorhynchus mykiss) by reverse transcription followed by polymerase chain reaction. Diagnostic validation].

Viral haemorrhagic septicaemia virus (VHSV) is a fish pathogen that attacks rainbow trout (Oncorhynchus mykiss) farming. The only diagnostic method recognised by the European community for VHS is based on the detection of the viral agent in cell culture followed by an immunological identification of the pathogen. Reverse transcription followed by a double amplification of the polymerase chain reaction (RT-PCR) for the gene encoding the viral glycoprotein G is proposed here as an alternative method to the virus assay in cell culture. The RT-PCR was found to have three advantages over the viral …

Does gill boundary layer carbonic anhydrase contribute to carbon dioxide excretion: a comparison between dogfish (Squalus acanthias) and rainbow trout (Oncorhynchus mykiss).

In vivo experiments were conducted on spiny dogfish (Squalus acanthias) and rainbow trout (Oncorhynchus mykiss) in sea water to determine the potential role of externally oriented or gill boundary layer carbonic anhydrase in carbon dioxide excretion. This was accomplished by assessing pH changes in expired water using a stopped-flow apparatus. In dogfish, expired water was in acid-base disequilibrium as indicated by a pronounced acidification (delta pH=-0.11+/-0.01; N=22; mean +/- s.e.m.) during the period of stopped flow; inspired water, however, was in acid-base equilibrium (delta pH=-0.002+…

Characterization of a novel cDNA obtained through differential-display PCR of phorbol ester-stimulated ovarian tissue from the brook trout (Salvelinus fontinalis).

A cDNA (DRC1, differentially regulated clone 1) was obtained from differential-display polymerase chain reaction (PCR) of brook trout ovarian tissue stimulated with phorbol-12-myristate-13-acetate (PMA) and A23187. Using 5' RACE (rapid amplification of cDNA ends), two full-length clones were obtained from DRC1 that were 425 and 660 base pairs long and contained the same open reading frame. On Northern blots, DRC1 hybridized with two ovarian mRNAs of 0.45 and 0.7 kb that were significantly suppressed in the presence of PMA and/or A23187. The mRNAs were not observed in ovaries prior to the resum…

Evaluation of Protective Efficacy of Formalin-Killed and Auxotrophic-Live Vibrio anguillarum Against Homologous and Heterologous Strains in Rainbow Trout (Oncorhynchus mykiss).

Vibrio anguillarum infection poses a significant threat to freshwater and marine aquaculture. Although whole-cell killed and live attenuated vaccines are used to control vibriosis, their comparative efficacy against homologous and heterologous V. anguillarum strains remains limited. Here, we compared the protective efficacy and antibody responses of a formalin-killed V. anguillarum (FKC) and an alanine racemase gene knockout auxotrophic-live V. anguillarum (Δalr1Δalr2 V. anguillarum, LAV), both derived from the V. anguillarum JJ strain, in rainbow trout against homologous (JJ strain) and heter…

Nonlethal detection of Renibacterium salmoninarum in Greenback Cutthroat Trout Oncorhynchus clarkii stomias comparing mucus, blood, and ovarian fluid samples to kidney tissues.

OBJECTIVE: Renibacterium salmoninarum, the causative agent of bacterial kidney disease, poses a major threat to both wild and aquaculture salmonid populations. Traditional detection methods typically involve lethal sampling to collect kidney tissues but are often impractical for species of conservation concern. This study evaluates nonlethal sampling techniques for detecting R. salmoninarum in Greenback Cutthroat Trout Oncorhynchus clarkii stomias by comparing mucus, blood, and ovarian fluid samples to conventional kidney tissue. METHODS: During the 2019 spawning season, we collected samples …

Lake Trout (Salvelinus namaycush) Naturally Infected with Salmovirus salmonidallo3 (SalHV-3; Family Alloherpesviridae) Continue to Harbor the Virus for Nearly a Decade.

Salmovirus salmonidallo3 (SalHV-3) causes Epizootic Epitheliotropic Disease (EED), which has resulted in the death of millions of lake trout (Salvelinus namaycush) over the past 40 years. Although advancements in understanding this virus's pathogenicity and control strategies have been made, the duration and effects of chronic SalHV-3 infections remain unknown. This study focused on lake trout that survived a natural outbreak of EED in 2012 and were maintained under quarantine conditions until 2021. Following exposure to either repeated or intermittent handling stress designed to mimic typical…

Physiological and transcriptomic responses in brown trout, Salmo trutta, to multiple stressors: Pesticide mixtures, elevated water temperature and the proliferative kidney disease.

Contamination with pesticides, rising water temperatures and pathogen pressure represent a multiple stressor scenario relevant to surface water ecosystems globally.
