Antifungal susceptibility of genital yeast isolates, Auckland, 2016-2025.

AIM: Our aim was to record current susceptibility results of genital yeast isolates and to compare the results to the previous report covering the period 2001-2015. METHODS: Genital yeast isolates had their antifungal susceptibility determined by disc diffusion or minimum inhibitory concentration following standard methods. Sequential isolates from the same person had their initial and last susceptibility results compared. RESULTS: Disc testing was performed on 1,307 initial isolates; 17 (1.3%) were from males. The most frequent isolates were Candida albicans (64%), Nakaseomyces glabratus co

Development of human monoclonal antibodies against TARM1 by yeast display.

TARM1, a leukocyte immunoglobulin-like receptor expressed on myeloid cells, functions as a costimulatory receptor promoting proinflammatory cytokine secretion. Recent studies have revealed the involvement of TARM1 in immune-mediated diseases. Despite its biological importance, research tools for elucidating human TARM1 function have been limited. Here, we generated monoclonal antibodies (mAbs) against human TARM1 by a yeast display platform combined with a human single-chain variable fragment (scFv) library. Two scFv clones were isolated and converted into IgG1 antibodies, both of which bound

Stress-induced remodeling of membrane-bound organelles in budding yeast.

Membrane-bound organelles undergo extensive remodeling during environmental stress, yet systematic side-by-side comparisons of organelle responses in budding yeast remain limited. Here, a panel of fluorescent markers was used to examine multiple organelles in Saccharomyces cerevisiae exposed to heat, hydrogen peroxide, acetic acid, or ethanol. Across all conditions, mitochondria consistently shifted from tubular networks to fragmented puncta, representing a common stress response. Quantitative scoring showed that heat stress induced mitochondrial fragmentation in more than 90% of cells within

[Microenvironmental regulation strategies and applications of yeast surface display systems].

Yeast surface display (YSD) is an important technological platform integrating protein engineering, synthetic biology, and industrial biocatalysis. Its catalytic efficiency and stability highly depend on the dynamic synergy of the three-tier microenvironments: the cell wall, intracellular environment, and extracellular compartments. This review systematically elaborates on the core components of the YSD microenvironment, including the cell wall, anchor proteins, intracellular secretion pathways, and extracellular physicochemical properties, and clarifies their critical roles in the folding, ac

Yeast vaccine production platform for human and animal infectious diseases.

Yeasts have contributed to human and animal health through functional antigen production for vaccine formulations. Some yeast-made vaccines have become a reality for humankind because they have reached commercialization (hepatitis B, HPV, and tick parasitosis). Many other vaccine prototypes are under preclinical and clinical evaluations, hoping for their usage soon. Currently, genomes, genetic modification techniques, and industrial vaccine manufacturing have been successfully developed for Saccharomyces cerevisiae, Komagataella phaffii (formerly Pichia pastoris), and Hansenula polymorpha. Mor…

Kodamaea ohmeri: An emergent yeast from a One Health perspective.

Kodamaea ohmeri is an emerging and opportunistic yeast associated with a high mortality rate in humans. As it is commonly found in the environment, it is possible that environmental conditions and agricultural practices contribute to the adaptation of this yeast and the selection of antifungal resistance. During a multicentric study in Brazil, conducted under a One Health perspective, 14 isolates of K. ohmeri were identified from different sources: three from blood cultures, three from animals (swine and poultry), and eight from animal environments (swine and poultry). Yeasts were isolated usi…

Molecular mechanism of host-yeast interactions and prevention by nanoformulation approaches.

Fungal infections are a major source of morbidity and mortality in people with compromised immune systems, such as those with human immunodeficiency virus, cancer, organ transplant recipients, and patients undergoing chemotherapy in healthcare settings. According to a recent World Health Organization (WHO) fungal priority pathogens list, Cryptococcus spp., Candida spp., Aspergillus spp., and Candida auris cause severe invasive infections in human. These opportunistic pathogens cause a significant number of mycoses, which affect over a billion people annually. Around two million infections can

Long telomere inheritance through budding yeast sexual cycles.

The ends of linear eukaryotic chromosomes are protected from being recognized as DNA double-strand breaks by telomeres, containing repetitive DNA sequences that bind specific proteins. In humans, mutations in telomere regulatory genes lead to short or long telomere syndromes. These syndromes often show genetic anticipation, where the disease has an earlier onset and a more severe manifestation in each new generation. Later generations inherit not only the mutation affecting telomere length, but also abnormal telomere length. Many aspects of telomere length homeostasis are conserved between mam

Bioenergy production from yeast through a thermo-chemical platform.

Alternative fuels are urgently needed to mitigate greenhouse gas emissions. This study was conducted to recover bioenergy from non-edible feedstock, an oleaginous yeast biomass obtained during fed-batch cultivation of Yarrowia lipolytica. Yeast oil (lipids) was extracted from the harvested biomass and readily converted into biodiesel using the non-catalytic transesterification method. The conversion yield of the convertible lipids was 97.4 wt%, even with a high content of unidentified impurities (> 12.7 wt%). To maximize bioenergy production and minimize waste generation, the yeast biomass res

Nutrient factors specifically promoting ethanol fermentation but not growth in yeast.

Yeast ethanol fermentation and growth are affected by environmental factors such as nutrients, pH, and temperature. Ethanol fermentation in yeast is typically accompanied by cell proliferation. Thus, the specific nutrients required for fermentation remain unclear. To determine nutrients required for fermentation, first a semi-synthetic medium with a small inoculum size (OD600 = 2) was used, and CO2 gas emissions were monitored. The addition of sodium aspartate (Asp) and MgSO4 to a medium containing 0.2 % yeast extract and 9.1 % glucose efficiently increased gas emissions. Further addition of K

Response of pulcherrimin-producing Metschnikowia yeast to different iron sources.

Metschnikowia pulcherrima clade yeasts produce pulcherriminic acid, which binds with iron to form pulcherrimin, a red pigment. However, the functions of pulcherriminic acid in yeasts and bacteria that synthesize it remain incompletely understood. To gain deeper understanding of these functions the responses of pulcherriminic acid-secreting Metschnikowia yeast in the presence of various solid iron sources (metallic iron, magnetite, and lepidocrocite) containing iron in different valence states were examined. By applying the classical plate method, the formation of pulcherrimin, was observed bot…

De novo biosynthesis of plant lignans by synthetic yeast consortia.

Reconstructing the biosynthesis of complex natural products such as lignans in yeast is challenging and can result in metabolic promiscuity, affecting the biosynthetic efficiency. Here we divide the lignan biosynthetic pathway across a synthetic yeast consortium with obligated mutualism and use ferulic acid as a metabolic bridge. This cooperative system successfully overcomes the metabolic promiscuity and synthesizes the common precursor, coniferyl alcohol. Furthermore, combined with systematic engineering strategies, we achieve the de novo synthesis of key lignan skeletons, pinoresinol and la

Successful Resolution of Suspected Red Yeast Rice-induced Nephropathy.

Recently, several Japanese patients have developed interstitial nephritis or acute tubular injury/necrosis, both accompanied by Fanconi syndrome, after consuming a specific red yeast rice supplement. In most cases, the renal function did not fully recover even after discontinuation of supplements and glucocorticoid treatment. A 52-year-old woman developed interstitial nephritis and Fanconi syndrome after receiving supplements. A renal biopsy revealed mild lymphocytic infiltration. Her renal function recovered after discontinuing the supplement without glucocorticoids. The supplement was contam

Regulation of misfolded protein aggregation and degradation by SUMOylation in budding yeast.

Protein misfolding is linked to many neurodegenerative disorders, such as Huntington's disease. The increase of glutamine-encoding CAG repeats in the first exon of huntingtin (HTT) causes Huntington's disease. Protein fragments of Htt exon 1 with polyQ expansion (mutant HTT, mHtt) are prone to aggregation, resulting in oligomers, amyloid fibrils, or large inclusion bodies. Previous studies demonstrate mHtt SUMOylation, a process of covalent attachment of small ubiquitin-like modifiers (SUMO) to target proteins. Protein polySUMOylation further triggers its ubiquitination and segregation by the

The Promising Role of Selenium and Yeast in the Fight Against Protein Amyloidosis.

In recent years, increasing attention has been paid to research on diseases related to the deposition of misfolded proteins (amyloids) in various organs. Moreover, modern scientists emphasise the importance of selenium as a bioelement necessary for the proper functioning of living organisms. The inorganic form of selenium-sodium selenite (redox-active)-can prevent the formation of an insoluble polymer in proteins. It is very important to undertake tasks aimed at understanding the mechanisms of action of this element in inhibiting the formation of various types of amyloid. Furthermore, yeast ce

Engineering a human P2X2 receptor with altered ligand selectivity in yeast.

P2X receptors are a family of ligand gated ion channels found in a range of eukaryotic species including humans but are not naturally present in the yeast Saccharomyces cerevisiae. We demonstrate the first recombinant expression and functional gating of the P2X2 receptor in baker's yeast. We leverage the yeast host for facile genetic screens of mutant P2X2 by performing site saturation mutagenesis at residues of interest, including SNPs implicated in deafness and at residues involved in native binding. Deep mutational analysis and rounds of genetic engineering yield mutant P2X2 F303Y A304W, wh
