Lymphatic and portal venous absorption of gastrically administered thiopental and phenobarbital were compared in the dog. Thiopental concentrations in lymph were significantly higher than phenobarbital concentrations for 30 min after drug administration. Disparity between portal plasma and lymph concentrations for thiopental during the first 15 min was about 1/3 as great as the same difference for phenobarbital. We have measured, by an intubation method, gastric evacuation and gastrointestinal absorption of alcohol ingested with a meal in seven healthy nonalcoholic subjects. A homogenized meal containing [14C]PEG and ethanol (1 g/kg body wt) was given intragastrically while saline containing [57CO]vitamin B12 was perfused into the duodenum. The in vitro adsorption of sodium pentobarbital by activated charcoal (USP), Darco G-60 and SuperChar was studied. Various solutions of sodium pentobarbital, ranging in concentration from 2.5 to 10 mg/ml (pH = 9), were prepared in distilled water. Radiolabeled (14C) sodium pentobarbital was added to serve as a concentration marker. Two millimeters of each solution was added to from 5 to 350 mg of each charcoal in a test tube. An in situ experimental model of rectal absorption was studied in rabbits compared with in vivo determination of drug concentration in the plasma of postcaval vein using indomethacin as drug. The effect of sodium salicylate and anaesthesia on the rectal absorption of theophylline was evaluated in rats. Theophylline proved to be slowly but completely absorbed on rectal infusion in conscious rats, compared with intravenous infusion. Pentobarbital anaesthesia did not influence absorption. In contrast to literature data, the results with salicylate showed that rectal absorption of theophylline was not enhanced.

Altered tissue levels of trace metals have been reported in streptozotocin-diabetic (STZ) rats. To determine whether increased hepatic and renal levels of Cu and Zn were associated with enhanced intestinal absorption, trace metal absorption was studied in control (C) and STZ rats using dietary balance and in situ ligated-loop techniques. The effect of picolinic acid and hydroxyquinolines in intestinal zinc absorption of cattle was studied. In contrast to reports of the absorption in rat and in man, picolinic acid did not enhance zinc absorption in cattle. In two calves suffering from Adema disease, the bovine parallel to Acrodermatitis enteropathica, hydroxyquinoline supplementation resulted in an increase in intestinal 65Zn absorption, while picolinic acid had no such effect. The results may indicate a difference in the zinc absorption mechanism between cattle and other species.

Serum concentrations of diazepam and N-desmethyldiazepam were measured in six adult patients following administration of 10 mg diazepam in solution by the rectal, intravenous, and intramuscular routes. Maximum serum concentrations of 121--200 ng/ml were recorded from 10 to 20 min. after the rectal instillation, whereas following intramuscular injection the levels rose slowly and irregularly, reaching a maximum (62--186 ng/ml) in 1 to 24 hours.
