[The therapeutic effect of metadoxine on alcoholic and non-alcoholic steatohepatitis].

The therapeutic effect of metadoxine on alcoholic and non-alcoholic steatohepatitis. Alcohol-induced liver disease is one of the main epidemic problems of nowadays. Non-alcoholic steatohepatitis has been described only for some years, but it also needs much more attention in the future. The etiologic factors of both entities are quite different, but the pathologic changes of the liver are nearly the same, therefore there could be certain drugs, that are equally effective in their therapy. Metadoxine is one of them, mainly because its protective effect against the damage done by free radicals.

Alcohol dependence: a critical look at the effects of alcohol metabolism.

This paper explores the metabolic consequences of alcohol misuse and identifies the pathophysiological reasons why alcohol, no matter what quantity is taken regularly, is not beneficial for the normal functioning of most body systems. Although moderate ethanol consumption may reduce stress and the risk of coronary heart disease, ethanol also exerts a direct toxicological effect because it interferes with hepatic metabolism and immune functions. Liver transplantation may be necessary for end-stage liver disease in alcoholics. A causal effect between alcohol intake and several cancers has been r

A descriptive study of trauma, alcohol, and alcoholism in young adults.

Young adults, 18-20 years of age, admitted to a trauma center via the emergency department, were studied to determine if they had been drinking prior to their injury event. The prevalence of self-reported chronic alcohol problems was examined using the short Michigan Alcohol Screening Test (SMAST). Of the 319 subjects, 131 (41%) tested positive for alcohol, including about one-half of those with intentional injuries and 38% with unintentional injuries. Approximately 22% had blood alcohol concentrations of 100 mg/dL or more, indicating they were legally intoxicated at the time of their injury.

Parental alcoholism: an examination of male and female alcoholics in treatment.

This study investigated the implications of parental alcoholism in 639 male and female alcoholics who were in treatment. In comparison with persons who did not report a parental history of alcoholism (PA-), the 264 offspring of alcoholics (PA+) were characterized by higher levels of alcohol dependency and alcohol-related consequences, increased reports of use of other drugs and higher levels of MMPI-measured psychopathology. Comparisons of PA+ patients with PA- patients with the same age of onset of problematic alcohol use suggested that PA+ persons are characterized more by an early onset of

Prognosis of alcoholic cirrhosis in the presence and absence of alcoholic hepatitis.

Liver biopsy specimens (178 percutaneous and 39 transjugular) were assessed from 217 consecutive patients with alcoholic liver disease, 77 noncirrhotics and 140 cirrhotics, whose cases were followed for 5 yr. Cirrhotic patients were categorized into two groups, with and without "hepatitis" using a criteria to define "hepatitis" that included only degrees of inflammation, necrosis, and Mallory bodies that had a prognostic weight in terms of mortality in 1 yr. This classification resulted in a sharp separation between a group of 42 patients with cirrhosis without "hepatitis" and with low mortali

[Importance of alcoholism markers in the diagnosis of alcoholic heart lesions].

A study was made of opportunities for the use of some signs of alcoholic intoxication in the verification of alcoholic etiology of heart lesion. Clinical signs like hyperemia of the face with telangiectasia, venous plethora of the eyeballs, tremor of the lips, tongue, limbs, Dupuytren's contracture, enlarged liver size combined with a positive macrocytosis test and, to a lesser degree, with a higher activity of gamma-glutamyl transpeptidase, and the detection of fatty hepatosis in liver puncture biopsy were shown to suggest alcoholic intoxication, and excluding other cases of heart lesion they

Alcoholic women in fertile age treated at an alcohol clinic.

The consumption of alcohol by women in Sweden is strongly increasing, especially in younger individuals. Since the rediscovery of the teratological properties of alcohol most of the studies concerning foetal alcohol damage in man have covered female skid row alcoholics. This investigation describes the medical and social characteristics of a group of women (n = 92) receiving inpatient care for alcoholism compared with an age-matched control group, in relation to obstetrical history. The conditions in the control group were in accord with those of the general population. Social problems and deg

Sensitisation to Mallory bodies (alcoholic hyalin) in alcoholic hepatitis.

Using a leucocyte migration inhibition test sensitisation to Mallory bodies (alcoholic hyalin) was found in a statistically significant 41% of 17 patients with alcoholic hepatitis. Patients with alcohol-induced fatty liver and cirrhosis did not demonstrate sensitisation. Mallory bodies are a characteristic feature of alcohol-induced liver damage, and immunological sensitisation to them might lead to liver cell death and cell progression of the hepatitis process.

Alcoholism--the second great imitator. An introduction to the problem of alcoholism.

The problem of alcohol abuse and alcoholism pervades our society. The economic losses run into billions of dollars annually. What is more important is the number of fatalities associated with the problem and the effect of alcohol on decision-making processes within all areas of government and nongovernment. The pathologist must understand the fundamental diagnostic processes, recognize the anatomic and clinical findings, and treat alcoholism as a medical problem.

Increase in the incidence of alcoholic pancreatitis and alcoholic liver disease in Iceland: impact of per capita alcohol consumption.

Objective: To analyze the incidence of acute alcoholic pancreatitis and of severe alcoholic liver disease (ALD) and its association with per capita alcohol consumption with identification of both alcoholic cirrhosis (AC) and severe alcoholic hepatitis (AH), in a population-based setting. Methods: A search was undertaken in diagnoses database for diagnostic codes in order to find patients hospitalized with incident acute alcoholic pancreatitis (AP) and alcoholic liver disease in Iceland in 2001-2015. Diagnoses were verified in all patients who were retrospectively reviewed. Those with ALD had ei

[A new sight on alcohol metabolism and alcoholism--role of high Km alcohol dehydrogenase ADH3 (Class III)].

Alcohol metabolism is known to be mainly carried out by the classic ADH1 (Class I) of the liver. However, another pathway has been also suggested to play important roles in alcohol metabolism especially at high levels of blood ethanol and under chronic drinking. Over the past three decades, vigorous attempts to identify the enzyme responsible for the non-ADH1 pathway have focused on the microsomal oxidizing system (MEOS) and catalase, but have failed to clarify their roles in systemic alcohol metabolism. Recently, we used ADH3-null mutant mice to demonstrate that high Km ADH3 (Class III), a ub

An unfolded protein response-inducible protein (ULI1) mediates aromatic alcohols biosynthesis through reinforcing the Ehrlich pathway in Saccharomyces cerevisiae.

Aromatic alcohols such as 2-phenylethanol (2-PE) and tryptophol (TOL) possess diverse biological functions. Here, an unfolded protein response-inducible gene ULI1 and an aldehyde dehydrogenase gene ALD3 in Saccharomyces cerevisiae were disrupted using the CRISPR-Cas9 system. Fermentation assays revealed that both the ULI1 frameshift (Kmuli1) and knockout (KΔuli1) mutants exhibited significantly reduced 2-PE and TOL yields relative to the wild-type strain KMLY1-6, retaining 46.32 and 64.54% for 2-PE, and 34.61 and 77.73% for TOL, respectively. In contrast, ALD3 knockout moderately elevated 2-PE…
