Identification of potential modulator of Anopheles gambiae odorant binding protein 1 by hierarchical virtual screening and molecular dynamics.

Malaria is a protozoan infection transmitted by the bite of the infected female mosquito belonging to the genus Anopheles spp., which causes more than 445 million annual deaths worldwide. Available drugs have serious adverse effects (e.g. blurred vision, hypotension and headache) and species-dependent efficacy. An alternative to overcome these problems involve the use of molecules with affinity to the Anopheles gambiae mosquito odor receptors, minimizing the reinfection process as well as reducing the problems related to pharmacological therapy. The vector control can interrupt the epidemiolog…

Application of matrix-assisted laser desorption/ionization mass spectrometry to identify species of Neotropical Anopheles vectors of malaria.

BACKGROUND: Malaria control in Panama is problematic due to the high diversity of morphologically similar Anopheles mosquito species, which makes identification of vectors of human Plasmodium challenging. Strategies by Panamanian health authorities to bring malaria under control targeting Anopheles vectors could be ineffective if they tackle a misidentified species. METHODS: A rapid mass spectrometry identification procedure was developed to accurately and timely sort out field-collected Neotropical Anopheles mosquitoes into vector and non-vector species. Matrix-assisted laser desorption/ioni…

Hydrogen cyanide produced by the soil bacterium Chromobacterium sp. Panama contributes to mortality in Anopheles gambiae mosquito larvae.

Mosquito larvae continuously encounter microbes in their aquatic environment, which serve as food and play a critical role in successful development. In previous work, we isolated a Chromobacterium sp. (C.sp_P) with larvicidal activity from the midgut of dengue vector Aedes mosquitoes in Panama. In this study, we found a positive correlation between initial concentrations of C.sp_P and larval mortality rates, and that C.sp_P is more efficient at inducing larval mortality in a high nutrient environment. Multiple Chromobacterium species induce larval mortality with similar efficacy to C.sp_P exc

Bio-efficacy of deltamethrin based durable wall lining against wild populations of Anopheles gambiae s.l. in Northern Tanzania.

BACKGROUND: Indoor residual spraying (IRS) is one of the preferred tools used for control of malaria in many settings in the world. However, this control tool still faces challenges that include lack of long lasting active ingredient, limited number of well-trained personal, and need of repeated treatment which increases operational costs and reduces acceptability by residents. As a result there is need to develop and validate other methods which can complement the existing controls. The current study compared the bio-efficacy of durable wall lining (DL) (treated with deltamethrin 265 mg/m2) a

Some strains of Plasmodium falciparum, a human malaria parasite, evade the complement-like system of Anopheles gambiae mosquitoes.

Plasmodium falciparum lines differ in their ability to infect mosquitoes. The Anopheles gambiae L3-5 refractory (R) line melanizes most Plasmodium species, including the Brazilian P. falciparum 7G8 line, but it is highly susceptible to some African P. falciparum strains such as 3D7, NF54, and GB4. We investigated whether these lines differ in their ability to evade the mosquito immune system. Silencing key components of the mosquito complement-like system [thioester-containing protein 1 (TEP1), leucine-rich repeat protein 1, and Anopheles Plasmodium-responsive leucine-rich repeat protein 1] pr…

Co-infections of Plasmodium knowlesi, P. falciparum, and P. vivax among Humans and Anopheles dirus Mosquitoes, Southern Vietnam.

A single Anopheles dirus mosquito carrying sporozoites of Plasmodium knowlesi, P. falciparum, and P. vivax was recently discovered in Khanh Phu, southern Vietnam. Further sampling of humans and mosquitoes in this area during 2009-2010 showed P. knowlesi infections in 32 (26%) persons with malaria (n = 125) and in 31 (43%) sporozoite-positive An. dirus mosquitoes (n = 73). Co-infections of P. knowlesi and P. vivax were predominant in mosquitoes and humans, while single P. knowlesi infections were found only in mosquitoes. P. knowlesi-co-infected patients were largely asymptomatic and were conce…

Present habitat suitability for Anopheles atroparvus (Diptera, Culicidae) and its coincidence with former malaria areas in mainland Portugal.

Malaria was a major health problem in the first half of the 20th Century in mainland Portugal. Nowadays, although the disease is no longer endemic, there is still the risk of future endemic infections due to the continuous occurrence of imported cases and the possibility of transmission in the country by Anopheles atroparvus Van Thiel, 1927. Since vector abundance constitute one of the foremost factors in malaria transmission, we have created several habitat suitability models to describe this vector species' current distribution. Three different correlative models; namely (i) a multilayer per…

Comparative analysis of BAC and whole genome shotgun sequences from an Anopheles gambiae region related to Plasmodium encapsulation.

The only natural mechanism of malaria transmission in sub-Saharan Africa is the mosquito, generally Anopheles gambiae. Blocking malaria parasite transmission by stopping the development of Plasmodium in the insect vector would provide a useful alternative to the current methods of malaria control. Toward this end, it is important to understand the molecular basis of the malaria parasite refractory phenotype in An. gambiae mosquito strains. We have selected and sequenced six bacterial artificial chromosome (BAC) clones from the Pen-1 region that is the major quantitative trait locus involved in…

Life-table analysis of Anopheles malaria vectors: generational mortality as tool in mosquito vector abundance and control studies.

BACKGROUND & OBJECTIVES: Vector control will for sometime remain a primary weapon in the war against vector borne diseases. Malaria is of paramount importance in this with its associated high morbidity and mortality especially in sub-Saharan Africa. This study on generational mortality associated factors in Anopheles mosquitoes life-table analysis was designed to investigate the fecundity, levels of mortality and mortality associated factors at the aquatic stages of anopheline malaria vectors. METHODS: Mortality associated factors were investigated at the eggs, I and II instar larval, III and…

Efficacy of mosquito nets treated with a pyrethroid-organophosphorous mixture against Kdr- and Kdr+ malaria vectors (Anopheles gambiae).

In order to prevent the resistance of Anopheles gambiae s.l. to pyrethroids from spreading too quickly and to lengthen the effectiveness of insecticide impregnated mosquito nets, it has recently been suggested to use mixtures of insecticides that have different modes of action. This study presents the results obtained with tulle mosquito nets treated with bifenthrin (a pyrethroid) and chlorpyrifos-methyl (an organophosphorous) both separately and in mixture on two strains of An. gambiae, one sensitive to all insecticides, and the other resistant to pyrethroids. The values of KDt50 and KDt95 an…

Immunological localization of Bacillus thuringiensis serovar israelensis toxins in midgut cells of intoxicated Anopheles gambiae larvae (Diptera: Culicidae).

Fourth instar larvae of Anopheles gambiae were intoxicated with doses of purified crystals from Bacillus thuringiensis serovar israelensis (Bti) corresponding to 50-fold the LC50 after 24 h. Midguts were dissected after various contact times, then processed for immuno-light and -electron microscopy. Immunodetection on thin sections was performed using affinity-purified rabbit IgG against Bti crystal CryIVD or CytA polypeptides, in combination with anti-rabbit IgG/peroxidase. Both polypeptides were detected by optical and electron microscopy after 15 min of contact with Bti crystals on the apic…

Site-specific ribosomal DNA insertion elements in Anopheles gambiae and A. arabiensis: nucleotide sequence of gene-element boundaries.

The nucleotide sequence of the junctions between the 28S ribosomal gene and site-specific insertion elements from two sibling mosquito species, Anopheles gambiae and A. arabiensis, is reported. In both species, elements insert at the same point within the 28S gene, but this site is 634 basepairs (bp) 3' of the R1 (Type I) insertion site in Drosophila melanogaster. The two mosquito elements each have poly A tails and a polyadenylation signal, but the extreme 3' and 5' ends show no other similarity to each other or to any other insertion element. In both mosquito species, identical target site d…

Comparative evaluation of synthetic attractants against an important malaria vector, Anopheles stephensi (Diptera: Culicidae) mosquitoes in laboratory conditions.

Mosquito-borne diseases are a major public health concern, and exploiting the mosquito's reliance on host odors is an effective way to prevent human exposure. We evaluated the attraction of non-blood-fed female Anopheles stephensi Liston, 1901 mosquitoes (Bandar Abbas strain, laboratory population) to four chemical components found in vertebrate host odors (at 5 dilutions) and their blends using an olfactometer in laboratory conditions. When each compound was combined with CO2, three of the four tested compounds showed significant attractancy (P < 0.05) at their optimal dilutions. Hexanoic aci…

Bacterial and Fungal Microbiota in Anopheles darlingi Exhibit Differences in Diversity Across Three Main Colombian Malaria-endemic Regions.

Anopheles darlingi is the main malaria vector in the Neotropics. Despite their importance, little is known about the microbiota composition in areas where these vectors are found. Since the microbiota may influence host biology and vector competence, it is essential to understand these microbial communities in endemic regions. This study explores the bacterial and fungal communities in An. darlingi from three malaria-endemic areas in Colombia and examined how geography influences microbial composition. Mosquitoes were collected from the Bajo Cauca, Pacific and Amazonas regions, and their micro…

Metarhizium pingshaense infection reverses insecticide resistance in Anopheles gambiae sensu lato by altering energy reserves and gene expression.

BACKGROUND: Entomopathogenic fungi like Metarhizium are emerging as effective biopesticides against malaria vectors. They reduce mosquito survival, fecundity, and flight ability, and reverse insecticide susceptibility in resistant Anopheles gambiae sensu lato strains. To elucidate the unclear underlying mechanisms, this study investigates the effects of fungal infections and insecticide exposure on the mosquito's energy reserves and the expression of key metabolic and immune genes. METHODS: Three mosquito types: (i) pyrethroid-resistant An. gambiae sensu lato and two laboratory colonies: (ii)…

Standing genetic variation underlies divergence of nicotinic acetylcholine receptor subunits among cryptic species of the Anopheles gambiae complex.

Arthropod species differ in insecticide susceptibility, yet how pre-existing polymorphism at target sites shapes variable responses within and among populations remains poorly understood. Recently diverged taxa provide ideal systems to test how target-site divergence modulates species sensitivity. Using whole-genome sequencing data from 573 mosquitoes representing six cryptic species of the Anopheles gambiae complex, we analyzed standing genetic variation across all 11 nicotinic acetylcholine receptor (nAChR) subunit genes to establish a baseline for natural diversity before the large-scale de…
