Gyrodactylosis, caused by Gyrodactylus spp., poses a significant challenge to sustainable aquaculture, highlighting the need for effective and safe anthelmintics. In this study, atovaquone, a specific inhibitor of the mitochondrial cytochrome bc1 complex, was repurposed as a therapeutic agent against Gyrodactylus kobayashii. In vivo bath treatments demonstrated high potency, with a 48-h EC50 of 0.005 mg/L, which is lower than that reported for most existing natural and synthetic compounds. Notably, atovaquone exhibited greater efficacy in vivo than in vitro, likely due to its lipophilic accumu…

Cases of infection with the canine hookworm Ancylostoma caninum that are resistant to all anthelmintic classes registered in the United States of America (USA) for control of this parasite in dogs were first reported in 2019. Termed Multiple Anthelmintic Drug Resistance (MADR), cases have subsequently been reported in pet dogs across the USA. Recent studies using molecular tests that detect mutations known to cause resistance to benzimidazole anthelmintics have revealed an alarming frequency of this trait in pet dogs throughout the USA, with some reports also from Canada. Since the initial rep…

Strongyloidosis is a parasitic disease caused by Strongyloides stercoralis, a nematode with a complex life cycle that facilitates long-term persistence within the host. The infection affects millions of people in tropical and subtropical regions and poses a particular challenge in immunocompromised individuals. Although conventional treatments, such as ivermectin and albendazole, are generally effective, emerging concerns regarding drug resistance and adverse effects have prompted the search for alternative therapeutic options. In this context, natural products-including plant extracts, bioact

Gastrointestinal nematode (GIN) infections are a major concern for the ruminant industry worldwide and result in significant production losses. Naturally occurring polyparasitism and increasing drug resistance that potentiate disease outcomes are observed among the most prevalent GINs of veterinary importance. Within the five major taxonomic clades, clade Va represents a group of GINs that predominantly affect the abomasum or small intestine of ruminants. However, the development of effective broad-spectrum anthelmintics against ruminant clade Va GINs has been challenged by a lack of comprehen…

ETHNOPHARMACOLOGICAL RELEVANCE: Infections caused by parasitic worms or helminth continue to pose a great burden on human and animal health, particularly in underdeveloped tropical and subtropical countries where they are endemic. Current anthelmintic drugs present serious limitations and the emergence of drug resistance has made it increasingly challenging to combat such infections (helminthiases). In Bangladesh, medicinal plants are often used by indigenous communities for the treatment of helminthiases. Knowledge on such plants along with screening for their anthelmintic activity has the po

This study focuses on developing novel antimicrobials to combat drug-resistant pathogens, addressing compounds failing clinical trials due to inadequate physicochemical properties. Sixteen imidazolidine-4-one derivatives were synthesized by extensive evaluation using molecular docking, absorption, distribution, metabolism, excretion (ADME) predictions, and antimicrobial testing. Molecular docking studies conducted with Schrödinger's Glide revealed that compounds S4 and G8 exhibited superior docking scores of -7.839 and -7.776, respectively. The G series outperformed the S series in scores. ADM

In the face of increasing drug resistance, the development of new anthelmintics is critical for controlling nematodes that parasitise livestock. Although hymenopteran venom toxins have attracted attention for applications in agriculture and medicine, few studies have explored their potential as anthelmintics. Here we assessed hymenopteran venoms as a possible source of new anthelmintic compounds by screening a panel of ten hymenopteran venoms against Haemonchus contortus, a major pathogenic nematode of ruminants. Using bioassay-guided fractionation coupled with liquid chromatography-tandem mas…

Ascarid parasites infect a variety of hosts and regular anthelmintic treatment is recommended for all species. Parascaris spp. is the only ascarid species with widespread anthelmintic resistance, which allows for the study of resistance mechanisms. The purpose of this study was to establish an in vitro drug exposure protocol for adult anthelmintic-naïve Parascaris spp. and report a preliminary transcriptomic analysis in response to drug exposure. Live worms were harvested from foal necropsies and maintained in RPMI-1640 at 37 °C. Serial dilutions of oxibendazole (OBZ) and ivermectin (IVM) were
