Effect of chlorpheniramine on muricide induced by thiamine deficiency: pharmacokinetic and behavioral studies.

Chlorpheniramine (5-10 mg/kg) showed a dose-dependent suppressive effect on muricide induced by thiamine deficiency at 1 hr after oral administration. The ED50 value for muricidal suppression was about 7.1 mg/kg (95% confidence limits, 5.4-9.3 mg/kg). The maximum effect of muricidal suppression lasted for 1-2 hr after oral dosage (10 mg/kg). Chlorpheniramine was detectable at 10 min in the blood and brain of thiamine deficient killer-rats by using a high-performance liquid chromatographic method. Maximum brain and blood levels of chlorpheniramine appeared at 45 and 60 min, respectively. In ter

Potential role of glucagon like peptide 1 in taste receptors.

The perception of taste is a complex physiological process that extends far beyond the simple detection of flavor molecules, serving as a critical interface between nutrient sensing, metabolic regulation, and feeding behavior. Emerging evidence reveals that this process is profoundly modulated by endocrine and neuromodulatory systems, creating a sophisticated gut-brain-taste axis that integrates peripheral gustatory signals with central homeostatic and hedonic mechanisms. Hormones such as glucagon-like peptide-1, leptin, ghrelin, and CCK not only regulate appetite and energy balance but also d

FGF21-inducing rare sugars for addressing obesity and alcoholism.

Obesity and alcoholism are major global health burdens driven in part by maladaptive ingestive behaviors. Accordingly, functional foods that curb overeating and excessive alcohol consumption may help prevent these disorders. Ingestion of simple sugars or alcohol increases plasma fibroblast growth factor 21 (FGF21), which activates oxytocin (OXT) neurons in the paraventricular nucleus of the hypothalamus (PVH) and suppresses the specific appetite for sugar and alcohol. D-Allulose and D-tagatose are generally recognized as safe rare sugars and serve as potent inducers of FGF21. Oral administrati

Feeding decision-making by a single neuron via disparate neurotransmitters.

Animals use gustatory information to decide whether to ingest nutritious substances or avoid toxic ones. Although certain neurons in the gustatory circuits respond to both aversive and appetitive signals, how these neurons resolve inputs with opposing valences is unknown. Here, we examine how the Drosophila melanogaster neuropeptide leucokinin (LK) affects gustatory information processing to elicit the appropriate feeding behaviors. We identify the subesophageal LK neurons (SELKs) as downstream synaptic partners of gustatory receptor neurons and show that these two groups are functionally conn

A trichobezoar in a child with undiagnosed celiac disease: a case report.

Celiac disease is a chronic, immune-mediated enteropathy caused by a permanent sensitivity to ingested gluten cereals that develops in genetically susceptible individuals. The classic presentation of celiac disease includes symptoms of malabsorption but has long been associated with cognitive, emotional, and behavioral disorders. We describe an 8-year-old patient with non-scarring alopecia and diagnosed with trichotillomania. Furthermore, she presented with a 3-year history of poor appetite and two or three annual episodes of mushy, fatty stools. Laboratory investigations showed a normal hemog

[Judicious use of anorectics].

Physicians treating adipose patients often hesitate to include drugs. Considering new developments in obesity research and drug-therapy, this attitude should be discussed. Anorectics of the amphetamine-group and analogues (derivates) react with the noradrenergic system and induce appetite-suppression. Fenfluramine in the d-1 configuration and isomeric dextro-rotation influence release of serotonin, reduce intake of food and increase sensation of saturation. In addition, various eating behavior disorders like nibbling and addiction to sweets are discussed. Use of Dextro-fenfluramine for these s

Anatomical evidence links the stomach to the central amygdala, a region responsive to local GLP-1R agonist induced feeding and nausea-like behaviors in male mice.

BACKGROUND: The glucagon-like peptide-1 receptor (GLP-1R) agonist liraglutide is an effective therapeutic agent for obesity, primarily through its ability to suppress appetite and delay gastric emptying. However, the central neural substrates mediating its effects on food intake remain incompletely defined. METHODS: Male mice received subcutaneous liraglutide injections in the cervical region to evalutates its effects on feeding behavior and body weight regulation. Retrograde transsynaptic tracing using pseudorabies virus (PRV) was employed to identify central amygdala (CeA) involvement in ga

Tumor-derived cytokine enhances bitter sensing through remote control of bitter taste neurons via the Upd3/Spz5/Toll-6 axis in Drosophila.

The sense of taste is essential as it governs appetite and the feeding process. However, it is largely unknown whether and how tumors in a host communicate with peripheral taste sensing. Using the well-established ykiS168A tumor models in Drosophila melanogaster, we found that flies carrying brain or gut tumors exhibit enhanced avoidance of bitter compounds such as caffeine but showed no avoidance of sucrose, and the degree of the avoidance was correlated with severity of the tumor phenotype. Through RNAi screening of upregulated cytokines secreted by malignant tumors, we identified Upd3 as a

Postoral receptors and alimentary taste: implications for energy intake and appetite.

Eating behavior is shaped by genetic, psychological, and physiological factors, with nutrient sensing playing a central role in modulating intake. The tongue, as the primary gustatory organ, initiates this process by influencing hedonic preferences, food choices, and feeding behavior. Recent sensory research has highlighted the potential role of an emerging class of taste modalities known as alimentary tastes. This concept refers to the gustatory detection of compounds that produce weak or subtle taste perceptions but elicit strong postoral effects. While most studies have focused on umami and

Neuroinflammation disorders exacerbated by environmental stressors.

Neuroinflammation is a condition characterized by the elaboration of proinflammatory mediators within the central nervous system. Neuroinflammation has emerged as a dominant theme in contemporary neuroscience due to its association with neurodegenerative disease states such as Alzheimer's disease, Parkinson's disease and Huntington's disease. While neuroinflammation often is associated with damage to the CNS, it also can occur in the absence of neurodegeneration, e.g., in association with systemic infection. The "acute phase" inflammatory response to tissue injury or infections instigates neur

DNA methylation-modifiers reduced food intake in juvenile chickens (Gallus gallus) and Japanese quail (Coturnix japonica).

S-Adenosylmethionine (SAM) is the major endogenous methyl donor for methyltransferase reactions, while 5-Azacytidine (AZA) is a synthetic drug inhibiting DNA methyltransferase activity. Both molecules can thus influence DNA methylation patterns in an organism and thereby affect gene expression and ultimately behavior in the long-term. Whether or not effects on behavior are exerted on a shorter time scale is unclear. The goal of this study was to explore the direct effects of SAM and AZA on appetite regulation, using broiler chicken and Japanese quail as the animal models. Fed or 180 min-fasted

Involvement of mesolimbic structures in short-term sodium depletion: in situ hybridization and ligand-binding analyses.

Acute treatment with the diuretic furosemide (Lasix) produces a reduction in plasma Na(+) and volume as well as increased thirst and salt appetite. The resulting hypovolemia stimulates the well-known counter-regulatory physiological response from the renin-angiotensin-aldosterone system. However, the neurochemical players underpinning the behavioral responses of thirst and salt appetite are less clear. Previously, we have reported that salt-replete deoxycorticosterone (DOCA) treatment activates mesolimbic structures associated with reward and goal-seeking behavior. The present study was design

Dopamine D2 receptors in the bed nucleus of the stria terminalis modulate alcohol-related behaviors.

Dysregulation of the dopamine (DA) system is a hallmark of substance abuse disorders, including alcohol use disorder (AUD). Of the DA receptor subtypes, the DA D2 receptors (D2Rs) play a key role in the reinforcing effects of alcohol. D2Rs are expressed in numerous brain regions associated with the regulation of appetitive behaviors. One such region is the bed nucleus of the stria terminalis (BNST), which has been linked to the development and maintenance of AUD. Recently, we identified alcohol withdrawal-related neuroadaptations in the periaqueductal gray/dorsal raphe to BNST DA circuit in ma…

Behavioral Symptomatology in the Premenstruum.

OBJECTIVE: Sleep and eating behaviors are disturbed during the premenstrual phase of the menstrual cycle in a significant number of reproductive-age women. Despite their impact on the development and control of chronic health conditions, these behaviors are poorly understood. In the present study, we sought to identify affective and psychological factors which associate with premenstrual changes in sleeping and eating behaviors and assess how they impact functionality. METHODS: Fifty-seven women provided daily ratings of premenstrual symptomatology and functionality across two-three menstrual

Association between MC4R rs17782313 polymorphism and overeating behaviors.

BACKGROUND/OBJECTIVES: Melanocortins have a crucial role in appetite and weight regulation. Although the melanocortin 4 receptor (MC4R) gene has been repeatedly linked to obesity and antipsychotic-induced weight gain, the mechanism behind how it leads to this effect in still undetermined. The goal of this study was to conduct an in-depth and sophisticated analysis of MC4R polymorphisms, body mass index (BMI), eating behavior and depressed mood. SUBJECTS/METHODS: We genotyped 328 individuals of European ancestry on the following MC4R markers based on the relevant literature on obesity and anti

Clinical Management of Obesity in Women: Addressing a Lifecycle of Risk.

The World Health Organization estimates that nearly 2 billion people worldwide are overweight, 600 million of whom are obese. The increasing prevalence of this condition in women is of particular concern given its impact on reproductive health and mortality. Burgeoning data implicating maternal obesity in fetal programming and the metabolic health of future generations further suggest that obesity in women is one of the most pressing public health concerns of the twenty-first century. However, health care professionals are infrequently engaged in obesity management. This article provides a con
