Agelas Wasting Syndrome Alters Prokaryotic Symbiont Communities of the Caribbean Brown Tube Sponge, Agelas tubulata.

The brown tube sponge Agelas tubulata (cf. Agelas conifera) is an abundant and long-lived sponge on Caribbean reefs. Recently, a disease-like condition, Agelas wasting syndrome (AWS), was described from A. tubulata in the Florida Keys, where prevalence of the syndrome increased from 7 to 35% of the sponge population between 2010 and 2015. In this study, we characterized the prokaryotic symbiont community of A. tubulata for the first time from individuals collected within the same monitoring plots where AWS was described. We also sampled tissue from A. tubulata exhibiting symptoms of AWS to det

Ferroplasma acidarmanus RPA2 facilitates efficient unwinding of forked DNA substrates by monomers of FacXPD helicase.

The strand-separation activity that is important for many cellular DNA processing machineries is provided by DNA helicases. In order to understand the physiological properties of a helicase acting in the context of its macromolecular machinery, it is imperative to identify the proteins that interact with the enzyme and to analyze how these proteins affect its helicase activities. The archaeal Rad3 helicase XPD (xeroderma pigmentosum group D protein) from Ferroplasma acidarmanus (FacXPD) is a superfamily II 5'-->3' DNA helicase. Similar to its mammalian homolog working as an integral part of th…

Microbial community dynamics and first quantification of the toxin domoic acid in a eutrophic bay in the United Arab Emirates.

The marine planktonic community (eukaryotic, bacterial and archaeal) has been little investigated in the Persian/Arabian Gulf (PAG), despite its crucial role in sustaining biogeochemical cycles and the ecological health of this subtropical ecosystem, which during summer is the hottest marine body of water on Earth. This study explored the temporal changes in the planktonic community in a densely populated, semi-enclosed bay in the United Arab Emirates (UAE), in which seasonal succession of persistent phytoplankton blooms resulted in beach closures. We surveyed the microbial community by analyz…

Ingredients for microbial life preserved in 3.5 billion-year-old fluid inclusions.

It is widely hypothesised that primeval life utilised small organic molecules as sources of carbon and energy. However, the presence of such … lled microbial communities associated with the barite deposits. Our findings demonstrate that early Archaean hydrothermal fluids contained essential primordial ingredients that provided fertile substrates for earliest life on our planet.

Molecular characterization of a dechlorinating community resulting from in situ biostimulation in a trichloroethene-contaminated deep, fractured basalt aquifer and comparison to a derivative laboratory culture.

Sodium lactate additions to a trichloroethene (TCE) residual source area in deep, fractured basalt at a U.S. Department of Energy site have resulted in the enrichment of the indigenous microbial community, the complete dechlorination of nearly all aqueous-phase TCE to ethene, and the continued depletion of the residual source since 1999. The bacterial and archaeal consortia in groundwater obtained from the residual source were assessed by using PCR-amplified 16S rRNA genes. A clone library of bacterial amplicons was predominated by those from members of the class Clostridia (57 of 93 clones), …

A hidden archaeal lineage with an ultra-reduced genome retaining only its replicative core.

Host-associated prokaryotes can undergo extreme genome reduction, but the most reduced genomes belong to mutualistic endosymbionts whose hosts compensate for lost functions, eroding the symbiont's identity as an autonomous cell. Whether comparable reduction can occur while preserving the informational autonomy of a cell remains unclear. Here, we report the discovery of Candidatus Sukunaarchaeum mirabile, an archaeon with a genome of only 238 kbp (less than half the size of the smallest known archaeal genome) obtained by single-cell genomics from a dinoflagellate cell. Phylogenetic analyses rev…

Thermal stratification-mediated DOM transformation shapes the vertical coupling-to-decoupling of carbon and nitrogen in an oxic alpine reservoir.

Owing to depth and distinctive geography, alpine reservoirs develop persistent seasonal thermal stratification that reshapes habitats and sh … relieve heterotrophic carbon limitation and accelerate mineralization, supporting ammonia-oxidizing archaea (AOA) nitrification. Incomplete denitrification potential and oxygen suppression of nosZ favored N2O accumulation via nitrification by-products, reflecting labile-carbon-associated C-N coupling. Structural and biochemical insights into the thermostable esterase Ta0887 from Thermoplasma acidophilum.

Microbial esterases are versatile and stable enzymes with a wide range of biotechnological applications. However, few esterases have been characterized from archaea, an important source of extremophilic enzymes. In this study, we report the biochemical characterization and crystal structure of Ta0887, a novel esterase from the thermoacidophilic archaeon Thermoplasma acidophilum. The protein was successfully cloned, expressed, and purified in Escherichia coli. Light scattering assays revealed that Ta0887 is a monomer in solution. Activity assays using p-nitrophenyl (p-NP) esters confirmed its e…

Developing a robust biomethanation process by improving oxygen tolerance of Methanothermobacter wolfeii strain BSEL.

Application of biomethanation for conversion of CO2 and H2 to renewable natural gas (RNG) is constrained by the strong O2 sensitivity of methanogenic archaea. In biogas facilities, O2 enters through leaks, sampling ports, and intentional micro-aeration for hydrogen sulfide control, creating intermittent oxidative conditions that can destabilize a downstream biomethanation process for biogas upgrading to RNG. An O2-tolerant biocatalyst reduces reliance on strict anaerobic techniques for handling the biocatalyst as well as purification of the gases used for operating biomethanation units. For re…

Genotypic Variation in Rice Root Exudates Controls Biological Nitrification Inhibition Efficiency in the Rhizosphere.

Nitrification in agricultural soils drives significant nitrogen fertiliser losses and nitrous oxide (N2O) emissions, particularly in intensi … tively targeted nitrifiers with little effect on the broader microbial community. Ammonia-oxidising archaea (AOA), the dominant nitrifiers in this acidic flooded soil, were more inhibited than ammonia-oxidising bacteria (AOB) in soil and in exudate-amended culture assays. The Gut-Lung Microbiome Crosstalk and Pulmonary Disease.

Both the gut and the lungs possess a microbiome, a community of commensal bacteria, archaea, fungi, and viruses that perform important housekeeping functions in those organs. The colonic microbiome primarily ferments indigestible dietary fibers into essential short-chain fatty acids, synthesizes essential vitamins, regulates the mucosal immune system, and forms a protective barrier against pathogenic colonization. The lung microbiome maintains respiratory health primarily by regulating mucosal immunity, providing a physical barrier against invading pathogens, and producing beneficial metabolit…

Introduction.

Microorganisms colonize nearly all anatomical sites of the human body, with the oral cavity hosting one of the most diverse, accessible, and densely populated microbial ecosystems. The oral microbiome comprises a complex consortium of bacteria, fungi, viruses, archaea, and protozoa that inhabit distinct ecological niches. Each niche provides unique physicochemical conditions that shape microbial composition, structure, and function. In addition to oral and dental sites, oral biofilms frequently develop on dental materials, appliances, and prostheses, where surface characteristics such as rough…

Tumor Topography Remodels TME Signalling via Compartment-Specific Host-Microbiome-Tumor Crosstalk in Hepatoblastoma.

Background & Aims: Signaling within the tumor microenvironment (TME) emerges from dynamic crosstalk among cancer cells, host tissues, and the microbiome. Orthotopic and ectopic xenograft models are widely used in preclinical liver cancer research, yet it remains unclear to what extent tumor location shapes systemic host responses and gut-liver communication. We therefore investigated how anatomical context influences host transcriptomes and spatially resolved intestinal microbial ecosystems in hepatoblastoma (HB). Methods: We integrated bacterial/archaeal ecology with host and xenograft transc…
