[Identification of residual antibiotics in the tissues of slaughter animals using electrophoresis bioautography].

An electrophoretic identification chart of the antibiotics penicillin, streptomycin, neomycin, erythromycin, tylosin and tetracyclines was made. Minimum inhibiting concentrations of the above antibiotics were determined for the bacterial tester-strains Bacillus subtilis BGA, Micrococcus luteus ATCC 9341 and Bacillus stearothermophillus v. calidolactis C 953. The obtained results were applied to identify residues of antibacterial substances in the tissues of slaughter animals, milk and other samples, as a follow-up of detection microbiological methods of plate agar diffusion.

Suppressive Effects of Volatile Compounds from Bacillus spp. on Magnaporthe oryzae Triticum (MoT) Pathotype, Causal Agent of Wheat Blast.

The Magnaporthe oryzae Triticum (MoT) pathotype is the causal agent of wheat blast, which has caused significant economic losses and threatens wheat production in South America, Asia, and Africa. Three bacterial strains from rice and wheat seeds (B. subtilis BTS-3, B. velezensis BTS-4, and B. velezensis BTLK6A) were used to explore the antifungal effects of volatile organic compounds (VOCs) of Bacillus spp. as a potential biocontrol mechanism against MoT. All bacterial treatments significantly inhibited both the mycelial growth and sporulation of MoT in vitro. We found that this inhibition was

Effects of levels of purity of spores of two Bacillus species on their resistance, germination, and core CaDPA and water content.

UNLABELLED: Previous work (V. K. Juneja, M. Osoria, E. G. Altuntas, N. K. Taneja, et al., Food Res Int 177:113904, 2024, https://doi.org/10.1016/j.foodres.2023.113904) showed that less well-purified spores of Bacillus cereus and Bacillus subtilis were more wet heat resistant than more well-purified spores. Extending the previous work, the current work used (i) B. subtilis spores identical to the one used previously but lacking a plasmid providing kanamycin resistance and (ii) spores of a different B. cereus strain, and with spores made on both plates and in liquid. There were larger difference…

Conservation and Specificity in Bacillus Biofilm Dynamics: On Structure and Function of B. cereus Camelysins.

The B. cereus family comprises members highly pathogenic for mammals or insects, with B. anthracis and B. thuringiensis respectively as notable examples. The biofilm operon of these bacteria encodes two TasA-like proteins, the 60% identical Camelysins CalY1 and CalY2. In this study, we observed that at neutral pH CalY2 alone polymerizes readily into filaments, whereas CalY1 forms a polydispersed mixture of oligomers without filament formation. However, at basic or acidic pH CalY1 also modestly polymerizes. CalY2 polymerization into filaments involves ß-sheet remodeling via donor strand complem

Semi-Artificial Photosynthesis Enables Light-Driven Biomanufacturing in Non-Photosynthetic Bacteria.

Semiconductor biohybrid systems integrate microbial biosynthesis with semiconductor light harvesting to sustainably power energy-intensive chemical production. Although such systems have been widely explored in photosynthetic bacteria, extending them to non-photosynthetic biomanufacturing chassis such as Bacillus subtilis (B. subtilis) remains substantially more challenging, owing to peptidoglycan-induced nanoparticle aggregation and mass-transfer limitations, cytotoxicity, and inefficient interfacial electron transfer from loose adsorption between inorganic semiconductors and cells. Here, we …

Antitermination of transcription of catabolic operons.

Antitermination of transcription mediated by proteins interacting with mRNA sequences is described for nine operons/regulons. Eight of the systems are catabolic, while the ninth, the Klebsiella pneumoniae nas regulon, is involved in the assimilation of nitrate and nitrite. Six of the catabolic operons/regulons are found in Bacillus subtilis, one is found in Escherichia coli, and one in Pseudomonas aeruginosa. The antitermination system of five of the operons/regulons (E. coli blg, and sacPA, sacB, bgl, and lic from B. subtilis) are assigned to the bgl-sac family on the basis of extensive simil…

¹H, ¹³C, and ¹⁵N resonance assignments and secondary structure prediction of the full-length transition state regulator AbrB from Bacillus anthracis.

The AbrB protein is a transcription factor that regulates the expression of numerous essential genes during the cells transition phase state. AbrB from Bacillus anthracis is, nototriously, the principal protein responsible for anthrax toxin gene expression and is highly homologous to the much-studied AbrB protein from Bacillus subtilis having 85% sequence identity and the ability to regulate the same target promoters. Here we report backbone and sidechain resonance assignments and secondary structure prediction for the full-length AbrB protein from B. anthracis.

Genomic determinants of sporulation in Bacilli and Clostridia: towards the minimal set of sporulation-specific genes.

Three classes of low-G+C Gram-positive bacteria (Firmicutes), Bacilli, Clostridia and Negativicutes, include numerous members that are capable of producing heat-resistant endospores. Spore-forming firmicutes include many environmentally important organisms, such as insect pathogens and cellulose-degrading industrial strains, as well as human pathogens responsible for such diseases as anthrax, botulism, gas gangrene and tetanus. In the best-studied model organism Bacillus subtilis, sporulation involves over 500 genes, many of which are conserved among other bacilli and clostridia. This work aim…

Investigating the Modes of Action of the Antimicrobial Chalcones BC1 and T9A.

Xanthomonas citri subsp. citri (X. citri) is an important phytopathogen and causes Asiatic Citrus Canker (ACC). To control ACC, copper sprays are commonly used. As copper is an environmentally damaging heavy metal, new antimicrobials are needed to combat citrus canker. Here, we explored the antimicrobial activity of chalcones, specifically the methoxychalcone BC1 and the hydroxychalcone T9A, against X. citri and the model organism Bacillus subtilis. BC1 and T9A prevented growth of X. citri and B. subtilis in concentrations varying from 20 µg/mL to 40 µg/mL. BC1 and T9A decreased incorporation …

The Listeria monocytogenes Fur-regulated virulence protein FrvA is an Fe(II) efflux P1B4 -type ATPase.

Listeria monocytogenes FrvA (Lmo0641) is critical for virulence in the mouse model and is an ortholog of the Bacillus subtilis Fur- and PerR-regulated Fe(II) efflux P1B4 -type ATPase PfeT. Previously, FrvA was suggested to protect against heme toxicity. Here, we demonstrate that an frvA mutant is sensitive to iron intoxication, but not to other metals. Expression of frvA is induced by high iron and this induction requires Fur. FrvA functions in vitro as a divalent cation specific ATPase most strongly activated by ferrous iron. When expressed in B. subtilis, FrvA increases resistance to iron bo…

The Conserved Spore Coat Protein SpoVM Is Largely Dispensable in Clostridium difficile Spore Formation.

The spore-forming bacterial pathogen Clostridium difficile is a leading cause of health care-associated infections in the United States. In order for this obligate anaerobe to transmit infection, it must form metabolically dormant spores prior to exiting the host. A key step during this process is the assembly of a protective, multilayered proteinaceous coat around the spore. Coat assembly depends on coat morphogenetic proteins recruiting distinct subsets of coat proteins to the developing spore. While 10 coat morphogenetic proteins have been identified in Bacillus subtilis, only two of these …

Bacillus spores in the mail: "ironing" out the anthrax problem.

INTRODUCTION: Following a terrorist release of Bacillus anthracis spores through the U.S. Postal Service, it was suggested that decontamination of spores in mail envelopes could be accomplished at home by utilizing the steam heat from a household iron. This study investigated the use of a standard household iron, in a dry heat mode, for sterilizing mail envelopes laced with bacterial spores. METHODS: The Bacillus subtilis var. niger spore, a more heat resistant spore, was used as a surrogate for B. Anthracis spores. Standard mail envelopes containing 2.1 x 10(6) spores were sealed and subject…
