DNA Origami-Based Multivalent Nanobody Display Platform for Potent Neutralization of Botulinum Neurotoxin Type A.

Botulinum neurotoxins (BoNTs) are the most potent biological toxins discovered to date. Rapid capture and clearance of BoNTs from the bloodstream is a critical strategy for poisoning treatment. Nanobodies, with their high affinity, represent promising molecules for neutralizing BoNTs. Due to its small molecular weight, it has the limitation of a short half-life in the bloodstream. The BoNT/A-neutralizing nanobody, ciA-C2, binds to the receptor-binding domain of BoNT/A, blocking its interaction with the neuronal receptor SV2 and thereby preventing toxin entry into cells to achieve detoxificatio

Preparation of Antibody-Modified mRNA-LNPs via a Combination of Post-Encapsulation and Post-Insertion.

Using ligand-modified lipid nanoparticles (LNPs) to encapsulate messenger RNA (mRNA) has become an important modality for tissue/organ/cell-specific gene delivery. Modifying mRNA-LNPs either with antibodies or their derivatives (Ab-mRNA-LNPs) has emerged as a promising drug delivery system (DDS) due to its efficacy and target specificity. Manufacturing Ab-mRNA-LNPs, however, involves complex and multi-step procedures that include the encapsulation of mRNA followed by the anchoring of antibodies via polyethylene glycol (PEG) linker-conjugated lipids. In this work, we present a seamless strategy

A novel medical trauma plate using bioactive ceramic composites - design and numerical modelling analysis.

In traditional orthopaedics and traumatology, titanium alloy trauma plates (TATPs) are commonly used as internal fixators for fractured bones, typically with screws. However, this technique presents challenges: TATPs often require removal after healing due to non-degradability and potential corrosion, and their stiffness mismatch with bone can cause stress-shielding that may delay healing. To address these issues, a nacre-like biomimetic apatite-wollastonite composite has been introduced to develop a new trauma plate for orthopedic treatments and prosthetics. This bioactive ceramic composite t

Comparison of mechanical properties of third generation silk protein for use in osteosynthesis fixation systems.

BACKGROUND: Osteosynthesis systems are widely used in the bony skeleton, such as in post-oncologic or traumatic reconstruction and orthognathic surgery. Silk protein-based biomaterials are novel alternatives to metal fixation systems, with studies showing degradability, good biocompatibility, nonsensitivity to temperature, and minimized stress-shielding. Mechanical properties of three generations of silk-based hardware were studied and compared to conventional metal and resorbable systems. METHODS: Silk plates and screws were prepared using three techniques: (1) hexafluoro-2-propanol-derived

Soft Depth Neural Probes Enable Chronic Recordings from the Rat Brainstem.

Implanted microelectrodes offer a direct method for electrically interfacing with individual neurons in vivo. A major challenge preventing long-term deployment of such interfaces is the degradation of recording quality over time. This is especially evident when targeting deep brain structures, as surgical access is challenging and invasive, and depth probes are subjected to elevated levels of micro-motion. To address these challenges, miniaturization and biomimetic softness are emerging as effective strategies to improve stability and biointegration of neural implants. Here, we propose a penet

A Triple-Layer Amniotic Membrane Dressing Drives Robust Wound Healing: In-Depth Protein Profiling and In Vivo Validation in Rat and Human Subjects.

In chronic ulcers, early intervention to obtain wound closure is a critical requirement for optimal reparative success. Human dried amniotic membrane (HDAM) has been widely used as a wound covering due to three characteristics: its content of highly enriched extracellular matrix (ECM) and bioactive cytokines, its long shelf life, and its suitable biomechanical properties adequate for handling in the dried state. The ECM composition can be enhanced by the creation of a sandwich composed of three distinct layers of the amniotic membrane without creating any detrimental manufacturing shortcomings

Deciphering Graphene-Homocysteine Interactions via Density Functional Theory: Impact on Platelet Function through an Experimental Approach.

The biomedical usefulness of graphene materials depends on their stability in blood and how they interact with plasma biomolecules. In circulation, they can bind to fibrinogen, platelets, and lipids, key players in thrombosis, potentially influencing coagulation and thrombotic risk. Homocysteine (Hcy), a sulfur-containing amino acid associated with cardiovascular disorders, plays a crucial role in platelet activation and oxidative stress, yet its interaction with graphene derivatives remains poorly understood. Given the experimental complexity of systematically screening graphene derivatives f

Composite Decellularized Corneal Hydrogel for Effective Corneal Injury Repair and Regeneration.

Repairing corneal stromal defects remains a significant clinical challenge. While hydrogels have been explored for corneal injury repair, their structural and compositional dissimilarity to natural corneal tissue has limited their potential as a viable alternative to corneal transplantation. In this study, a decellularized corneal hydrogel system (HCSL) was developed to address these limitations. The HCSL integrates decellularized corneal stromal lenticules, gelatin methacrylate (GelMA), and oxidized dextran (Odex) to create a composite decellularized corneal hydrogel with superior mechanical

Transparent and Antifouling Lubrication Coating for Medical Applications Formed by a Self-Assembled Method.

Lubrication surfaces reduce the risk of cross-contamination and enhance the long-term stability of medical devices, which holds significance in the realm of antifouling medical materials. However, the complexity of constructing micronano structures to immobilize lubricating fluids and the fluorine content typically found in silane coupling agents restrict their widespread adoption. In this study, we prepared a biomimetic lubricating coating (BLC) through the one-step self-assembly of octadecyltrichlorosilane and oil infusion. The BLC exhibits pronounced repellency to liquids of different surfa

Collagen Hybridizing Peptides Promote Collagen Fibril Growth In Vitro.

Recreating the structural and mechanical properties of native tissues in vitro presents significant challenges, particularly in mimicking the dense fibrillar network of extracellular matrixes such as skin and tendons. This study develops a reversible collagen film through cycling collagen self-assembly and disassembly, offering an innovative approach to address these challenges. We first generated an engineered collagen scaffold by applying plastic compression to the collagen hydrogel. The reversibility of the collagen assembly was explored by treating the scaffold with lactic acid, leading to

Improving the Efficiency of Ultrasound and Microbubble Mediated Gene Delivery by Manipulation of Microbubble Lipid Composition.

Ultrasound and microbubble-mediated gene delivery is emerging as a powerful nonviral gene delivery approach due to its ability to target various tissues. Since microbubble cavitation plays a crucial role in gene delivery, factors affecting cavitation, such as microbubble composition, size, ultrasound pressure, frequency, and pulse interval, can directly affect the efficiency of gene delivery. The effect of ultrasound parameters on gene delivery efficiency has been systematically investigated in numerous studies. However, relatively few studies have investigated the influence of different micro

A-BioHPG: A Hydrophilic Biodegradable and Hemocompatible Hyperbranched/Dendritic Polyketal Polyglycerol with Tunable Degradation.

We report the design and synthesis of a biodegradable hyperbranched polyether polyketal polyol, containing acid-labile ketal groups with controlled degradation, high water solubility, excellent hemocompatibility, and cell compatibility. For this, we have developed a new AB2-type acid-labile monomer, 2-((4-(2-(oxiran-2-ylmethoxy) ethoxy)tetrahydro-2H-pyran-4-yl)oxy)ethan-1-ol (OTPE). The OTPE monomer is copolymerized with glycidol through anionic ring-opening multibranching polymerization generating OTPE-incorporated hyperbranched polyglycerol (A-BioHPG) with varying degradable monomer content.

Enhancing the biocompatibility of phakic intraocular lens via selective fibronectin trapping.

Myopia has become a significant public health problem in recent decades, resulting in a profound public health and financial burden. The phakic intraocular lens (PIOL) utilized in myopia intraocular refractive surgery is constantly facing challenges in terms of uveal biocompatibility. Inspired by the "sandwich theory", this study proposes the hypothesis that fibronectin (FN) can improve biocompatibility, and then creatively constructs a selective in-situ trap FN strategy. Specifically, PIOL surfaces with amide bonds covalently linking collagen were prepared, where selectivity was achieved by s
