A Planar Culture Model of Human Absorptive Enterocytes Reveals Metformin Increases Fatty Acid Oxidation and Export.

BACKGROUND & AIMS: Fatty acid oxidation by absorptive enterocytes has been linked to the pathophysiology of type 2 diabetes, obesity, and dyslipidemia. Caco-2 and organoids have been used to study dietary lipid-handling processes including fatty acid oxidation, but are limited in physiological relevance or preclude simultaneous apical and basal access. Here, we developed a high-throughput planar human absorptive enterocyte monolayer system for investigating lipid handling, and then evaluated the role of fatty acid oxidation in fatty acid export, using etomoxir, C75, and the antidiabetic drug m

Agmatine ameliorates sepsis-related intestinal injury via the AhR-STAT3-IL-10 pathway.

BACKGROUND: Sepsis-related intestinal injury is essential in multi-organ dysfunction induced by the systemic inflammatory response. Agmatine (AGM) has anti-inflammatory, antioxidative, and immunomodulatory properties. However, no reports of AGM alleviating sepsis-related intestinal injury have been found. Therefore, this study intends to investigate the protective effects of AGM on sepsis-related intestinal injury and its potential mechanism. METHODS: We first established a sepsis model by lipopolysaccharide (LPS) or cecum ligation perforation (CLP), and then used H&E staining, transmission e

Uptake of Micro- and Nano-Sized Plastic Particles in Human Cells: Size-Dependent Endocytosis and Cellular Responses.

Micro- and nanoplastics (MNPs) are increasingly detected in human tissues, yet the mechanisms governing their cellular uptake remain poorly understood. This study evaluated size-dependent cellular association of polystyrene MNPs (0.25-1.12 μm) across seven human cell lines representing skin, intestine, lung, kidney, liver, brain, and vascular endothelium. No cytotoxicity was observed in six cell lines after 24 or 48 h at concentrations up to 100 μg/mL, and epithelial and endothelial barrier integrity remained unaffected. Several cell types showed increased metabolic activity after 48 h. Cell-a

Intestinal absorption of S-nitrosothiols: Permeability and transport mechanisms.

S-Nitrosothiols, a class of NO donors, demonstrate potential benefits for cardiovascular diseases. Drugs for such chronic diseases require long term administration preferentially through the oral route. However, the absorption of S-nitrosothiols by the intestine, which is the first limiting barrier for their vascular bioavailability, is rarely evaluated. Using an in vitro model of intestinal barrier, based on human cells, the present work aimed at elucidating the mechanisms of intestinal transport (passive or active, paracellular or transcellular pathway) and at predicting the absorption site

Cannabidiol and Terpene Formulation Reducing SARS-CoV-2 Infectivity Tackling a Therapeutic Strategy.

UNLABELLED: In late 2019, COVID-19 emerged in Wuhan, China. Currently, it is an ongoing global health threat stressing the need for therapeutic compounds. Linking the virus life cycle and its interaction with cell receptors and internal cellular machinery is key to developing therapies based on the control of infectivity and inflammation. In this framework, we evaluate the combination of cannabidiol (CBD), as an anti-inflammatory molecule, and terpenes, by their anti-microbiological properties, in reducing SARS-CoV-2 infectivity. Our group settled six formulations combining CBD and terpenes pu

Ibuprofen, Flurbiprofen, Etoricoxib or Paracetamol Do Not Influence ACE2 Expression and Activity In Vitro or in Mice and Do Not Exacerbate In-Vitro SARS-CoV-2 Infection.

SARS-CoV-2 uses the human cell surface protein angiotensin converting enzyme 2 (ACE2) as the receptor by which it gains access into lung and other tissue. Early in the pandemic, there was speculation that a number of commonly used medications-including ibuprofen and other non-steroidal anti-inflammatory drugs (NSAIDs)-have the potential to upregulate ACE2, thereby possibly facilitating viral entry and increasing the severity of COVID-19. We investigated the influence of the NSAIDS with a range of cyclooxygenase (COX)1 and COX2 selectivity (ibuprofen, flurbiprofen, etoricoxib) and paracetamol o

Deglycosylation Enhances the Toxicity of Plant-Derived Glycosyl Bisphenol A during Gastrointestinal Digestion.

Bisphenol A (BPA) is one of the most used plasticizers and a known endocrine disrupting chemical. In plants, BPA is easily conjugated to form conjugates such as glycosyl BPA, and humans can be exposed to such conjugates when ingesting BPA-contaminated plant foods. This study considered the potential of deconjugation of glycosyl BPA in a simulated human gastrointestinal tract system and evaluated the subsequent changes in biological activity using the Caco-2 colon cell model. Glycosyl BPA was found in both soil and lettuce with concentrations ranging from not detected (ND) to 82.51 ng/g dry wei

[Prediction of drug intestinal absorption and drug-induced intestinal toxicity with the use of human crypt-derived intestinal stem cells and their differentiated cells].

For the development of orally-administered drugs, quantitative prediction of drug intestinal absorption and drug-induced intestinal toxicity is one of the essential evaluation items. However, the current experimental systems have some critical problems such as large inter-species difference in animal studies and different gene expression levels between Caco-2 cells and human intact intestinal tissue. Thus, the construction of novel experimental systems which can reflect human intestinal physiology has been desired. We established stable 3D culture of intestinal stem cells in Matrigel originall…

Serotonin modifies cytoskeleton and brush-border membrane architecture in human intestinal epithelial cells.

Serotonin or 5-hydroxytryptamine (5-HT) influences numerous functions in the gastrointestinal tract. We previously demonstrated that 5-HT treatment of Caco-2 cells inhibited Na(+)/H(+) exchangers (NHE) and Cl(-)/OH(-) exchange activities via distinct signaling mechanisms. Since regulation of several ion transporters such as NHE3 is influenced by intact cytoskeleton, we hypothesized that 5-HT modifies actin cytoskeleton and/or brush-border membrane architecture via involvement of signaling pathways. Ultrastructural analysis showed that 5-HT (0.1 muM, 1 h) treatment of Caco-2 cells caused the ap…
