Complete Genome and Molecular Characterization of a New Cyprinid Herpesvirus 2 (CyHV-2) SH-01 Strain Isolated from Cultured Crucian Carp.

Cyprinid herpesvirus 2 (CyHV-2) is a causative factor of herpesviral hematopoietic necrosis (HVHN) in farmed crucian carp (Carassius carassius) and goldfish (Carassius auratus). In this study, we analyzed the genomic characteristics of a new strain, CyHV-2 SH-01, isolated during outbreaks in crucian carp at a local fish farm near Shanghai, China. CyHV-2 SH-01 exhibited a high sensitivity to goldfish and crucian carp in our previous research. The complete genome of SH-01 is 290,428 bp with 154 potential open reading frames (ORFs) and terminal repeat (TR) regions at both ends. Compared to the se

Host Range of Carp Edema Virus (CEV) during a Natural Mortality Event in a Minnesota Lake and Update of CEV Associated Mortality Events in the USA.

Mass mortality events of common carp (Cyprinus carpio, carp) associated with carp edema virus (CEV) alone or in coinfections with koi herpesvirus (KHV), is an emerging issue. Despite recent outbreaks of CEV in wild carp populations, the host range of North American species has not been well studied. To that end, we intensively sampled carp (n = 106) and co-habiting native fish species (n = 5 species; n = 156 total fish) from a CEV-suspect mass-mortality event of carp in a small Minnesota lake (Lake Swartout). Additionally, fecal and regurgitant samples (n = 73 each) from double-crested cormora

Molecular identification of glucose transporter 4: The responsiveness to starvation, glucose, insulin and glucagon on glucose transporter 4 in common carp (Cyprinus carpio L.).

Glucose transporter 4 (GLUT4) is comprehensively investigated in mammals, while the comparative research of GLUT4 in common carp is deficient. To investigate the function of GLUT4, carp glut4 was first isolated. The open reading frame of carp glut4 was 1518 bp in length, encoding 505 amino acids. A high-sequence homology was identified in carp and teleost, and the phylogenetic tree displayed that the carp GLUT4 was clustered with the teleost. A high level of glut4 mRNA was analysed in fat, red muscle and white muscle. After fasting treatment, glut4 mRNA expression was increased significantly i

Innate cell-mediated cytotoxicity of CD8+ T cells against the protozoan parasite Ichthyophthirius multifiliis in the ginbuna crucian carp, Carassius auratus langsdorfii.

Cytotoxic T cells are known to have the ability to kill microbe-infected host cells, which makes them essential in the adaptive immunity processes of various vertebrates. In this study, we demonstrated innate cell-mediated cytotoxicity of CD8+ T cells against protozoan parasites found in the ginbuna crucian carp. When isolated effector cells such as CD8+, CD4+ (CD4-1+), or CD8- CD4- (double-negative, DN), from naïve ginbuna crucian carp were co-incubated with target parasites (Ichthyophthirius multifiliis), CD8+ cells from the kidney and gill showed the highest cytotoxic activity. Proteomic and metabolomic basis for improved textural quality in crisp grass carp (Ctenopharyngodon idellus C.et V) fed with a natural dietary pro-oxidant.

Reactive oxygen species (ROS) regulate metabolism and chemical composition of various tissues. To understand how ROS affect the textural quality of fish muscle, we performed a multi-omics analysis on an established crisp grass carp model fed with a natural pro-oxidant faba bean. ROS levels were systemically and significantly increased up to three-fold in crisp grass carp, improving the muscle texture. Lipid metabolism was significantly enhanced up to five-fold in muscle and liver possibly to compensate the impaired carbohydrate metabolism of these tissues, but this caused further local ROS pro

The regulatory effects of pyridoxine deficiency on the grass carp (Ctenopharyngodon idella) gill barriers immunity, apoptosis, antioxidant, and tight junction challenged with Flavobacterium columnar.

The effects of dietary pyridoxine (PN) on the gill immunity, apoptosis, antioxidant and tight junction of grass cap (Ctenopharyngodon idella) were investigated in this study. Fish were fed semi-purified diets containing graded levels of PN for 10 weeks, and then challenged with Flavobacterium columnare by bath immersion exposure for 3 days. The results indicated that compared with the optimal PN level, PN deficiency resulted in a decline in the antimicrobial compound production of gill. In addition, PN deficiency up-regulated the pro-inflammatory cytokines and down-regulated the anti-inflammat

Impact of Short-Term Fasting on The Rhythmic Expression of the Core Circadian Clock and Clock-Controlled Genes in Skeletal Muscle of Crucian Carp (Carassius auratus).

The peripheral tissue pacemaker is responsive to light and other zeitgebers, especially food availability. Generally, the pacemaker can be reset and entrained independently of the central circadian structures. Studies involving clock-gene expressional patterns in fish peripheral tissues have attracted considerable attention. However, the rhythmic expression of clock genes in skeletal muscle has only scarcely been investigated. The present study was designed to investigate the core clock and functional gene expression rhythms in crucian carp. Meanwhile, the synchronized effect of food restricti

Does information on novel identified autoantibodies contribute to predicting the progression from undifferentiated arthritis to rheumatoid arthritis: a study on anti-CarP antibodies as an example.

BACKGROUND: The presence of autoantibodies is considered an important characteristic of rheumatoid arthritis (RA); therefore, both anticitrullinated protein antibodies (ACPA) and rheumatoid factor (RF) are included in the 2010 classification criteria for rheumatoid arthritis (RA). However, a considerable number of RA patients lack both these autoantibodies. Recently, several novel autoantibodies have been identified but their value for the classification of RA patients is unclear. Therefore, we studied the value of novel autoantibodies using the presence of anticarbamylated protein (anti-CarP)

Identification of reproduction-related genes and SSR-markers through expressed sequence tags analysis of a monsoon breeding carp rohu, Labeo rohita (Hamilton).

Labeo rohita (Ham.) also called rohu is the most important freshwater aquaculture species on the Indian sub continent. Monsoon dependent breeding restricts its seed production beyond season indicating a strong genetic control about which very limited information is available. Additionally, few genomic resources are publicly available for this species. Here we sought to identify reproduction-relevant genes from normalized cDNA libraries of the brain-pituitary-gonad-liver (BPGL-axis) tissues of adult L. rohita collected during post preparatory phase. 6161 random clones sequenced (Sanger-based) f

Antibody screening identifies 78 putative host proteins involved in Cyprinid herpesvirus 3 infection or propagation in common carp, Cyprinus carpio L.

Cyprinid herpesvirus 3 (CyHV-3) is the aetiological agent of a serious and notifiable disease afflicting common and koi carp, Cyprinus carpio L., termed koi herpesvirus disease (KHVD). Significant progress has been achieved in the last 15 years, since the initial reports surfaced from Germany, USA and Israel of the CyHV-3 virus, in terms of pathology and detection. However, relatively few studies have been carried out in understanding viral replication and propagation. Antibody-based affinity has been used for detection of CyHV-3 in enzyme-linked immunosorbent assay and PCR-based techniques, a

Ferric iron availability and species identity jointly shape the metabolic profiles and spoilage potentials of grass carp spoilage bacteria: A combined metabolomic and physicochemical evaluation.

Ferric iron is essential for bacterial growth and spoilage-associated phenotypes, yet its specific influences on the metabolism and spoilage potential of fish spoilage bacteria remain unclear. Therefore, this study aimed to investigate the metabolic and spoilage potential changes of grass carp spoilage bacteria under varying ferric iron accessibilities, using combined metabolomic and physicochemical detection. The results showed that high ferric iron accessibility stimulated bacterial carbohydrate metabolism (glycolysis and tricarboxylic acid cycle) and fatty acid β-oxidation. Conversely, the

Effects of grilling process on the formation pattern of heterocyclic amines and advanced glycation end products and physicochemical quality of premade grilled grass carp fish blocks.

This study found a temperature-dependent pattern in heterocyclic amines (HAs) and advanced glycation end-products (AGEs) formation, as well as the physicochemical quality attributes of pre-made grilled grass carp. Raising the center temperature from 50 °C to 80 °C increased total HAs by 20% and total AGEs (CML + CEL) by 107%. At grilling temperatures of 220-280 °C, HAs formation dominated, with tryptophan, glyoxal, and creatine/creatinine as key precursors. In contrast, AGE formation prevailed at 310 °C, primarily involving lysine and methylglyoxal. Notably, grilled fish skin contained only 23

Redox imbalance and NF-κB activation drives 1,3-diphenylguanidine (DPG)-induced hepatotoxicity in grass carp (Ctenopharyngodon idella): HSP70 as a protective therapeutic target.

As a widely utilized rubber vulcanization agent, 1,3-diphenylguanidine (DPG) has been identified as inherently toxic to aquatic organisms. The study elucidates the molecular pathways of DPG-induced hepatotoxicity and evaluates potential therapeutic strategies. In this study, grass carp exposed to DPG exhibited mild to moderate hepatic edema, sinusoidal congestion, with serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels elevated by 25.0 % and 46.3 %, respectively. Furthermore, a 2.46-fold increase in TUNEL-positive hepatocytes was observed following DPG exposure. R

Combined exposure to cypermethrin and sulfamethoxazole is associated with muscle injury in grass carp through oxidative stress, endoplasmic reticulum stress, MAPK activation, autophagy, and apoptosis.

The co-occurrence of pesticides and antibiotics in aquatic environments has raised increasing concern because of their potential effects on aquatic organisms. Cypermethrin (CMN) and sulfamethoxazole (SMZ) are representative pyrethroid pesticides and sulfonamide antibiotics, respectively, but their combined effects on fish muscle remain poorly understood. In this study, grass carp (Ctenopharyngodon idella) were exposed to CMN (0.65 μg/L), SMZ (0.30 μg/L), or their combination for 42 days. Network toxicology, histopathology, oxidative stress assays, Western blotting, protein-protein interaction

Dietary Ellagic Acid Mitigates High Stocking Density-Induced Oxidative Stress and Immune Suppression in Common Carp (Cyprinus carpio): An In Vivo and In Silico Study.

This study examined the effects of dietary ellagic acid (EA) on growth performance, survival, immune responses, and oxidative status of common carp (Cyprinus carpio) exposed to high stocking density (HSD). A 2 × 3 factorial design was used, with two stocking densities (25 and 100 kg/m3) and three EA levels (0, 50, and 100 mg/kg diet). HSD significantly reduced growth performance, as indicated by lower weight gain and specific growth rate (SGR), increased feed conversion ratio (FCR), and decreased survival (p < 0.05). It also suppressed immune responses, including white blood cell count (WBC),
