Social novelty preference-the tendency to interact more with unfamiliar than familiar conspecifics-is conserved across species and disrupted in disorders such as autism spectrum disorder, schizophrenia, and social anxiety. While the hippocampus and related circuits are known to encode social recognition memory, the mechanisms that translate familiarity signals into behavioral differences remain unclear. Here, we show that male mice exhibit a robust preference for engaging with unfamiliar over familiar conspecifics. Using c-Fos labeling, RNAscope, immunohistochemistry, and fiber photometry, we

The substantia nigra pars reticulata (SNr), the principal output nucleus of the basal ganglia, is traditionally viewed as a binary gate that permits actions through disinhibition. However, this framework fails to explain the fluid, high-dimensional control required for skilled behavior. Using high-resolution 3D kinematic tracking and automated behavioral classification, we show that SNr population activity paradoxically increases during skilled forelimb reaching and scales with motif-specific kinematics. Systematic optogenetic and chemogenetic perturbations reveal that the SNr bidirectionally

Alcohol use disorder (AUD) lacks effective brain-targeted treatments. Here, using mouse models, we show that glucagon-like peptide-1 receptor (GLP-1R) signaling in the dorsal lateral septum (dLS) regulates alcohol consumption and reward. Systemic administration of the GLP-1R agonist liraglutide decreased alcohol intake and ethanol-evoked dopamine release in the nucleus accumbens, requiring GLP-1R expression in the dLS. Alcohol consumption suppressed dLSGLP-1R neuronal activity, whereas liraglutide prevented alcohol-induced suppression of transient calcium dynamics. Inactivation of these neuron

Chronic stress disrupts emotional homeostasis, leading to resilience or susceptibility through unclear mechanisms. Here, we identify a dual-pathway mechanism in the dorsomedial prefrontal cortex (dmPFC)-lateral habenula (LHb) circuit in male mice, where glutamatergic and GABAergic projections coordinately regulate stress responses. Chronic stress differentially modulates these pathways, inducing susceptibility-specific hyperactivity in dmPFC-innervated LHb neurons by potentiating excitatory and suppressing inhibitory inputs, disrupting long-range excitatory/inhibitory (E/I) balance. Chemogenet

Stress modulates itch, with acute stress suppressing and chronic stress exacerbating pruritus, yet the underlying neural mechanisms remain unclear. In this study, we investigate the role of lateral hypothalamic area (LHA) neurons in stress-induced itch modulation. Using neural-activity-dependent genetic labeling and chemogenetic tools, we selectively engaged a stress-sensitive population of LHA neurons (LHAstress-TRAP neurons). Transient activation of these neurons elicited anxiety-like behavior and place aversion while suppressing both acute (chloroquine-induced) and chronic (psoriatic) itch;

The ventromedial hypothalamus (VMH) is a key regulator of energy homeostasis, linking central neural activity to peripheral metabolic function. Within the VMH, neurons expressing steroidogenic factor-1 (SF-1; VMHSF1) are essential for regulating energy expenditure, yet the projection-defined pathways through which they differentially control distinct adipose depots remains unclear. Here, we use projection-specific optogenetic and chemogenetic manipulations in SF1-Cre mice to identify two anatomically and functionally distinct VMHSF1 output pathways with complementary metabolic roles. The vagus nerve regulates inflammation via sensory-motor arcs. While vagal sensory neurons relay signals from within the body (interoception), whether external inputs can regulate visceral neuroimmune responses (exteroception) remains poorly understood. We unveiled a skin-lung reflex by which sensory neurons from the auricular skin suppressed Alternaria alternata-induced allergic airway inflammation. Viral and chemical neural tracing revealed distinct sensory projections from the vagal ganglia to the auricular skin. Pharmacologic, chemogenetic, and optogenetic activation of auricular transient

Regeneration is triggered by cells dying due to various types of injuries. However, it remains unclear whether different types of cell death elicit distinct regeneration responses. Here, we systematically profile dynamic transcriptional responses of macrophages and lateral line cells to different cell death modalities during zebrafish sensory hair cell regeneration. We show that chemogenetically induced programmed hair cell death triggers a diminished inflammatory response compared to pharmacologically induced cell lysis, characterized by minimal neutrophil recruitment, distinct transcriptiona

Methamphetamine (METH) is a highly addictive psychostimulant, yet its addiction mechanisms remain unclear. Oxytocin (OXT), a neuropeptide, shows promise in reducing METH addiction, but how OXT exerts its effects is poorly understood. Using conditioned place preference (CPP), we first found that intranasal OXT other than Arginine Vasopressin (AVP) administration suppressed METH-CPP in mice, which could be reversed by OXT receptors (OXTRs) blockade in the nucleus accumbens (NAc) core. Activating OXTRs in the NAc core similarly reduced METH-CPP. Then, we found repeated METH exposure inhibited oxy

Atypical sensory integration and contextual learning are common symptoms in autism spectrum disorder (ASD), but how sensory circuits are affected remains elusive. Here we focused on the early visual information processing, and performed in vivo two-photon calcium imaging and pupillometry of mice engaged in an implicit learning task in stable and volatile visual contexts. Wild-type (WT) mice show stimulus-specific contextual modulation of the visual responses in the superior colliculus (SC) and pupil dynamics, whereas SCN2A-haploinsufficient ASD-model mice exhibit abnormal modulation patterns.

Blue light has been shown to impair spatial memory in mice. However, the underlying neural circuit mechanisms of this impairment remain elusive. Our study revealed that 600-lux blue light disrupted spatial memory retrieval in male mice through the secondary visual cortex (V2M) → the prelimbic cortex (PrL) neural pathway. Specifically, viral tracing and multichannel fiber optic recordings demonstrated that glutamatergic V2M neurons (V2MGlu) projected to GABAergic neurons in the PrL (PrLGABA), thereby inhibiting the excitability of glutamatergic PrL neurons (V2MGlu→PrLGABA→Glu). The selective ab

Methamphetamine (METH) addiction involves escalating intake with strong cue reactivity, and high relapse risk, yet its neural mechanism remains unclear. Using c-Fos mapping and machine learning, we identified the claustrum (CLA), a subcortical region reciprocally connected with the anterior cingulate cortex (ACC), as key mediators of both METH taking and seeking in self-administering male rats. Chemogenetic inhibition of CLA suppressed both drug consumption and cue-induced reinstatement, while ACC inhibition selectively reduced drug-seeking. Circuit tracing and manipulation revealed that the C

Animal models of drug addiction simulate core behaviors such as active drug seeking, compulsive drug use, and relapse, thereby establishing a system for studying the mesolimbic reward circuit. Rats and mice serve as the most commonly used model organisms in this field. Mainstream models are developed based on the principles of operant conditioning and classical conditioning, among which the self-administration paradigm is regarded as the "gold standard" because it can directly simulate active drug seeking and compulsive drug use. These models demonstrate face validity, construct validity, and

Neuronal plasticity in the central amygdala (CeA) is essential for modulating feeding behaviors and emotional responses, potentially influencing reactions to Deoxynivalenol (DON). Acute oral administration of DON elicits a dose-responsive reduction in food intake, accompanied by pronounced alterations in locomotor activity and feeding frequency. This study investigates circuitry adaptations that mediate DON's effects on feeding, by targeting of GABA neurons in the CeA. Following exposure to DON, an increase in connectivity between the paraventricular nucleus of the thalamus (PVT) and CeAGABA n

Antipsychotic drugs have severe metabolic side effects. Acute use can induce hypothermia, while chronic use often leads to weight gain and associated disorders. However, no treatment is currently available for drug-induced hypothermia, and weight control measures lack evidence for long-term effectiveness. Here we demonstrate that a glucagon-like peptide 2 analogue, teduglutide, effectively prevents olanzapine-induced hypothermia and weight gain, and restores glucose tolerance and insulin sensitivity in mice. Mechanistically, olanzapine suppresses prodynorphin-expressing neurons in the ventrome

Spontaneous recovery following an ischemic stroke is often limited, largely attributed to age-related decline in neuroplasticity. To overcome this, we demonstrated that ectopic expression of a cocktail of transcriptional factors (Oct4, Sox2, and Klf4, referred to as OSKTFs) reset developmental decline of epigenetic signatures in adult corticospinal neurons, without affecting their spinal projection patterns and function in controlling skilled locomotion. Corticospinal expression of OSKTFs had moderate effects on promoting collateral sprouting of the corticospinal tract axons and recovery of sk

Central glucagon-like peptide-1 (GLP-1), secreted by a distinct population of nucleus tractus solitarius neurons, suppresses feeding but the exact mechanisms of action in the brain remain unclear. Here, we investigate a descending circuit formed by GLP-1 receptor (GLP-1R) neurons in the paraventricular hypothalamic nucleus (PVNGLP-1R) projecting to the dorsal vagal complex (DVC) of the brain stem in mice. PVNGLP-1R→DVC synapses release glutamate and are augmented by GLP-1. Chemogenetic activation of PVNGLP-1R→DVC suppresses feeding. Under an energy deficit (that is, hunger) state, synaptic str

Deficits in attention are common across a range of neuropsychiatric disorders. A multitude of brain regions, including the frontal cortex (FC) and locus coeruleus (LC), have been implicated in attention. Regulators of these brain regions at the molecular level are not well understood, but might elucidate underlying mechanisms of disorders with attentional deficits. To probe this, we used chemogenetic stimulation of neurons in the LC with axonal projections to the FC, and subsequent bulk RNA-sequencing from the mouse FC. We found that stimulation of this circuit caused an increase in transcript

Individuals with Down syndrome (DS) have a higher prevalence of obesity compared to the general population. Conventionally, this has been attributed to endocrine issues and lack of exercise. However, deficits in neural reward responses and dopaminergic disturbances in DS may be contributing factors. To investigate this, we focused on a mouse model (Ts65Dn) bearing some triplicated genes homologous to trisomy 21. Through detailed meal pattern analysis in male Ts65Dn mice, we observed an increased preference for energy-dense food, pointing towards a potential "hedonic" overeating behavior. Moreo

As a central part of the mammalian brain, the prefrontal cortex (PFC) has been implicated in regulating cocaine-induced behaviors including compulsive seeking and reinstatement. Although dysfunction of the PFC has been reported in animal and human users with chronic cocaine abuse, less is known about how the PFC is involved in cocaine-induced behaviors. By using two-photon Ca2+ imaging to simultaneously record tens of intact individual networking neurons in the frontal association cortex (FrA) in awake male mice, here we report that a systematic acute cocaine exposure decreased the FrA neural
