Chorionic villi chromosome analysis was performed on 1,186 cases of induced abortion between the 5th and 11th week of gestation. The total incidence of major chromosome abnormalities, including numerical and structural chromosomal changes as well as mosaics and polyploids, was 4.5% (53 cases). The most common abnormalities were trisomy 21 (5 cases), trisomy 16 (4 cases), and monosomy X (4 cases). The incidence of chromosome abnormalities increased with the advancing age of the mother.

To determine the mechanism for the coexistence of XX chromosomal maleness and true hermaphroditism in the same family, we performed cytogenetic and molecular genetic analyses, using DNA probes from the short arm of the Y chromosome. These studies excluded the following possible mechanisms: (1) an inherited, mitotically unstable Y chromosome that results in chromosomal mosaicism, (2) an inherited Y-to-X or Y-autosomal translocation, (3) recurrent Y-to-X translocation, and (4) incomplete inactivation of the X chromosomal homolog for the testicular determining factor. We conclude that the disorde

Clonal chromosome abnormalities were found in cultured fibroblasts from three sibs and one sporadic case with porokeratosis of Mibelli. Chromosome 3 especially involved region p12-14. This region includes the most common fragile site in humans, and the proximal region of chromosome 3 short arm is involved in a variety of neoplastic conditions. We conclude that porokeratosis of Mibelli, an autosomal dominant disorder, is associated with chromosomal instability. Porokeratosis of Mibelli is known to also be associated with increased susceptibility to malignant disease. The chromosome instability

Rate division of human lymphocytes was studied in 85 healthy siblings and 142 normal non-related individuals using sister chromatid differential staining in standard conditions of cell culturing. It was shown that proliferation of male lymphocytes exceeds that of female cells. This sexual dimorphism does not depend on the time of fixation of cell cultures and probably is conditioned by differential chromosomal constitution of the two sexes. The study of sex chromosome mosaics revealed rate modification of cell proliferation in the line: 45,X greater than 46,XY greater than 46,XX. The possibili

A sessile adenoma of the left flexure of the colon was studied after surgical colectomy. Specimens were obtained for complete histologic evaluation. The tumor consisted of glandular tubes with decreased mucin production and a papillary structure on the luminal aspect. The muscularis mucosa was not involved; there was no carcinomatous focus. Cytogenetic study was carried out on 56 cells; none was normal, 77% were hyperdiploid (52-87 chromosomes), 16% were hypodiploid (18-39 chromosomes), and 7% were paradiploid. The supernumerary chromosomes were chromosomes #3, #6, #13, #19, and #20; chromosom…

Human sperm chromosomes were studied in a man heterozygous for a paracentric inversion of chromosome 7 (q11q22). The pronuclear chromosomes were analysed after in vitro penetration of golden hamster (Mesocricetus auratus) eggs. Ninety-four sperm chromosome spreads were examined, of which 34 contained the normal number 7 chromosome and 59 the inverted 7. This segregation was significantly different from the expected 1:1 ratio. The number of X- to Y-bearing sperm was 48 and 46 respectively. No sperm contained a recombinant chromosome caused by a crossover within the inversion. The frequency of c…

During oocyte maturation and zygote development chromosomes undergo cyclic changes, alternaing the condensed and decondensed states. In oocytes, zygotes and perhaps in other cells, the chromosome cycle appears to be controlled in same way by common cytoplasmic factors. Among them, maturation-promoting factor (MPF) plays a particularly important role, although the germinal vesicle substances and cytoplasmic membrane vesicles are indispensable for the chromosomal changes. MPF precursor is stored in fully grown oocytes of most species, but replenishing MPF after its fall during cell cycles requir…

In chromosome sets of three patients with Turner's syndrome non-fluorescent Y-chromosomes of normal size were found in part of the cells. C-, Q- and G-techniques have shown that they were dicentric Yp-chromosomes, resulting from a junction of long arms of two Y-chromosomes with simultaneous loss of the entire distal fluorescent segments. It is supposed that in some cases the non-fluorescent Y-chromosomes, previously described in literature, are as a matter of fact undiscernible dicentric Yp-chromosomes.

A fetus was identified by prenatal cytogenetic diagnosis as having a karyotype 46,XY,r(13) (p11q13). Termination of the pregnancy yielded a severely malformed fetus. Fetal abnormalities included anencephaly, imperforate anus and urethral meatus, severe talipes, syndactyly, cardiac defects and other anomalies. Confirmatory studies on cultured placental villi cells indicated a second cell line, 46,XY, -13,+13qter leads to cen::13q13 leads to qter. This cell line was not detectable in cells derived from the fetus despite extensive studies. It seems likely that the two cell lines arose simultaneou

Two ring chromosomes 13 were studied by means of various cytogenetic techniques in two psychomotorically retarded male infants. Common features of our patients include microcephalia, hypertelorism, wide and prominent nose bridge, and cryptorchism. Various configurations of the aberrant chromosomes could be identified in cultured skin fibroblasts and peripheral lymphocytes from both patients. Chromosome heteromorphisms and analysis of silver stained nucleolus organizer regions (Ag-NOR) substantiated the parental origin of the ring chromosomes. The more severely affected patient showed a break p…

Premature chromosome pulverization (PCC) or prophasing is a much misunderstood cytological entity. It must be separated from chromosome damage caused by a number of chemical, physical and biological agents. Prophasing is observed in fused cells in which one of the constituent cells must be in metaphase and another in interphase. The morphology of the "pulverized" interphase nucleus will depend on the phase of the cell cycle in which the interphase cell was in when exposed to a substance present in the cytoplasm of the metaphase cell leading to "prophasing". Prophasing is a normal cellular phen

For medical genetic counseling, estimating the chance of a child being born with chromosome abnormality is crucially important. Cytogenetic diagnostics of parents with a balanced karyotype are a special case. Such chromosome rearrangements cannot be detected with comprehensive chromosome screening. In the current paper, we consider chromosome diagnostics in two cases of chromosome rearrangement in patients with balanced karyotype and provide the results of a detailed analysis of complex chromosomal rearrangement (CCR) involving three chromosomes and a small supernumerary marker chromosome (sSM…

Pathogenic multidrug-resistant bacteria with hybrid chromosomes have emerged as a significant global healthcare threat. These include the pandemic Escherichia coli ST1193, the product of homologous recombination events involving two phylogenetically distant strains of E. coli, in which mutant alleles of the widely separated genes, gyrA and parC, generating high-level fluoroquinolone resistance were acquired. The mechanisms and frequency of hybrid formation are poorly understood. We developed a robust hybrid selection procedure and applied it to 118 clinical UTI isolates of E. coli mixed with s…

Common fragile sites (CFSs) are large genomic loci that are frequently deleted under replication stress and are thought to play a role in carcinogenesis as well as developmental disorders during early embryogenesis. They often appear as gaps or breaks in mitotic chromosomes by conventional optical microscopy and are also loci that are replicated in mitosis in a process called mitotic DNA synthesis (MiDAS). However, we still have a very poor understanding of the potential mechanisms underlying their genomic instability. We recently found that in normal mitotic chromosomes, there is a two-level …

Sex chromosomes are involved in three major empirical patterns: (i) Y (or W) chromosomes are often nonrecombining and degenerate; (ii) heterogametic offspring (XY or ZW) from interspecific crosses are more often sterile or inviable compared with homogametic offspring (Haldane's rule); and (iii) the X (or Z) has a disproportionately large effect on reproductive isolation between species compared with autosomes (the large X effect). Each observation has received its own tailored explanation involving multiple genetic and evolutionary causes. In this work, we show that these empirical patterns al…
