Fifty years of development of neuroscientific insights into oro-facial pain and its control.

When the Journal of Oral Rehabilitation was established in 1973, there was very limited understanding of the mechanisms underlying neurally based functions, including those unique to the face, mouth and jaws (e.g. dental pain, taste, chewing, swallowing and salivation). Since that time, technological and other advances have led to new insights into the structure, connectivity and function of cranial nerves and areas of the central nervous system (CNS) involved in oro-facial functions and disorders or related functions (e.g. cognition, emotion, stress, consciousness, sleep, learning and memory)

GLP-1 and dual GIP/GLP-1 receptor agonists' psychopharmacology and putative neuropsychiatric associated effects: a Bradford Hill-informed, systematic, evaluation.

PURPOSE OF REVIEW: This review evaluates evidence linking both GLP-1 and GIP/GLP-1RAs' exposure to a range of neuropsychiatric outcomes using the Bradford Hill criteria for causal inference. RECENT FINDINGS: Glucagon-like peptide-1 (GLP-1) and the dual glucose-dependent insulinotropic polypeptide-GIP/GLP-1 receptor agonists (RAs) are incretin-based medications widely prescribed for type 2 diabetes mellitus and obesity, with demonstrated cardiometabolic benefit and rapidly expanding population exposure. In parallel, post-marketing surveillance and emerging translational research have raised qu

Folic Acid Metabolism and Its Impact on Neurogenesis: Molecular Mechanisms and Therapeutic Potential.

Folic acid (FA), a vital water-soluble B vitamin, is indispensable for the development and maintenance of the nervous system. By modulating one-carbon metabolism, FA critically influences DNA and RNA synthesis, methylation reactions, and cell division, thereby profoundly affecting neurogenesis. Neurogenesis, encompassing the proliferation, differentiation, and maturation of neural stem cells, is tightly regulated by FA through its role in DNA synthesis and methylation. Impaired neurogenesis is implicated in various neurological disorders, highlighting its critical role in cognitive function, b

Neuromodulation in sleep disorders.

Sleep disorders are a growing global health issue with profound consequences on cardiovascular health, metabolic functioning, cognitive output and overall quality of life. Although there have been numerous clinical advances in brain-based treatments for sleep disorders, there are still a number of sleep disorders that are not well understood and need new treatment options to address the underlying neural mechanisms of sleep-wake regulation. Recent advances in neuroscience have highlighted neuromodulation as a promising treatment option for the complex neural mechanisms of sleep-wake dysfunctio

Advances in non-human primate models for Alzheimer's disease research.

Alzheimer's disease (AD) is the most common progressive neurodegenerative disease. It is characterized by cognitive decline and brain pathology, including amyloid-β (Aβ) plaques, neurofibrillary tangles, and neuroinflammation. While rodent models have contributed to our understanding of a multiplicity of disease mechanisms, their limitations in replicating human age-related neurodegenerative diseases pose challenges for translating research findings to the clinical setting. The common marmoset (Callithrix jacchus), a small non-human primate, has emerged as a promising model for neuroscience an

40 Hz light stimulation restores early brain dynamics alterations and associative memory in Alzheimer's disease model mice.

Visual gamma entrainment using sensory stimuli (vGENUS) is a promising non-invasive therapeutic approach for Alzheimer's disease (AD), showing efficacy in improving memory function. However, its mechanisms of action remain poorly understood. Using young AppNL-F/MAPT double knock-in (dKI) mice, a model of early AD, we examined brain dynamics alterations before amyloid plaque onset. High-density EEG recordings and metrics from fields outside neuroscience were used to assess brain dynamics fluidity-a measure of the brain's ability to transition between activity states. We revealed that dKI mice e

Molecularly-guided spatial proteomics captures single-cell identity and heterogeneity of the nervous system.

Single-cell proteomics is an emerging field with significant potential to characterize heterogeneity within biological tissues. It offers complementary insights to single-cell transcriptomics by revealing unbiased proteomic changes downstream of the transcriptome. Recent advancements have focused on enhancing proteome coverage and depth, mostly in cultured cell lines, and a few recent studies have explored the potential of analyzing tissue micro-samples but were limited to homogenous peripheral tissues. In this current work, we utilize the power of spatial single cell-proteomics through immuno

[A study on the effects of transcranial direct current stimulation combined with motor imagery on brain function based on electroencephalogram and near infrared spectrum].

Motor imagery is often used in the fields of sports training and neurorehabilitation for its advantages of being highly targeted, easy to learn, and requiring no special equipment, and has become a major research paradigm in cognitive neuroscience. Transcranial direct current stimulation (tDCS), an emerging neuromodulation technique, modulates cortical excitability, which in turn affects functions such as locomotion. However, it is unclear whether tDCS has a positive effect on motor imagery task states. In this paper, 16 young healthy subjects were included, and the electroencephalogram (EEG) …

Pharmacotherapeutic strategies for the treatment of anorexia nervosa - novel targets to break a vicious cycle.

INTRODUCTION: Anorexia nervosa (AN) has one of the highest mortality rates of all mental illnesses. No approved pharmacological treatments exist for AN, but novel neurobiological targets show promise. AREAS COVERED: Studies show that in individuals with AN, there are alterations in brain neurotransmitter signaling, alongside associated mental rigidity and comorbid anxiety and depression. Available and new therapies could be used to improve alterations in neurobiology and behavior. This narrative review serves as a review of previously published literature assessing the efficacy of traditional

Functional disorders - new proposals for definition, psychosomatics, somatization.

Functional Disorders are common medical problems both in primary and in secondary health care. The mechanisms that cause symptoms such as primary pain, fatigue, dizziness are still unknown. Various classifications, including ICD-10 or DSM-5, describe these conditions differently, and new proposals are being developed e.g. in ICD-11, RDoC. Many controversies are evoked by lack of unequivocal explanatory theory. The early psychoanalytical concept has been modified by other explanations, such as immunological abnormalities, dysfunction of vegetative system and HPA axis, central sensitization, div

Positive hysteresis in emotion recognition: Face processing visual regions are involved in perceptual persistence, which mediates interactions between anterior insula and medial prefrontal cortex.

Facial emotion perception can be studied from the point of view of dynamic systems whose output may depend not only on current input but also on prior history - a phenomenon known as hysteresis. In cognitive neuroscience, hysteresis has been described as positive (perceptual persistence) or negative (fatigue of current percept) depending on whether perceptual switching occurs later or earlier than actual physical stimulus changes. However, its neural correlates remain elusive. We used dynamic transitions between emotional expressions and combined behavioral assessment with functional magnetic …

Epilepsy and brain network hubs.

Epilepsy is a disorder of brain networks. A better understanding of structural and dynamic network properties may improve epilepsy diagnosis, treatment, and prognostics. Hubs are brain regions with high connectivity to other parts of the brain and are typically situated along the brain's most efficient communication pathways, supporting large-scale brain wiring and many higher order neural functions. The visualization and analysis of hubs offers a perspective on regional and global network organization and can provide novel insights into brain disorders and epilepsy. By notably supporting the

Asymmetry in the Central Nervous System: A Clinical Neuroscience Perspective.

Recent large-scale neuroimaging studies suggest that most parts of the human brain show structural differences between the left and the right hemisphere. Such structural hemispheric asymmetries have been reported for both cortical and subcortical structures. Interestingly, many neurodevelopmental and psychiatric disorders have been associated with altered functional hemispheric asymmetries. However, findings concerning the relation between structural hemispheric asymmetries and disorders have largely been inconsistent, both within specific disorders as well as between disorders. In the present

Pharmacotherapeutic strategies for the treatment of anorexia nervosa - too much for one drug?

INTRODUCTION: Anorexia nervosa is a severe psychiatric illness and no medication has been approved for its treatment. This lack of biological treatments requires the development of new directions for pharmacological research. AREAS COVERED: There is modest but emerging evidence that dopamine D2 and serotonin 1A and 2A receptor agonistic and antagonist medication might be beneficial for weight gain, although the underlying mechanisms are uncertain. Improving quality of life including treating comorbid conditions is an additional important outcome measure, but this has not been well researched.

Recovering Brain Dynamics During Concurrent tACS-M/EEG: An Overview of Analysis Approaches and Their Methodological and Interpretational Pitfalls.

Transcranial alternating current stimulation (tACS) is increasingly used as a tool to non-invasively modulate brain oscillations in a frequency specific manner. A growing body of neuroscience research utilizes tACS to probe causal relationships between neuronal oscillations and cognitive processes or explore its capability of restoring dysfunctional brain oscillations implicated in various neurological and psychiatric disease. However, the underlying mechanisms of action are yet poorly understood. Due to a massive electromagnetic artifact, overlapping with the frequency of interest, direct ins

Obsessive-compulsive disorder.

Obsessive-compulsive disorder (OCD) is a highly prevalent and chronic condition that is associated with substantial global disability. OCD is the key example of the 'obsessive-compulsive and related disorders', a group of conditions which are now classified together in the Diagnostic and Statistical Manual of Mental Disorders, Fifth Edition, and the International Classification of Diseases, 11th Revision, and which are often underdiagnosed and undertreated. In addition, OCD is an important example of a neuropsychiatric disorder in which rigorous research on phenomenology, psychobiology, pharma

The possible role of the insula in the epilepsy and the gambling disorder of Fyodor Dostoyevsky.

Background The retrospective diagnosis of Fyodor Mikhailovich Dostoyevsky's (1821-1881) neurological and psychiatric disease proves to be particularly interesting. Recent neurobiological data suggest a solution to the questions regarding the writer's retrospective diagnosis, claiming the insular cortex to be the origin of the rare ecstatic seizures. Regarding Dostoyevsky's pathological gambling, this hypothesis is consistent with another finding from recent neuroscience, namely that the malfunction of the insula could be an important underlying pathology in gambling disorder.
