Type X collagen is principal extracellular matrix (ECM) in natural dermis. To prepare artificial dermis, collagen is traditional, and most superior biomaterial. But beside collagen, the dermis also contains many other ECM. Among them, glycosaminoglycan (GAG) is another important substance. To imitate the natural dermis, and modificate the scaffold materials, two types of scaffolds were prepared: one is traditional type X collagen spongy scaffold, the other is collagen-chondroitin sulfate (CS) spongy scaffold. Collagen was blended with CS, one kind of GAG, and cross-linked by 1-ethyl-3-(3-dimet

Cyclopeptides have emerged as promising candidates for drug development. However, owing to the scarcity of efficient cyclization strategies for natural peptides, the development of diverse cyclopeptides remains a formidable challenge. In this study, a variety of macrocyclic peptides featuring (thio)urea bridges are developed through the site-selective lysine‑lysine crosslinking of native peptides, employing N, N'-carbonyldiimidazole or N, N'-thiocarbonyldiimidazole as bridging reagents. In particular, dual thiourea-bridged macrocyclic peptides are further synthesized by using a one-pot two-st

The activation of chemical reactions in living cells using ultraviolet (UV) light enables the interrogation of biomolecules in their native environment with photoreactive probes or crosslinking reagents. Although numerous photo-crosslinking approaches have been successfully employed, they often suffer from common limitations, including low reaction yields, the need for long exposure times, and irradiation-induced cellular damage from heat, desiccation, or side reactions. We recently showed that 365 nm light-emitting diodes enable rapid, biorthogonal protein-DNA crosslinking in living cells, in

Uremic toxins pose a significant threat to the health and longevity of kidney dialysis patients, and their broad range of molecular weights and chemical properties presents a challenge to adsorption-based removal strategies. Developing efficient adsorbent materials capable of selective toxin capture is crucial for improving the dialysis outcomes. In this work, macroporous polymer resins (MPRs) with different chemical functionality were synthesized via bulk polymerization to investigate how uremic toxin adsorption is impacted by different molecular structures in a cross-linked network structure

Polyion complex (PIC) vesicles are attractive carriers for mRNA delivery, yet their assembly is often compromised by charged cargos that disrupt the electrostatic interactions required for vesicle formation. Here, we report triblock polyampholyte vesicles (TBPVs) that encode preferential polymer-polymer interactions, enabling robust vesicle assembly independent of cargo-mediated disruption. Constructed from poly(ethylene glycol)-b-poly(L-lysine)-b-poly(aspartic acid) copolymers, TBPVs encapsulate both free mRNA and pre-condensed mRNA polyplexes while preserving vesicle integrity. Notably, the

Intrauterine adhesions (IUA) are primarily caused by damage to the basal layer of the endometrium during uterine surgery. Hysteroscopic adhesiolysis is the treatment of choice; however, recurrent adhesions following surgery often require management with physical barriers. These barriers, however, are insufficiently effective and may cause endometrial damage, thereby hindering tissue regeneration. In this study, a cross-linked recombinant type III collagen hydrogel (CRC) was developed using 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC) and N-hydroxysuccinimide (NHS) as cross-linking agent

Minoxidil is the only FDA approved topical treatment for androgenetic alopecia, which is male- or female-pattern hair loss. However, its use is limited by its efficacy and tolerability. Here, we describe a novel bioengineered approach to improve local delivery to the skin. We showed that Schiff bases form between the diamines of minoxidil and aldehyde-modified hyaluronic acid, leading to cross-linking and formation of injectable hydrogels. Hydrogels exhibited tunable release of minoxidil over 6 weeks in vitro with induction of hair growth in vivo in a mouse model after intradermal injection. T

Current methods for sequence-selective biomolecule isolation suffer from high cost, off-target effects, and limited flexibility. Here, we introduce LASSO (crossLink-Assisted Sequence-Selective isOlation), a versatile platform using programmable polymer phase separation to capture biomolecules under native conditions. LASSO relies on combinatorial crosslinker libraries-diverse mixtures of DNA strands that collectively trigger the formation of highly swollen polymer agglomerates with near-zero background binding. We demonstrate >80% pulldown efficiency for diverse targets, including DNA, SARS-Co

In addressing the challenges of recovery and microparticle residue associated with biochar, loading biochar with starch presents a viable solution. However, starch's high hydrophilicity and viscosity introduce water resistance and uniform loading issues. This study proposes an innovative ionically synergistic dual crosslinking modification method, utilizing an extruder barrel as a novel reactor. This method enables the construction of highly water-resistant starch gels and the uniform and robust loading of biochar micro-nanoparticles within the gel matrix, achieving the "one-step" preparation

Hydrogel has been widely studied as a carrier model system for drug delivery. Efficient encapsulation of drug molecules and their controlled release are important factors in the design of hydrogels to control drug release at a desired time and location. In this study, we propose a photoresponsive hydrogel with tunable mechanical properties for drug delivery. The hydrogel was synthesized by cross-linking maleimide-functionalized 4-armed polyethylene glycol (4-armed PEG-Mal) with reversibly photodissociable green fluorescent protein pdDronpa. Chemical modification of naturally derived glycosaminoglycans (GAGs) expands their potential utility for applications in soft tissue repair and regenerative medicine. Here we report the preparation of a novel crosslinked chondroitin sulfate (~200 to 2000 kilodaltons) that is both soluble in aqueous solution and microfilterable. We refer to these materials as "SuperGAGs." One can further conjugate these materials with diverse capture agents to further modify polymer properties and add new capabilities. A representative material (GLX-100) demonstrated durable restoration of bladder impermeabilit

Alcohol is broken down in the body into acetaldehyde, a toxic chemical that can damage DNA by creating interstrand crosslinks (AA-ICL). These crosslinks block DNA replication and threaten the stability of the genome. A rare genetic disease, Fanconi anaemia (FA), is marked by extreme sensitivity to DNA crosslinking agents, including acetaldehyde. Although the Fanconi anaemia DNA repair pathway is known to fix this type of damage, exactly how it repairs acetaldehyde crosslinks is not yet understood. Here we show that the FA nuclease Slx4-Xpf-Ercc1 (SXE) plays a key role in the repair of AA-ICL.

Moldable tissue-sealant hydrogels were developed herein by combining the yield stress fluidity of a Carbomer and in situ cross-linking of 3-arm PEG-thiol (PEG-SH) and 4-arm PEG-acrylate (PEG-AC). The Carbomer was mixed with each PEG oligomer to form two aqueous precursors: Carbomer/PEG-SH and Carbomer/PEG-AC. The two hydrogel precursors exhibited sufficient yield stress (>100 Pa) to prevent dripping from their placement on the tissue surface. Moreover, these hydrogel precursors exhibited rapid restructuring when the shear strain was repeatedly changed. These rheological properties contribute t

The present work consist of the synthesis of photo-cross-linkable materials, based on unsaturated polyesters (UPs), synthesized from biobased monomers from renewable sources such as itaconic acid and 1,4-butanediol. The UPs were characterized to assess the influence of polycondensation reaction temperature and cross-linking time on their final properties. For this purpose, different UV irradiation exposure periods were tested. Homogeneous, uniform, and transparent films were obtained after 1, 3, and 5 min of UV exposure. These cross-linked films were then characterized. All materials presented

Alzheimer's disease (AD) is a progressive neurological disorder featuring abnormal protein aggregation in the brain, including the pathological hallmarks of amyloid plaques and hyperphosphorylated tau. Despite extensive research efforts, understanding the molecular intricacies driving AD development remains a formidable challenge. This study focuses on identifying key protein conformational changes associated with the progression of AD. To achieve this, we employed quantitative cross-linking mass spectrometry (XL-MS) to elucidate conformational changes in the protein networks in cerebrospinal

Hydrogels appear to be an attractive class of biomaterial for cartilage tissue engineering due to their high water content, excellent biocompatibility, tunable stiffness, etc. The crosslinking density of the hydrogel can affect their viscoelastic property, and therefore potentially impact the chondrogenic phenotype of re-differentiated chondrocytes in a 3D microenvironment through physical cues. To understand the effect of crosslinking densities on chondrocytes phenotype and cellular interaction with the hydrogel, this study utilized a clinical grade thiolate hyaluronic acid and thiolate gelat

Previous studies have demonstrated that UV cross-linking (CXL) increases stromal stiffness and produces alterations in extracellular matrix (ECM) microstructure. In order to investigate how CXL impacts both keratocyte differentiation and patterning within the stroma, and fibroblast migration and myofibroblast differentiation on top of the stroma, we combined CXL with superficial phototherapeutic keratectomy (PTK) in a rabbit model. Twenty-six rabbits underwent a 6 mm diameter, 70 μm deep phototherapeutic keratectomy (PTK) with an excimer laser to remove the epithelium and anterior basement mem

Anti-neoplastic effect of DNA cross-linking agents such as cisplatin, mitomycin C, and psoralen is attributed to their ability to induce DNA interstrand cross-links (ICLs), which block replication, transcription, and linear repair pathways by preventing DNA strand separation and trigger apoptosis. It is generally agreed that the Fanconi anemia (FA) pathway orchestrates the removal of ICLs by the combined actions of various DNA repair pathways. Recently, attention has been focused on the ability of the NEIL3-initiated base excision repair pathway to resolve psoralen- and abasic site-induced ICL
