OBJECTIVE: To compare pregnancy rates after fallopian tubal and uterine transfer of cryopreserved embryos.
DESIGN: Prospective randomized trial with assignment to treatment groups by a random number table.
SETTING: University of Iowa Hospitals and Clinics, a tertiary care academic institution.
PATIENTS: Forty patients with patent fallopian tubes and at least three cryopreserved embryos.
INTERVENTIONS: Cryopreserved embryos were thawed and transferred to the fallopian tube by laparoscopy or to the uterus by a transcervical catheter.
MAIN OUTCOME MEASURES: Clinical and ongoing pregnancy rat

Live frozen vaccines containing Babesia bovis, Babesia bigemina or Anaplasma centrale were prepared using glycerol as cryoprotectant and stored in liquid nitrogen. The viability of the vaccines was tested inoculating calves 1 h (n = 12), 2 h (n = 12), 12 h (n = 6) and 24 h (n = 6) after thawing. Since September, 1985, 20 patients have undergone implantation of a homograft valve in the pulmonary position (16 pulmonary, 4 aortic). There were 11 primary operations and 9 reoperations. In 7 of 11 primary operations the homograft valve was utilized as a composite conduit with a short Dacron extension. In four of five reoperations for a failed porcine valved conduit, a composite homograft conduit was used. Here, we present an optimized workflow for PBMC isolation from human whole blood, followed by short-term cryopreservation at -80°C. The protocol includes rapid thawing to preserve cell viability, washing steps to minimize platelet contamination, and quality-control assessment of PBMC number, viability, and composition before HRR measurements.
