Vocal fold bowing in elderly male monozygotic twins: a case study.

This case study examined case histories, diagnostic features, and treatment response in two 79-year-old male monozygotic (identical) twins with vocal fold bowing, exploring both genetic and environmental factors. DNA concordance was examined via cheek swab. Case histories, videostroboscopy, auditory- and visual-perceptual assessment, electromyography, acoustic measures, and Voice Handicap ratings were undertaken. Both twins underwent surgical intervention and subsequent voice therapy. Monozygosity was confirmed for DNA polymorphisms, with 10 of 10 concordance for STR DNA markers. For both twin

DNA Methylation Variation Is Identified in Monozygotic Twins Discordant for Non-syndromic Cleft Lip and Palate.

Non-syndromic cleft lip with or without cleft palate (NSCLP) is the most common craniofacial birth defect. The etiology of NSCLP is complex with multiple genes and environmental factors playing causal roles. Although studies have identified numerous genetic markers associated with NSCLP, the role of epigenetic variation remains relatively unexplored. Because of their identical DNA sequences, monozygotic (MZ) twins discordant for NSCLP are an ideal model for examining the potential contribution of DNA methylation to non-syndromic orofacial clefting. In this study, we compared the patterns of wh

Exposure to environmental tobacco smoke during pregnancy in rats yields less effect on indices of brain cell number and size than does postnatal exposure.

While there is evidence that human perinatal exposure to environmental tobacco smoke (ETS) can result in an increased risk of respiratory disorders and sudden infant death syndrome, evidence linking ETS exposure to neurodevelopmental handicaps is suggestive but less compelling. We previously noted that postnatal ETS exposure, rather than prenatal exposure, resulted in reduced concentration of hindbrain DNA and increased protein/DNA ratio when rat brain tissue was studied at 9 weeks postnatal age. We have now evaluated the effects of ETS exposure during pregnancy on brain development by assayin

Environmental Exposure-Induced Epigenetic Modifications and Adverse Pregnancy Outcomes: A Systematic Review.

Prenatal environmental exposures may alter epigenetic regulation, influencing fetal development and pregnancy outcomes. The aim of the study was to systematically review original studies examining associations between environmental exposures, epigenetic modifications, and pregnancy-related disorders. A systematic search of PubMed, ScienceDirect, Cochrane Library, and Google Scholar identified studies evaluating prenatal exposures, epigenetic markers, and clinical outcomes. Study quality was assessed using the Joanna Briggs Institute criteria. Thirty-four studies (2009-2024) were included. Pren

Epigenetic modification of hypothalamic neuropeptides and metabolic hormone receptors in metabolic health.

The hypothalamus plays a central role in regulating metabolism by integrating neuropeptide signaling with environmental cues to maintain energy homeostasis. Adverse environmental factors, such as obesogenic diet, undernutrition, stress, and sedentary lifestyles, can disrupt the normal regulation of key hypothalamic neuropeptides and metabolic hormone receptors through epigenetic mechanisms, including DNA methylation, histone modifications, and microRNA regulation. These epigenetic alterations are not merely transient; they can be heritable and may influence metabolic health across generations,

The Role of Environmental Toxicant-Induced Oxidative Stress in Male Infertility.

Infertility is a serious public health issue affecting around 15% of couples globally. Of the 60-80 million people of reproductive age affected by infertility, 40-50% are due to male factor while 30-40% of cases are still idiopathic. The recent global deterioration in sperm quality raises apprehensions regarding the toxic effects of environmental pollutants on reproductive health of males. Environmental toxicants have shown strong evidences for inducing oxidative stress affecting spermatogenesis severely, thereby leading to reduced sperm motility, count, and DNA damage. Reactive oxygen species

In vitro investigation and mass spectrometric characterization of novel DNA adducts of sesquimustard.

Sesquimustard (Q), a potent bifunctional alkylating vesicant, is a significant toxicological concern due to its environmental persistence and capability to induce severe genotoxic effects. It can create several metabolites, some of which can be utilized as biomarkers, and it can extensively alkylate key macromolecules in organisms, including DNA. Although prior research has primarily focused on glutathione and albumin adducts as retrospective biomarkers of Q exposure, no prior studies have identified DNA adducts of Q. This study aimed to fill this critical gap by characterizing, for the first

LncRNA DINOR is a virulence factor and global regulator of stress responses in Candida auris.

The emergent fungal pathogen Candida auris exhibits high resistance to antifungal drugs and environmental stresses, impeding treatment and decontamination1-3. The fungal factors mediating this stress tolerance are largely unknown. In the present study, we performed piggyBac, transposon-mediated, genome-wide mutagenesis and genetic screening in C. auris, and identified a mutant that grew constitutively in the filamentous form. Mapping the transposon insertion site revealed the disruption of a long non-coding RNA, named DINOR for DNA damage-inducible non-coding RNA. Deletion of DINOR caused DNA

Exposome and unhealthy aging: environmental drivers from air pollution to occupational exposures.

The aging population worldwide is facing a significant increase in age-related non-communicable diseases, including cardiovascular and brain pathologies. This comprehensive review paper delves into the impact of the exposome, which encompasses the totality of environmental exposures, on unhealthy aging. It explores how environmental factors contribute to the acceleration of aging processes, increase biological age, and facilitate the development and progression of a wide range of age-associated diseases. The impact of environmental factors on cognitive health and the development of chronic age

Blood mtDNA markers of mitochondrial subtype and early-onset Parkinson's disease biology.

Mitochondrial dysfunction is central to the pathogenesis of Parkinson's disease (PD), integrating both genetic and environmental factors. Therefore, reliable blood-based biomarkers reflecting mitochondrial alterations are needed. Emerging evidence suggests that somatic changes to mitochondrial DNA (mtDNA) may reflect early disease-associated processes relevant to PD conversion and clinical manifestation. In this study, we analysed somatic mtDNA major arc deletions as a measure of mitochondrial genome integrity and evaluated 7S DNA abundance as well as copy number as complementary readouts in w

Oxidative stress in allergic respiratory diseases.

There is ample evidence that allergic disorders, such as asthma, rhinitis, and atopic dermatitis, are mediated by oxidative stress. Excessive exposure to reactive oxygen and nitrogen species is the hallmark of oxidative stress and leads to damage of proteins, lipids, and DNA. Oxidative stress occurs not only as a result of inflammation but also from environmental exposure to air pollution and cigarette smoke. The specific localization of antioxidant enzymes in the lung and the rapid reaction of nitric oxide with reactive oxygen species, such as superoxide, suggest that antioxidant enzymes migh

Synergistic interplay between UV and urban particulate matter exposure induces melanocyte senescence and contributes to human skin aging.

Extrinsic skin aging is driven by environmental factors, including ultraviolet (UV) radiation and air pollution. While melanocytes serve as key protectors against UV-induced damage, their role in aging, particularly through the process of senescence, remains underexplored. Here, we exposed human neonatal melanocytes and ex vivo skin explants to UV (UVA + UVB), urban particulate matter (UPM), and their combination (UV + UPM) to assess the effects on melanocyte function and skin aging. We demonstrate that combined UV + UPM exposure triggers oxidative stress, mitochondrial and DNA damage, senesce

Variably methylated retrotransposons are refractory to a range of environmental perturbations.

The agouti viable yellow (Avy) allele is an insertional mutation in the mouse genome caused by a variably methylated intracisternal A particle (VM-IAP) retrotransposon. Avy expressivity is sensitive to a range of early-life chemical exposures and nutritional interventions, suggesting that environmental perturbations can have long-lasting effects on the methylome. However, the extent to which VM-IAP elements are environmentally labile with phenotypic implications is unknown. Using a recently identified repertoire of VM-IAPs, we assessed the epigenetic effects of different environmental contexts

Influence of environmental factors in the development of inflammatory bowel diseases.

Idiopathic inflammatory bowel diseases (IBD), Crohn's disease (CD) and ulcerative colitis (UC), are multifactorial diseases that are manifested after disruption of a genetic predisposed individual and its intestinal microflora through an environmental stimulus. Urbanization and industrialization are associated with IBD. Epidemiological data, clinical observations and family/immigrants studies indicate the significance of environmental influence in the development of IBD. Some environmental factors have a different effect on the subtypes of IBD. Smoking and appendectomy is negatively associated

Heavy Metals and Cancer: Unraveling the Environmental Sources and Molecular Carcinogenic Mechanisms.

Heavy metals are the emerging environmental pollutants. They may accumulate in soil, water, and air and enter the food chain. Humans are exposed to these agents through oral, dermal and nasal routes. After entering the human body, heavy metals may reach various organs and cause health issues. Long-term exposure increases the risk of cancers associated with the lungs, breast, skin, GI tract, and prostate. Toxic heavy metals trigger oxidative stress by increasing free radical production and disrupting redox homeostasis. Free radicals damage DNA bases, and if not repaired by the DNA repair mechan

LINE-1 DNA methylation mediates smoking-related risk in site-specific urothelial carcinoma: a Taiwan case-control study.

While environmental and lifestyle factors contribute to urothelial carcinoma (UC) development, their differential effects on bladder cancer (UB) versus upper tract urothelial carcinoma (UTUC) and the underlying epigenetic mechanisms remain unclear. This study investigated location-specific risk factors and the mediating role of LINE-1 DNA methylation in UC carcinogenesis. A hospital-based case-control study was conducted, comprising 478 UC cases (266 UB, 204 UTUC) and 569 controls. The risk factors, including smoking, Chinese herbal medicine use, comorbidities, and blood levels of arsenic, cad
