Phylogeny and morphology of some European cyathocotylid digeneans (Trematoda: Diplostomoidea).

The Cyathocotylidae Mühling, 1898 is a family of primitive diplostomoid trematodes important for understanding the evolution of the superfamily Diplostomoidea. However, cyathocotylids remain poorly studied with the use of molecular techniques. In this study we sequenced the 5.8S + ITS2 region, 28S rRNA, and cox1 genes of two cyathocotylid species and obtained new morphological data on them. We propose Georduboisia nom. nov. instead of the preoccupied name Duboisia Szidat, 1936 (junior homonym of Duboisia Stremme, 1911). Adults of Georduboisia cf. teganuma (Ishii, 1935) and Paracoenogonimus ova

Morphological and molecular characterization of Seuratascaris numidica (Seurat, 1917) (Ascaridida: Ascarididae).

Seuratascaris numidica (Seurat, 1917) is a specialized nematode species parasitizing amphibians only. In the present study, the detailed morphology of this poorly known species was studied using light and scanning electron microscopy based on the newly material collected from Hoplobatrachus chinensis (Osbeck) (Amphibia: Anura) in China. We found that the relative length of intestinal caecum in our male specimens (representing 68.4-71.1% of oesophageal length) is slighter longer than the previously reported data (not over 60.0% of oesophageal length). Our SEM observations also revealed the pres

The taxonomic and phylogenetic status of digeneans from the genus Timoniella (Digenea: Cryptogonimidae) in the Black and Baltic seas.

Timoniella spp. are cryptogonimid flukes (Digenea: Cryptogonimidae) that parasitize the guts of fish in brackish waters. Timoniella imbutiforme, a species from the Mediterranean Sea, is recorded in the Black Sea, while T. balthica has been described from the Baltic Sea. In this paper, we clarify the taxonomic status of Timoniella populations in the Baltic and Black seas. Adults and metacercariae of Timoniella spp. were sampled from localities in the Mediterranean Sea (France), Black Sea (Ukraine) and Baltic Sea (Germany) and subjected to molecular and morphological analysis, including Bayesian

Invalidation of Diphyllobothrium hottai (Cestoda: Diphyllobothriidae) based on morphological and molecular phylogenetic analyses.

Diphyllobothrium hottai Yazaki, Fukumoto & Abe, 1988 was described based on the morphology of adult worms recovered from golden hamsters that had been experimentally infected with plerocercoids obtained from Japanese surf smelts (Hypomesus pretiosus japonicus) and olive rainbow smelts (Osmerus eperlanus mordax). Although D. hottai was considered to be distinct from Diphyllobothrium ditremum (Creplin, 1825), their taxonomic relationship requires further clarification. In our study, D. hottai and D. ditremum obtained from hamsters experimentally infected with plerocercoids isolated from Japanese

Molecular characterization of Opisthorchis noverca (Digenea: Opisthorchiidae) based on nuclear ribosomal ITS2 and mitochondrial COI genes.

Opisthorchiasis is a public health problem in South-East Asian countries and Eastern Europe. The infection implicates mainly two species of Opisthorchis, namely O. viverrini and O. felineus, that occur mostly in fish-eating mammals and humans, although there are rare reports of human cases involving two other species, O. noverca and O. guayaquilensis. Opisthorchis noverca has been reported frequently in dogs and pigs from the Indian subcontinent, with rare reports from cattle and human subjects. With a view to supplementing morphology-based identification of this species, the present study aim

Molecular and morphological differentiation of two similar species of Accacoeliidae (Digenea): Accacladocoelium macrocotyle and A. nigroflavum from sunfish, Mola mola.

In a study of 106 sunfish, Mola mola (L.), from Mediterranean waters, 2,731 worms, belonging to 2 congeneric species of accacoeliids, Accacladocoelium macrocotyle (Diesing, 1858) Robinson, 1934 and Accacladocoelium nigroflavum (Rudolphi, 1819) Robinson, 1934 , were collected from the digestive system. It is often difficult to differentiate between these 2 species as they are sympatric and very similar; in fact, according to previous descriptions, the extent of the vitellarium is the only interspecific difference, described as extending posteriorly to the ovary in A. macrocotyle and as anterior

Fasciola hepatica in snails collected from water-dropwort fields using PCR.

Fasciola hepatica is a trematode that causes zoonosis mainly in cattle and sheep and occasionally in humans. Fascioliasis has been reported in Korea; however, determining F. hepatica infection in snails has not been done recently. Thus, using PCR, we evaluated the prevalence of F. hepatica infection in snails at 4 large water-dropwort fields. Among 349 examined snails, F. hepatica-specific internal transcribed space 1 (ITS-1) and/or ITS-2 markers were detected in 12 snails and confirmed using sequence analysis. Morphologically, 213 of 349 collected snails were dextral shelled, which is the sam

DNA types of aspermic Fasciola species in Japan.

In order to reveal DNA types of aspermic Fasciola forms in Japan, Fasciola specimens obtained from eight prefectures that had not been previously reported were analyzed for DNA of ribosomal internal transcribed spacer 1 (ITS1) and mitochondrial NADH dehydrogenase 1 (ND1) gene. Five combinations in DNA types of both ITS1 and ND1 were revealed from the results of this study and previous studies. The DNA type Fsp2, which is identical to that of F. gigantica in both ITS1 and ND1, was the most predominant in Japan, followed by Fsp1, which is the same DNA type as that of F. hepatica. Fasciola forms

Description and phylogenetic position of a new Serpentirhabdias (Nematoda: Rhabdiasidae) from Hydrodynastes gigas (Serpentes: Colubroidea) in Pantanal, Brazil.

Serpentirhabdias Tkach, Kuzmin and Snyder, 2014 comprises about 23 species of lung-dwelling nematodes parasites of snakes distributed worldwide. Among them, eight species were found in Brazil. We describe and examine the phylogenetic position of Serpentirhabdias kuzmini n. sp., a parasite of Hydrodynastes gigas from the Pantanal, Brazil. The new species is characterized by a triangular oral opening, absence of a buccal capsule, presence of onchia in the oesophastome, excretory glands longer than the esophagus, and a gradually tapering tail. Moreover, comparative analysis of partial sequences o

Assessing the taxonomic status of an opecoelid trematode from the evileye blaasop, Amblyrhynchote honckenii (Bloch), in South Africa.

The taxonomic status of Pseudoheterolebes cotylophorus (Ozaki 1935) Yamaguti 1959 (Opecoelidae: subfamily Opistholebetinae) has been a subject of taxonomic confusion since its initial description by Ozaki in 1935 as Opistholebes cotylophorus Ozaki 1935 from diodontid fish in Japan. The species' current recognised classification dates back to Yamaguti's reclassification of O. cotylophorus as Pseudoheterolebes cotylophorus; however, doubts persist regarding this classification and the validity of this species. The species was reported by Bray in 1986 from off the coast of South Africa, based on

The phylogenetic position of the unusual diplostomoidean genus Schwartzitrema Vigueras, 1941 (Digenea, Diplostomoidea) and description of 2 new species from Neotropics.

Schwartzitrema Vigueras, 1941, is a small, rarely reported genus of diplostomoideans that parasitize avian definitive hosts, predominantly anhingas, on several continents. No DNA sequences were previously available for any Schwartzitrema spp. In the present study, we describe adults of 2 new Schwartzitrema spp. from anhingas in Brazil. We also provide description of metacercaria of one of the new species from Argentina. Schwartzitrema macrotesticulata n. sp. lacks a clear separation between its testes, appearing almost as a contiguous, irregularly shaped, lobular mass. A highly characteristic

The first determination of Trichuris sp. from roe deer by amplification and sequenation of the ITS1-5.8S-ITS2 segment of ribosomal DNA.

Trichuris nematodes were isolated from roe deer (Capreolus capreolus). At first, nematodes were determined using morphological and biometrical methods. Subsequently genomic DNA was isolated and the ITS1-5.8S-ITS2 segment from ribosomal DNA (RNA) was amplified and sequenced using PCR techniques. With u sing morphological and biometrical methods, female nematodes were identified as Trichuris globulosa, and the only male was identified as Trichuris ovis. The females were classified into four morphotypes. However, analysis of the internal transcribed spacers (ITS1-5.8S-ITS2) of specimens did not c

Genetic identification of Fasciola hepatica by ITS-2 sequence of nuclear ribosomal DNA in Turkey.

The trematodes of the genus Fasciola are the common liver flukes of a range species of animals and have a global geographical distribution. They can generally be distinguished on the basic of their morphology. ITS-2 ribosomal DNA sequences have been used to characterize the liver flukes as a specific marker from different geographical regions which include F. hepatica, F. gigantica, and an intermediate Fasciola. To determine the phylogenetic location of F. hepatica of Turkey origin based on ITS-2 rDNA molecular data, adult F. hepatica trematodes were collected from the liver naturally infected

Molecular identification of a Trichinella isolate from a naturally infected pig in Tibet, China.

The first human case with trichinellosis was reported in 1964 in Tibet, China. However, up to the present, the etiological agent of trichinellosis has been unclear. The aim of this study was to identify a Tibet Trichinella isolate at a species level by PCR-based methods. Multiplex PCR revealed amplicon of the expected size (173 bp) for Trichinella spiralis in assays containing larval DNA from Tibet Trichinella isolate from a naturally infected pig. The Tibet Trichinella isolate was also identified by PCR amplification of the 5S ribosomal DNA intergenic spacer region (5S ISR) and mitochondrial

Natural hybridization between Paragonimus bangkokensis and Paragonimus harinasutai.

Paragonimus bangkokensis and Paragonimus harinasutai, which are morphologically distinguishable species, often co-infect in the same crab intermediate hosts. Recent molecular phylogenetic analyses revealed that these two species are genetically close to each other and are considered as the sister species. While we have been studying Paragonimus adult worms obtained from the lungs of a cat experimentally infected with Paragonimus metacercariae which were morphologically identified as P. harinasutai collected from central Viet Nam, one out of 6 adult worms has grouped cuticular spines, which is

Molecular cloning, expression, and serological evaluation of an 8-kilodalton subunit of antigen B from Echinococcus multilocularis.

Full-length cDNA and genomic DNA encoding an 8-kDa subunit of antigen B from Echinococcus multilocularis (designated EmAgB8/1) were isolated from an E. multilocularis metacestode cDNA library and a protoscolex genomic DNA library, respectively. The open reading frame of the cDNA clone encodes a polypeptide comprising 85 amino acids with a 20-amino-acid NH(2)-terminal signal sequence, which was confirmed following N-terminal sequencing of the native antigen. Reverse transcription-PCR analysis revealed that the clone encoding EmAgB8/1 is predominantly transcribed in larval E. multilocularis. The

Cloning and expression of DiT33 from Dirofilaria immitis: a specific and early marker of heartworm infection.

Dirofilaria immitis is an important filarial parasite of dogs and cats, and a useful model for human filariasis. Current diagnostic tests for heartworm infection in animals rely on the presence of fecund female worms (usually found 6.5 months post-infection or later) and therefore fail to detect pre-patent infections. Putative pepsin inhibitors from 2 filarial parasites of humans namely Onchocerca volvulus (Ov33, Oc3.6, OvD5B) and Brugia malayi (Bm33), have been shown to be useful in diagnosis of onchocerciasis and lymphatic filariasis, respectively. Previous studies have suggested that a homo

Immunization with plasmid DNA encoding the integral membrane protein, Sm23, elicits a protective immune response against schistosome infection in mice.

Schistosomes are helminth parasites infecting at least 200 million people worldwide. In this study, we evaluated the feasibility of using a nucleic acid vaccine to induce protective immune responses to the Schistosoma mansoni integral membrane protein Sm23. C57BL/6 mice were immunized by intramuscular injection in three separate vaccination trials. ELISA and Western Blot analyses indicated that mice immunized with a DNA plasmid construct encoding Sm23 (Sm23-pcDNA) generated specific IgG for Sm23, while sera from mice immunized with the control pcDNA plasmid did not. The vaccine elicited IgG(2a

Molecular cloning and characterization of a nematode polyprotein antigen/allergen from the human and animal hookworm Ancylostoma ceylanicum.

Nematodes are unable to synthesize fatty acids de novo and must acquire them from the environment or host. It is hypothesized that two unique classes of fatty acid and retinol binding proteins that nematodes produce (fatty acid and retinol binding (FAR) and nematode polyprotein antigen/allergen (NPA)) are used to meet this need. A partial cDNA has been cloned corresponding to four subunits of a putative Ancylostoma ceylanicum NPA (AceNPA). The translated amino acid sequence of AceNPA shares sequence identity with similar proteins from Dictyocaulus viviparus, Ascaris suum, and Ostertagia ostert
