Molecular cloning and characterization of duck CD83 and its effect on H9N2 avian influenza virus replication.

Cluster of Differentiation 83 (CD83) is a multifunctional regulatory molecule that has been identified in several mammals, birds, and fishes; however, no reports have described CD83 in ducks to date. Given the important role ducks play in the transmission of avian influenza viruses (AIVs), this study aimed to characterize the molecular features of duck CD83 (duCD83) and investigate its potential role during H9N2 AIV infection. We successfully cloned the coding sequence (CDS) and extracellular domain of duCD83 for the first time and expressed the recombinant protein in human embryonic kidney 29…

Duck plague virus gG is secreted, nonstructural glycoprotein, not essential for viral replication and responsible for the virulence.

Duck plague virus (DPV) is a highly pathogenic member of the herpesvirus family that can induce significant morbidity and mortality, primari … cycle, suggesting that gG is not essential for viral replication. In vivo studies demonstrated that ducks infected with the gG-knockout virus presented minimal organ lesions and a significantly lower mortality rate than did those infected with the wild-type virus. Furthermore, gG modulated the transcription levels of host interferons, proinflammatory cy …

Identification and subcellular localization of proteins that interact with Duck plague virus pUL14 in infected host cells.

Duck plague (DP), which is caused by duck plague virus (DPV), is an infectious disease that severely harms the waterfowl breeding industry. The UL14 protein (pUL14) is a tegument protein encoded by the UL14 gene, which is located in the unique long (UL) region of the DPV genome. DPV pUL14 plays a crucial role in viral replication, likely by interacting with host and viral proteins that have yet to be identified. In this study, glutathione-S-transferase (GST) pull-down combined with liquid chromatography-mass spectrometry (LC-MS/MS) was employed to identify pUL14-interacting proteins in DPV-inf

Bacillus subtilis expressing duck Tembusu virus E protein induces immune protection in ducklings.

Duck Tembusu virus (DTMUV) is an infectious disease that emerged in China in 2010. It has caused serious economic losses to the poultry industry and may pose a threat to public health. We aimed to develop a new Bacillus subtilis (B. subtilis)-based oral vaccine to control DTMUV transmission among poultry; to this end, we constructed a B. subtilis strain that can secrete DTMUV E protein. Ducklings were orally immunized, and serum antibodies, mucosal antibodies, and splenic cytokines were detected. The results showed that, in addition to high levels of specific IgG, there were also high levels o

The hydrodynamic performance of duck feet for submerged swimming resembles oars rather than delta-wings.

Waterfowl use webbed feet to swim underwater. It has been suggested that the triangular shape of the webbed foot functions as a lift-generating delta wing rather than a drag-generating oar. To test this idea, we studied the hydrodynamic characteristics of a diving duck's (Aythya nyroca) foot. The foot's time varying angles-of-attack (AoAs) during paddling were extracted from movies of ducks diving vertically in a water tank. Lift and drag coefficients of 3D-printed duck-foot models were measured as a function of AoA in a wind-tunnel; and the near-wake flow dynamics behind the foot model was ch…

Duck body condition affects the number of parasites in the gastrointestinal tract, as evaluated with the Akaike information criterion.

The aim of the study was to determine the relationship between the body condition, and the number of parasites in the gastrointestinal tract of the tufted duck (Aythya fuligula), using the Akaike information criterion. Absolute and relative measurements of 197 ducks were taken. Liver mass was positively associated with the number of parasites in the duodenum (β = 0.5). Heart mass affected positively the number of parasites in the rectum (β = 2.3), the number of nematodes in the jejunum (β = 7.45), the total number of trematodes (β = 1.7), their number in the ceca (β = 7.3) and rectum (β = 4.2)…

Duck plague virus US3 protein kinase phosphorylates UL47 and regulates the subcellular localization of UL47.

Duck plague virus (DPV) belongs to the alphaherpesvirinae and causes high morbidity and mortality in waterfowl. UL47 is a large abundant structural protein in DPV, which means that UL47 protein plays an important role in virus replication. US3 protein, as a viral protein kinase in alphaherpesviruses, has been reported to be critical for DPV virion assembly. In this study, we over-expressed UL47 and US3 proteins and found that DPV UL47 protein was a phosphorylated substrate of US3 protein, which interacted and co-localized with US3 protein in the cytoplasm. US3-regulated phosphorylation of UL47

Genetic characterization and phylogenetic analysis of duck-derived waterfowl parvovirus in Anhui province, eastern China.

Recently, a novel duck-origin goose parvovirus (N-GPV) was reported to cause short beak and dwarfism syndrome in ducks. In this study, we performed complete genome sequencing and analyzed three different duck-derived parvoviruses that infected different breeds of ducks. Phylogenetic trees based on gene sequences indicated that they were classical goose parvovirus (C-GPV), Muscovy duck parvovirus (MDPV), and N-GPV. Furthermore, potential recombination events were found. These results improve our understanding of the diversity of duck-derived parvoviruses in Anhui province, eastern China, and pr…

Impacts of Duck-Origin Parvovirus Infection on Cherry Valley Ducklings From the Perspective of Gut Microbiota.

Duck-origin goose parvovirus (D-GPV) is the causative agent of beak atrophy and dwarfism syndrome (BADS), characterized by growth retardation, skeletal dysplasia, and persistent diarrhea. However, the pathogenic mechanism of D-GPV remains undefined. Here, we first reported the gut microbiome diversity of D-GPV infected Cherry Valley ducks. In the investigation for the influence of D-GPV infection on gut microbiota through a period of infection, we found that D-GPV infection caused gut microbiota dysbiosis by reducing the prevalence of the dominant genera and decreasing microbial diversity. Fur…

LGP2 plays a critical role in MDA5-mediated antiviral activity against duck enteritis virus.

Duck viral enteritis (DEV) is a DNA virus that leads to heavy economic losses in the commercial duck industry. As a key cytoplasmic sensor, melanoma differentiation-associated gene 5 (MDA5) can recognize viral RNA and enhance the antiviral immune response. Retinoic acid-inducible gene-I (RIG-I) and MDA5 both belong to the RIG-I-like receptors family, and RIG-I is known to be involved in the anti-DEV signaling pathway. However, the role of MDA5 in DEV infection remains unclear. In this study, we used overexpression and knockdown methods to determine if MDA5 affected DEV infection in ducks. We c…

Copaiba oil-resin (Copaifera reticulata Ducke) modulates the inflammation in a model of injury to rats' tongues.

BACKGROUND: The regeneration of integrity and tissue homeostasis after injury is a fundamental property and involves complex biological processes fully dynamic and interconnected. Although there are medications prescribed to accelerate the process of wound healing by reducing the exaggerated inflammatory response, comes the need to search for different compounds of Amazonian biodiversity that can contribute to the acceleration of the healing process. Among these products, the copaiba oil-resin is one of the most prominent feature in this scenario, as they have been reported its medicinal prope
