Ellagic acid C and aspergone R, two undescribed pyrones from the endophytic fungus Diaporthe sp. CB10100: biological evaluation, molecular docking and molecular dynamics studies.

Two new compounds, ellagic acid C (5) and aspergone R (6), along with four previously identified compounds (7-10), were successfully isolated from the fungus Diaporthe sp. CB10100. Ellagic acid C (5) contains an uncommon tricyclic 5/6/6 ring system. A biosynthetic pathway for 6 was proposed, although additional investigations are needed to elucidate the ethyl biosynthesis mechanism. Both aspergone R (6) and (-)-terpestacin (9) decreased lipopolysaccharide (LPS)-induced protein expression of iNOS and COX-2. Molecular docking analysis revealed that 6 and 9 potentially interact with the active si

1H-Pyrrole-2,5-dicarboxylic acid, a quorum sensing inhibitor from one endophytic fungus in Areca catechu L., acts as antibiotic accelerant against Pseudomonas aeruginosa.

Pseudomonas aeruginosa has already been stipulated as a "critical" pathogen, emphasizing the urgent need for researching and developing novel antibacterial agents due to multidrug resistance. Bacterial biofilm formation facilitates cystic fibrosis development and restricts the antibacterial potential of many current antibiotics. The capacity of P. aeruginosa to form biofilms and resist antibiotics is closely correlated with quorum sensing (QS). Bacterial QS is being contemplated as a promising target for developing novel antibacterial agents. QS inhibitors are a promising strategy for treating

Anticancer effect of altersolanol A, a metabolite produced by the endophytic fungus Stemphylium globuliferum, mediated by its pro-apoptotic and anti-invasive potential via the inhibition of NF-κB activity.

Altersolanol A, a natural product from the endophytic fungus Stemphylium globuliferum isolated from the medicinal plant Mentha pulegium (Lamiaceae) growing in Morocco, shows cytotoxic, cytostatic, anti-inflammatory and anti-migrative activity against human chronic myeloid K562 leukemia and A549 lung cancer cells in a dose dependent manner without affecting the viability of non cancerous cells. Altersolanol A induces cell death by apoptosis through the cleavage of caspase-3 and -9 and through the decrease of anti-apoptotic protein expression. Moreover, we report here the importance of the disti

Scavenging organic nitrogen and remodelling lipid metabolism are key survival strategies adopted by the endophytic fungi, Serendipita vermifera and Serendipita bescii to alleviate nitrogen and phosphorous starvation in vitro.

Serendipitaceae represents a diverse fungal group in the Basidiomycota that includes endophytes and lineages that repeatedly evolved ericoid, orchid and ectomycorrhizal lifestyle. Plants rely upon both nitrogen and phosphorous, for essential growth processes, and are often provided by mycorrhizal fungi. In this study, we investigated the cellular proteome of Serendipita vermifera MAFF305830 and closely related Serendipita vermifera subsp. bescii NFPB0129 grown in vitro under (N) ammonium and (P) phosphate starvation conditions. Mycelial growth pattern was documented under these conditions to c…

Characterization of Culturable Plant Growth-promoting Bacteria Isolated from Amaranthus hypochondriacus L. Roots.

Amaranths plants grow under several abiotic stress conditions and produce high-quality edible seeds. Despite their high agronomic and nutritional potential, their endophytic bacteria have not yet been characterized. This study aimed to isolate Amaranthus hypochondriacus L. root endophytes and characterize their plant growth-promoting capacity. Twenty-six amaranth root putative diazotrophs were isolated and identified by 16S rRNA gene sequencing. Most of the isolated strains exhibited multiple plant growth-promoting traits, such as iron and phosphate solubilization and the capacity to form biof…

Soil and seed both influence bacterial diversity in the microbiome of the Cannabis sativa seedling endosphere.

INTRODUCTION: Phytobiomes have a significant impact on plant health. The microbiome of Cannabis sativa is particularly interesting both because of renewed interest in this crop and because it is commercially propagated in two different ways (i.e. clonally and by seed). Angiosperms obtain a founding population of seed-borne endophytes from their seed-bearing parent. This study examines the influence of both seed and soil-derived bacteria on the endospheres of cannabis seedlings of both hemp- and drug-types. METHODS: A multi-factorial metagenomic study was conducted with three cannabis genotype…

Application of glyphosate or paraquat before the emergence of maize plants (Zea mays L.) had no detectable effect on the rhizosphere or endophyte bacterial community structure under the tested field conditions.

Herbicides are applied in agricultural practices to manage weeds. How these herbicides applied before maize emergence (Zea mays L.) might affect the rhizosphere and endophytic bacterial community is still largely unknown. In this study, plots were treated with 400 g paraquat ha-1 (Gramoxone®, paraquat ion 20% w/w), 1,089 g glyphosate ha-1 (Faena®, potassium salt of glyphosate 446 g/L) or weeds were hand removed before maize emerged and the bacterial community was determined in the bulk soil at the start of the experiment, in the bulk soil, rhizosphere, roots and stem at flowering (after 101 da

Root Exposure of Graphitic Carbon Nitride (g-C3N4) Modulates Metabolite Profile and Endophytic Bacterial Community to Alleviate Cadmium- and Arsenate-Induced Phytotoxicity to Rice (Oryza sativa L.).

To investigate the mechanisms by which g-C3N4 alleviates metal(loid)-induced phytotoxicity, rice seedlings were exposed to 100 and 250 mg/kg graphitic carbon nitride (g-C3N4) with or without coexposure to 10 mg/kg Cd and 50 mg/kg As for 30 days. Treatment with 250 mg/kg g-C3N4 significantly increased shoot and root fresh weight by 22.4-29.9%, reduced Cd and As accumulations in rice tissues by 20.6-26.6%, and elevated the content of essential nutrients (e.g., K, S, Mg, Cu, and Zn) compared to untreated controls. High-throughput sequencing showed that g-C3N4 treatment increased the proportion of

Development and validation of an ultrahigh performance liquid chromatography-high resolution tandem mass spectrometry assay for nine toxic alkaloids from endophyte-infected pasture grasses in horse serum.

Endophyte fungi (e.g. Epichloë ssp. and Neotyphodium ssp.) in symbiosis with pasture grasses (e.g. Festuca arundinacaea and Lolium perenne) can produce toxic alkaloids, which are suspected to be involved in equine diseases such as fescue toxicosis, ryegrass staggers, and equine fescue oedema. The aim of this study was, therefore, to develop and validate a quantification method for these and related alkaloids: ergocristine, ergocryptine, ergotamine, ergovaline, lolitrem B, lysergic acid, N-acetylloline, N-formylloline, peramine, and paxilline in horse serum. Differential partitioning of seed-inhabiting methylotrophs in the endosphere of wheat plants.

BACKGROUND: Both endophytes, microorganisms that reside within plants, and methylotrophs, which grow using methanol produced from plant leaves, play key roles in protecting plants against biotic and abiotic stresses. However, the source of endophytes and the mechanisms of their selection in plants are poorly understood. Therefore, experiments were carried out to identify wheat seed methylotrophic endophytes and evaluate their partitioning in root, stem and leaf of aseptic-controlled plants cultivated from surface-sterilized seeds. RESULTS: The counts of endophytic methanol utilizers were high…

Transcriptome Analysis of Choke Stroma and Asymptomatic Inflorescence Tissues Reveals Changes in Gene Expression in Both Epichloë festucae and Its Host Plant Festuca rubra subsp. rubra.

Many cool-season grasses have symbiotic relationships with Epichloë (Ascomycota, Clavicipitaceae) fungal endophytes that inhabit the intercellular spaces of the above-ground parts of the host plants. The presence of the Epichloë endophytes is generally beneficial to the hosts due to enhanced tolerance to biotic and abiotic stresses conferred by the endophytes. Many Epichloë spp. are asexual, and those infections always remain asymptomatic. However, some Epichloë spp. have a sexual stage and produce a macroscopic fruiting body, a stroma, that envelops the developing inflorescence causing a synd…

The Heterogeneous Effects of Epichloë and Rhizophagus irregularis on the Physiological and Rhizosphere Microbial Community of Festuca rubra.

In nature, a significant number of plant species form symbiotic associations with microorganisms, with arbuscular mycorrhizal fungi (AMF) and endophytic fungi being two prevalent groups of these partners. However, the ability to establish such symbioses with AMF and endophytic fungi is limited to a small fraction of native grass species. Nitrogen is a crucial nutrient for plant growth, yet it is often a limiting factor, underscoring the importance of understanding how plants acquire it. AMF enhance plant growth by improving nitrogen uptake efficiency, but the combined effects of endophytic fun
