Developmental aspects of the effect of naloxone on control of breathing in piglets.

The respiratory effects of a stereospecific opiate antagonist, naloxone, were studied in two groups of paralyzed and vagotomized piglets who … greater than that of the older piglets (P less than 0.05). These findings indicate that endogenous endorphins have a significant, though changing, role in control of breathing in the developing piglet.

Nociceptive reactivity and precipitated abstinence in hypophysectomized rats.

Nociceptive reactions were determined using the hot plate technique (55 degrees C) in sham-operated and hypophysectomized rats. No significant differences were noticed in the latencies to lick, jump, or escape between the two groups. These experiments indicate that pituitary endorphins are not the sole endogenous substances involved in the regulation of thermonociception. In hypophysectomized rats rendered acutely dependent on morphine, following naloxone, some signs of precipitated abstinence (chewing and teeth chattering) were significantly diminished, some others (abnormal posture, body tre…

[Opiate receptors and sleep. II. Effects of micro-injection of ethyl alcohol and pentobarbital in the median thalamus, periaqueductal gray matter and nucleus of the tractus solitarius of the rabbit (author's transl)].

There is an analogy between sleep EEGs produced by microinjections of morphine in the bulbo-mesencephalo-thalamic recruiting system and EEGs … bable that, via an as yet unknown mechanism, ethyl alcohol and pentobarbital promote the release of endorphins or peptides, which are specific ligands of all or some opiate receptors.

Opioids in Remote Ischemic Preconditioning-Induced Cardioprotection.

Remote ischemic preconditioning (RIPC) is an intriguing process whereby transient regional ischemia and reperfusion episodes to remote tissu … found their critical involvement in evoking cardioprotective effects. Endogenous opioids including endorphins, enkephalins, and dynorphins are released during RIPC and are critically involved in mediating RIPC-induced cardioprotective effects. It has been suggested that during RIPC, the endogenous opioids may be released into the systemic circulation and ma …

Neurochemistry of the Rett syndrome.

The current status of neurochemistry research with respect to the Rett syndrome (RS) is reviewed and correlations developed with the previou … nosis. Evidence of pervasive growth failure is presented and the current status of the role of beta-endorphins in RS is described. Considerations regarding future directions for research in RS are presented. These strategies include an approach to the fundamental question, namely, the identification of the primary molecular defect.

Varieties of Pathological Self-mutilation.

Pathological self-mutilation appears as a non-specific symptom as well as a specific syndrome. Since psychotic persons may commit horrifying acts, such as enucleation of an eye or amputation of a body part, identification of high risk patients is crucial. Stereotypical self-mutilation, such as head banging and biting off of fingertips, is associated with mental retardation and with the syndromes of Lesch-Nyhan, deLange, and Tourette. This type of self-mutilation is the focus of biological research or endorphins and on dopamine receptors. Skin cutting and burning, the most common type of self-m…

Endogenous morphines and pain.

The involvement of endogenous morphines (enkephalins and endorphins) in the regulation of pain is demonstrated by the following experimental evidence: (a) their analgesic activities; (b) their distribution in the central nervous systems; (c) the effects of their modifiers, especially of their antagonists, on nociceptive reactions and (or) on various types of analgesia; (d) rare modifications of their brain levels in pain and (or) analgesic states. Besides the well-known facts, the following items are particularly stressed: the functional roles of hypothalamic structures and of the pituitary, t…

[Effect of Chinese herbal medicine for nourishing yin and removing fire on biosynthesis, secretion and regulative mechanism of gonadotropin-releasing hormone in hypothalamus].

OBJECTIVE: To elucidate the effect of Chinese herbal medicine (CHM) for nourishing Yin and removing fire on the biosynthesis, secretion and regulative mechanism of gonadotropin-releasing hormone (GnRH) in hypothalamus. METHODS: The brain slices of medial basal hypothalamus of adolescent rats, which had been fed with CHM, were incubated. The content of GnRH in incubative liquid was determined during the slices were stimulating with high KC1 to observe the change of GnRH biosynthesis from tonic secretory center of GnRH (arcuate nucleus and ventromedial nucleus) in hypothalamus. Naloxone and self-mutilation.

A 15-year-old male with a long history of self-mutilation resembling the Lesch-Nyhan syndrome, but with normal uric acid levels, was treated with naloxone. Pain-inducing behavior decreased in the evenings following infusion of the drug, and was markedly reduced for 2 days following the treatment period. Though not recommended as therapy, the observations suggest that naloxone may play a role in the regulation of endorphin/enkephalin neural systems.

Feeding behavior and its responsiveness to naloxone differ in lean and obese sheep.

Feeding and its regulation by opioids were studied in lean sheep and sheep in the static phase of dietary obesity. Sheep were fasted 16 h and on separate days were injected IV with 0 (saline), 0.01, 0.1, 1, or 3 mg/kg naloxone 5 min before they were allowed ad lib intake for the ensuing 32 h. All sheep were in chronic zero energy balance when not fed ad lib during naloxone treatment. After 0 mg/kg naloxone, intake rate was at least twice as fast (p < 0.05) in lean than obese sheep through the first 4 h of ad lib feeding, but was similar (approximately 0.5 g/min) in both groups of sheep after 8

[Study of the profiles of prolactin and other hormones in both maternal plasma and amniotic fluid during labor].

The effects of a dopamine antagonist and stress of labor on prolactin (PRL) concentrations were studied in maternal plasma and amniotic fluid during term delivery. In procedure 1, maternal blood and amniotic fluid samples were obtained from 5 normal full-term deliveries at 20-minute intervals for 180 minutes after the intravenous bolus of 10 mg metoclopramide (MCP) during labor just before delivery. Amniotic fluid samples were drawn through a transcervical intrauterine pressure catheter to avoid contamination with maternal blood. PRL, 17 beta-estradiol (E2), progesterone (P) and cortisol were

Menstrual migraine, old and new.

The authors reviewed the connection between sexual hormones and migraine crisis. Besides other exogenous factors, the fall of estradiol blood levels in the late luteal and premenstrual phase seems a causal factor in the origin of menstrual-related headache crisis. The behaviour of migraine crisis during the various events of female reproductive life, support this view. The role of PRL, T, FSH and LH was also discussed. On the other hand, menstrual migraine represents a model that fits perfectly with a neuroendocrine hypothesis which is based upon a faulty chronobiological response of the so-ca

EEG sleep patterns and endogenous opioids in infants of narcotic-addicted mothers.

The early symptoms of brain dysfunction in infants born to heroin-addicted mothers were studied during the first month of life by a multimethod approach. Neurological examination and polygraphic EEG studies of sleep patterns were performed; concomitantly plasma levels of beta-endorphin and met-enkephalin were measured. Whenever possible, these parameters were correlated with each other and with the severity of the withdrawal syndrome. While the neurological examination of addicted newborns was almost negative, altered sleep cycles with a decrease in active sleep and an increase in quiet sleep

Correlation of intratumoral mast cell quantity with psychosocial distress in patients with pancreatic cancer: the PancStress study.

Mast cells are commonly found in pancreatic ductal adenocarcinoma (PDAC), yet their role in the disease remains uncertain. Although mast cells have been associated with depression in several diseases, their connection to PDAC in this context remains unclear. This study explored the correlation between mast cells and psychosocial stress in patients with PDAC. Prior to surgery, 40 patients with PDAC (n = 29 primary resected, n = 11 neoadjuvant treated) completed four questionnaires assessing stress and quality of life. Immunostaining was performed on the resected tumor tissue. Spearman analysis

Folic acid ameliorates depression-like behaviour in a rat model of chronic unpredictable mild stress.

BACKGROUND: Depression is characterized by significant and low mood. Classical antidepressants are still not adequate in treating depression because of undesirable side effects. Folic acid, a member of the vitamin B complex, in considered to be strongly associated with the function and development of the central nervous system. Thus, in this study, we established a model of depression through chronic unpredictable mild stress (CUMS) in rats and assessed the antidepressant effects and mechanisms of folic acid. METHODS: Sprague-Dawley rats were randomly divided into four groups: control, chroni

Inhibiting roles of berberine in gut movement of rodents are related to activation of the endogenous opioid system.

Although Berberine (BER) is popular in treating gastrointestinal (GI) disorders, its mechanisms are not clear yet. In order to investigate the effects and possible mechanism of BER on GI motility in rodents, we first explored GI motility by recording the myoelectrical activity of jejunum and colon in rats, and upper GI transit with a charcoal marker in mice. Then, the plasma levels of gastrin, motilin, somatostatin and glucagon-like-peptide-1 (Glp-1) were measured by ELISA or radioimmunoassay (RIA). Furthermore, endogenous opioid-peptides (β-endorphin, dynorphin-A, met-enkephalin) were detecte

High-efficiency transduction of liver cancer cells by recombinant adeno-associated virus serotype 3 vectors.

Opioids have previously been shown to affect proliferation and differentiation in various neural cell types. In the present study, cultured rat adult hippocampal progenitors (AHPs) were shown to release beta-endorphin. Membrane preparations of AHPs were found to bind [125I]beta-endorphin, and immunoreactivity for mu- and delta-opioid receptors (MORs and DORs), but not for kappa-opioid receptors (KORs), was found on cells in culture. Both DNA content and [3H]thymidine incorporation were reduced after a 48-h incubation with 100 microM naloxone, 10 micro m naltrindole or 10 microM beta-funaltrexa

Hypertensive state, independent of hypertrophy, exhibits an attenuated decrease in systolic function on cardiac kappa-opioid receptor stimulation.

Opioids/opiates are commonly administered to alleviate pain, unload the heart, or decrease breathlessness in patients with advanced heart failure. As such, it is important to evaluate whether the myocardial opioidergic system is altered in cardiac disease. A hamster model of spontaneous hypertension was investigated before the development of hypertension (1 mo of age) and in the hypertensive state (10 mo of age) to evaluate the effect of prolonged hypertension on myocardial opioidergic activity. Plasma beta-endorphin was decreased before the development of hypertension and in the hypertensive

Naloxone, an opiate receptor antagonist, ameliorates acute experimental autoimmune myocarditis by reducing cytotoxic activities.

Little is known about effects of naloxone, an opiate receptor antagonist, upon experimental autoimmune myocarditis (EAM) in mice. The aim of the present study was to test the effects of naloxone upon EAM in mice. Four-week-old C57BL/6 mice were injected with porcine cardiac myosin. Naloxone 1 mg/kg/day was given intraperitoneally daily on days 0 to 21 in experiment I (acute stage) and on days 21 to 42 in experiment II (chronic stage). For the analysis of endogenous opiate and neurohumoral system, plasma beta-endorphin and cytokines were measured. The cytotoxic activity of lymphocytes was deter
