It has long been suspected that hormonal factors contribute directly and indirectly to the etiology and pathogenesis of multiple sclerosis (MS). The susceptibility of MS is higher in women than in men and women are even more susceptible to hormonal influences when onset occurs at an early or delayed age. Pregnancy has a short-term favorable effect on the course of the disease but there is an increased rate of relapse during the post-partum period. In addition, women often report premenstrual exacerbation of their symptoms with remission during menses. These findings suggest that in women estro

Estrogen receptor-positive (ER+) breast cancer accounts for 70-80% of all invasive breast malignancies worldwide and remains the dominant subtype driving rising global breast cancer incidence, representing a major clinical burden in China with a 2.84% annual age-standardized incidence increase. Endocrine therapy serves as the cornerstone intervention for ER+ disease, yet primary and acquired drug resistance severely undermine long-term therapeutic efficacy, creating an urgent demand for systematic integration of updated mechanistic and translational evidence. This comprehensive review systemat

Extensive progress has been made to understand the pathophysiology of stroke but it is still a major cause of mortality and disability worldwide. There are few strategies for the treatment of this disease and the use of thrombolytic tissue plasminogen activator is limited due to the narrow time window. However, the administration of neuroactive steroids could be considered as a potential treatment approach to decrease ischemia-induced lesions. Neurosteroids receptors play important roles in neuroprotection mediated by these hormones. Membrane and intracellular receptors are both involved in th

(1) Raloxifene is marketed in France for the prevention of non traumatic vertebral fracture in postmenopausal women. In animal pharmacology studies it was found to both agonise and antagonise oestrogen. (2) The assessment file is methodologically sound but fails to answer a good number of practical questions. (3) A placebo-controlled trial showed that raloxifene reduced the risk of vertebral collapse after two years of treatment, in both the primary and secondary prevention settings, but no effect was demonstrated on non vertebral fractures. Furthermore, raloxifene reduced the risk of breast c

Oral contraceptives (OC) and smoking-derived nicotine (N) are known to synergistically increase the risk and severity of cerebral ischemia in women. Although it has been known for some time that long-term use of OC and nicotine will have an increased risk of peripheral thrombus formation, little is known about how the combination of OC and nicotine increases severity of brain ischemia. Recent laboratory studies simulating the conditions of nicotine exposure produced by cigarette smoking and OC regimen of women in female rats confirms that the severity of ischemic hippocampal damage is far grea

G protein-coupled estrogen receptor 1 (GPER1), is a functional estrogen receptor involved in estrogen related actions on several systems including processes of the nervous, reproductive, metabolic, cardiovascular, and immune system. Regarding the latter, GPER is expressed in peripheral B and T lymphocytes as well as in monocytes, eosinophils, and neutrophils. Several studies have implicated GPER in immune-mediated diseases like multiple sclerosis, Parkinson's disease, and atherosclerosis-related inflammation, while a recent report suggests that its deletion could be responsible for a form of f

Estrogens, acting through its two receptors, ESR1 (hereafter designated ER alpha) and ESR2 (hereafter designated ER beta), have diverse physiological effects in the reproductive system, bone, cardiovascular system, hematopoiesis, and central and peripheral nervous systems. Mice with inactivated ER alpha, ER beta, or both show a number of interesting phenotypes, including incompletely differentiated epithelium in tissues under steroidal control (prostate, ovary, mammary, and salivary glands) and defective ovulation reminiscent of polycystic ovarian syndrome in humans (in ER beta-/- mice), and o

The endometrium is crucial for the perpetuation of human species. It is a complex and dynamic tissue lining the inner wall of the uterus, regulated throughout a woman's life based on estrogen and progesterone fluctuations. During each menstrual cycle, this multicellular tissue undergoes cyclical changes, including regeneration, differentiation in order to allow egg implantation and embryo development, or shedding of the functional layer in the absence of pregnancy. The biology of the endometrium relies on paracrine interactions between epithelial and stromal cells involving complex signaling p

Single nucleotide polymorphisms (SNPs) are well-known for shaping human drug responses and disease susceptibility, yet their role in mediating environmental responses remains largely unexplored. Here, we systematically investigated SNPs within the ligand-binding domains (LBDs) of 48 nuclear receptors (NRs), focusing on their influence on receptor-ligand interactions. Through the integration of genomic databases, we identified 27 missense SNPs with global minor allele frequencies (MAFs) of >0.001 and mapped their distribution across NR LBDs. Structure-based molecular docking revealed significan

Sarcopenia, defined as the loss of muscle mass and function, is a widely prevalent and severe condition in older adults. Since 2016, it is recognized as a disease. Strength exercise training and nutritional support are the frontline treatment of sarcopenia, with no drug currently approved for this indication. However, new therapeutic options are emerging. In this review, we evidenced that only very few trials have focused on sarcopenia/sarcopenic patients. Most drug trials were performed in different clinical older populations (e.g., men with hypogonadism, post-menopausal women at risk for ost

This paper focuses on the question whether different estrogens (E) have different qualitative pharmacodynamic effects when used by women for contraception, Hormone Replacement Therapy (HRT) or prevention of osteoporosis. In this context estrogens have been defined as the estrogen agonists estradiol (E2), estrone (E1), estriol (E3), conjugated equine estrogens (CEE), diethylstilbestrol (DES) and ethinylestradiol (EE). Selective Estrogen Receptor Modulator's (SERM's) have been excluded from this analysis primarily because of lack of comparative (clinical) data with estrogen agonists. A major pro

Postmenopausal osteoporosis is initiated by estrogen withdrawal and is characterized mainly by overactivated osteoclastic bone resorption. Targeting TNF receptor-associated factor 6 (TRAF6) or its downstream signaling pathways to modulate osteoclast formation and function is an appealing strategy for osteoclast-related disorders. In the present study, we determined the effect of tomatidine, a steroidal alkaloid derived from Solanaceae, on the formation and function of receptor activator of NF-κB (RANK) ligand-induced osteoclasts and the underlying mechanism. Tomatidine inhibited osteoclast for

Breast cancer is the major cause of cancer-related death in women. Its treatment is particularly difficult when metastasis occurs. The ability of cancer cells to move and invade the surrounding environment is the basis of local and distant metastasis. Cancer cells are able to remodel the actin cytoskeleton, which requires the recruitment of numerous structural and regulatory proteins that modulate actin filaments dynamics, including Paxillin or the Neural Wiskott-Aldrich Syndrome Protein (N-WASP). We show that 17-β estradiol (E2) induces phosphorylation of Paxillin and its translocation toward

Three different conformations of the ligand binding domain of the human estrogen receptor (ER-LBD) are observed for the native state when binding an agonist and when binding an antagonist. By conjugating ER-LBD conformation specific peptides to CdS nanoparticles, the three different states can be identified by anodic stripping voltammetry. This electrochemical sensor can detect and distinguish the binding of different ligands to the human estrogen receptor.

OBJECTIVE: To test the hypothesis that the expression of the different isoforms of the estrogen receptor alpha (ER-α) and beta (ER-β) and the progesterone receptor A (PR-A) and B (PR-B) would be differentially modulated in uteri with adenomyosis compared with controls and that modulation would be related to the menstrual cycle.
DESIGN: Case control, blinded comparison.
SETTING: University department.
PATIENT(S): 54 premenopausal women with and 35 without uterine adenomyosis as the sole pathology.
INTERVENTION(S): Multiple samples studied using immunohistochemistry for estrogen and progeste

Estrogens act by binding to estrogen receptors alpha and beta (ERα, ERβ), ligand-dependent transcription factors that play crucial roles in sex differentiation, tumor growth and cardiovascular physiology. Estrogens also activate the G protein-coupled estrogen receptor (GPER), however the function of GPER in vivo is less well understood. Here we find that GPER is required for normal heart rate in zebrafish embryos. Acute exposure to estrogens increased heart rate in wildtype and in ERα and ERβ mutant embryos but not in GPER mutants. GPER mutant embryos exhibited reduced basal heart rate, while

BACKGROUND: Acquired protein S (PS) deficiency is highly associated with elevated circulating estrogen levels resulting from pregnancy, oral contraceptives, and estrogen replacement therapy; however, the mechanism of estrogen-mediated acquired PS deficiency remains poorly understood. Increasing evidence indicates that estrogen receptor signaling can indirectly modulate the expression of target genes at the post-transcriptional level by modulating the expression of microRNAs (miRNAs), and miRNAs have also been demonstrated to be involved in the regulation of hemostasis.
OBJECTIVES: To investig

BACKGROUND: Titanium dioxide nanoparticles (TiO₂NPs) are widely used metal nanoparticles capable of accumulating in tissues and exerting endocrine-disrupting effects. Their impact on female reproductive physiology remains largely unclear. This study aimed to elucidate the endocrine-disrupting properties of TiO₂NPs by assessing ovarian and uterine histology, serum hormone levels, estrous cycle changes, and receptor expression patterns in both intact and ovariectomized female rats.
METHODS: Thirty-two Sprague Dawley rats were randomly divided into four groups (n = 8/group): intact control, inta

Osteoarthritis (OA) is the most prevalent degenerative joint disease worldwide and a leading cause of pain, disability, and reduced quality of life, with pronounced sex-specific differences in incidence and clinical presentation. Epidemiological evidence shows a sharp increase in OA prevalence in women after menopause, whereas in men, the elevated risk at older age is more closely associated with declines in bone mass, muscle strength, and biomechanical stability. Increasing experimental and clinical studies indicate that sex hormones-including estrogen, androgens, and progesterone-and their r
