Chitosan-based mucoadhesive films loaded with curcumin for topical treatment of oral cancer.

This study aimed to develop mucoadhesive chitosan-based films capable of enhancing the curcumin penetration into the oral mucosa to treat oral cancers. We developed three films containing medium molecular weight chitosan (190-310 KDa) and other excipients (polyvinyl alcohol, Poloxamer®407, and propylene glycol) that have proven to be compatible with each other and with curcumin in thermal analyses. The films were smooth, flexible, and precipitates free, with uniform weight and thickness, pH compatible with the oral mucosa, resistance to traction, and entrapped curcumin in a high proportion. Th

Thermosensitive hexanoyl glycol chitosan-based ocular delivery system for glaucoma therapy.

UNLABELLED: Conventional eye drops quickly move away from the surface of the eye; as a result, ocular bioavailability is very limited. To overcome this issue, we developed a thermosensitive hexanoyl glycol chitosan (HGC) as a carrier for topical drug delivery to the eye. Here, we modulated the degree of N-hexanoylation to control the thermogelling behavior and prepared a new ocular formulation of HGC for glaucoma therapy. The viscosity of the aqueous formulation sharply and significantly increases at body temperature. The results from cytotoxicity evaluation showed that HGC is non-toxic at up

An evaluation of 2-benzyl-1-naphthol (DuP 654) analogs as systemic anti-inflammatory agents.

DuP 654 (2-benzyl-1-naphthol) is a topically active anti-inflammatory agent that was evaluated in phase II clinical trials as an anti-psoriatic agent. The compound is a potent 5-lipoxygenase inhibitor and exhibits inhibitory activity against lipopolysaccharide-stimulated release of interleukin-1 from human monocytes. DuP 654 cannot be used as a systemic anti-inflammatory compound due to its rapid and extensive metabolism. Fifteen analogs were synthesized in an attempt to block the systemic route(s) of metabolism. The compounds were evaluated (IP and PO) in the rat carrageenan paw edema inflamm

Multifunctional System with Ferulic Acid: Increased Safety, Antioxidant Activity, and Efficacy of Sunscreen Formulations.

The topical application of ferulic acid (FA) is limited by its low solubility and high susceptibility to oxidation. To overcome these limitations, the multicomponent system Cicloferulic® (CF) was developed by mixing FA, hydroxypropyl β-cyclodextrin (HPβ-CD), and polyvinylpyrrolidone K30 (PVP K30), which demonstrated greater safety and antioxidant efficacy than FA alone. The physicochemical characterization of FA, CF, and their components showed greater thermal stability and reduced crystallinity of CF, indicating the formation of the system. In cytotoxicity assays on HaCaT cells, FA maintained

A dual-drug chitosan-hyaluronic acid bandage contact lens accelerates corneal re-epithelialization in a rabbit mechanical debridement model.

Rapid corneal re-epithelialization is crucial to prevent infection and scarring, yet the efficacy of conventional topical therapies is hindered by their limited ocular bioavailability. To address this, we developed a daily replaceable chitosan-hyaluronic acid (CS-HA) bandage contact lens co-loaded with levofloxacin and betamethasone using a soaking-based drug-loading approach. In a rabbit 8-mm mechanical corneal debridement model (n = 15/group), the dual-drug CS-HA lens was compared with a chitosan-only drug-loaded lens, a commercial silicone-hydrogel bandage lens plus drops, and drops alone.

Repurposing copper diethyldithiocarbamate (CuET) in a polymeric nanofibrous patch for topical therapy of psoriasis.

The management of psoriasis remains challenging due to the limitations of current therapies. This study explores the repurposing of copper diethyldithiocarbamate (CuET)-a metabolite of the anti-alcoholism drug disulfiram-as a novel topical agent for psoriasis. To this end, we developed an advanced delivery platform by fabricating a CuET-loaded nanofiber patch via electrospinning of a polyvinylpyrrolidone/polyethylene glycol/citric acid (PVP/PEG/CA) blend. This rationally designed matrix provided optimal mechanical strength, moisture-activated adhesion, and precise local drug delivery performan

Hair Growth-Promoting Effect of a Horse Oil-Folium Isatidis Topical Preparation via Modulation of VEGF, TGF-β1, and β-Catenin Signaling in C57BL/6 Alopecia Mice: A Pilot Study.

Wnt/β-catenin signaling is a key regulator of hair follicle cycle progression, and aberrant activation or suppression of this pathway has been implicated in the pathogenesis of alopecia. Horse oil (HO) and Folium Isatidis (FI) have been traditionally used in East Asian medicine for hair growth, but their combined molecular mechanism remains unclear. We evaluated a topical preparation combining HO and FI (HOFI) in two C57BL/6 mouse models: physiological depilation (telogen-to-anagen transition) and testosterone-induced seborrheic alopecia. Hair growth was assessed by gross appearance, hair leng

Anti-inflammatory efficacy of a novel diclofenac-loaded liposomal gel in carrageenan-induced paw edema model: a preclinical and radiographic study.

OBJECTIVE: This research investigates the anti-inflammatory efficacy of an innovative liposomal gel formulation containing diclofenac. SIGNIFICANCE: Topical administration of Non-Steroidal Anti-inflammatory Drugs (NSAIDs) provides significant local therapeutic effects, while substantially lowering the risk of systemic or central adverse effects. However, topically applied NSAIDs have also failed to achieve desired effects. This issue may arise from insufficient cutaneous permeation of active pharmaceutical agents to achieve clinically effective drug levels. Research indicates that effective t

Deployment and transcriptional evaluation of nitisinone, an FDA-approved drug, to control bed bugs.

BACKGROUND: Bed bugs are blood-feeders that rapidly proliferate into large indoor infestations. Their bites can cause allergies, secondary infections and psychological stress, among other problems. Although several tactics for their management have been used, bed bugs continue to spread worldwide wherever humans reside. This is mainly due to human-mediated transport and their high resistance to several classes of insecticides. New treatment options with novel modes of action are required for their control. In this study, we evaluated the use of nitisinone (NTBC), an FDA-approved drug, for bed
