Onchocerca volvulus: identification of cDNAs encoding a putative phosphatidyl-ethanolamine-binding protein and a putative partially processed mRNA precursor.

We have identified and sequenced cDNAs of the parasitic nematode Onchocerca volvulus which encode a homologue of phosphatidylethanolamine-binding proteins from mammals. These clones are also closely related to the O. volvulus Ov16 cDNA (Lobos et al., 1990). One identified cDNA clone appears to represent a partially processed precursor. These results suggest that these cDNAs are derived from a complex genomic locus, raising the possibility of polycistronic transcription in this nematode.

Analysis of the fit-1 gene encoding a ECM protein, flectin, in Caenorhabditis elegans.

Flectin is a new type of extracellular matrix protein and its function was suggested to provide a microenvironment of great elasticity. The C. elegans genome database revealed the presence of a flectin homologue, fit-1, which shows approximately 40% similarity (20% identity) to chick flectin. Here we propose a new gene structure for the fit-1 based on our experiments and the partial cDNA clones obtained from Y. Kohara and further suggest that the previous gene prediction is incorrect. FLT-1 is shown to be expressed in various neurons, hypodermal cells, distal tip cells and vulva epithelial cel

Transcriptional profiling and functional characterization of the Hc-NHR-49 gene in ivermectin resistance of Haemonchus contortus.

BACKGROUND: The barber's pole worm (BPW), Haemonchus contortus, poses a significant threat to sheep health and livestock husbandry. Control has historically relied on synthetic anthelmintics such as ivermectin; however, widespread resistance to this drug has emerged. The functional role of nuclear hormone receptors (NHRs) in H. contortus remains poorly understood. METHODS: The Hc-NHR-49 gene was polymerase chain reaction (PCR)-amplified and subjected to bioinformatic analysis. Expression profiles of the gene in resistant and sensitive strains across different developmental stages and under iv

Transcriptome profiling reveals the immunomodulatory role of Echinococcus granulosus EgG1Y162 on macrophages in vitro.

Echinococcus granulosus (E. granulosus) infection can induce host immunosuppression and chronic liver injury, with current therapeutic approaches exhibiting significant limitations. EgG1Y162, a promising vaccine candidate against E. granulosus, shows considerable potential; however, its specific effects on macrophages remain unknown, necessitating further investigation. This study employed RNA-seq to analyze the functional enrichment of differentially expressed genes (DEGs) in macrophages following EgG1Y162 transfection, aiming to elucidate its regulatory impact on cytokines. Additionally, EgG

Wenyonia gracilis sp. n. (Cestoda: Caryophyllidea) from Synodontis zambezensis (Siluriformes: Mochokidae): the first native caryophyllidean tapeworm from southern Africa.

Parasitological examination of freshwater fishes of the Phongolo River in north-eastern KwaZulu-Natal, South Africa resulted in the discovery and morphological and molecular characterisation of a new species of Wenyonia Woodland, 1923 (Cestoda: Caryophyllidea). The new species from the plain squeaker, Synodontis zambezensis Peters (Siluriformes: Mochokidae), is morphologically most similar to Wenyonia acuminata Woodland, 1923, a species reported from three species of Synodontis in north-eastern, western and central Africa (Sudan, Nigeria, Democratic Republic of the Congo). Both these species a

Helminth-induced Th2 cell dysfunction is distinct from exhaustion and is maintained in the absence of antigen.

T cell-intrinsic regulation, such as anergy, adaptive tolerance and exhaustion, is central to immune regulation. In contrast to Type 1 and Type 17 settings, knowledge of the intrinsic fate and function of Th2 cells in chronic Type 2 immune responses is lacking. We previously showed that Th2 cells develop a PD-1/PD-L2-dependent intrinsically hypo-responsive phenotype during infection with the filarial nematode Litomosoides sigmodontis, denoted by impaired functionality and parasite killing. This study aimed to elucidate the transcriptional changes underlying Th2 cell-intrinsic hypo-responsivene

Methods for Studying the Germline of the Human Parasite Schistosoma mansoni.

Schistosomes are flatworm parasites that claim the lives of more than 200,000 people in poverty-stricken regions every year. Much of the pathology due to infection is the direct result of injury spurred by the parasite's eggs becoming lodged in host tissues. Thus, asking basic questions about germ cell biology may not only identify novel therapeutic approaches, but could also uncover conserved mechanisms that regulate the germline in diverse metazoa. Here, we detail useful methods for studying the schistosome germline including EdU labeling, whole-mount in situ hybridization, and RNA interfere

GPCR and IR genes in Schistosoma mansoni miracidia.

BACKGROUND: Schistosoma species are responsible for the disease schistosomiasis, a highly prevalent helminthic disease that requires a freshwater snail as intermediate host. The S. mansoni free-living miracidium must utilize olfaction to find a suitable snail host, and certain types of rhodopsin G protein-coupled receptors (GPCRs) and ionotropic receptors (IRs) have been identified as olfactory receptors in other animal phyla. The Schistosoma genome project, together with the recent availability of proteomic databases, allowed for studies to explore receptors within S. mansoni, some of which m

Protection of pigs against Taenia solium cysticercosis by immunization with novel recombinant antigens.

Recombinant antigens from the oncosphere stage of the parasite Taenia solium were expressed in Escherichia coli. The TSOL16, TSOL45-1A and TSOL45-1B recombinant antigens, each consisting of fibronectin type III (FnIII) domain S, were produced as fusion proteins with glutathione S-transferase (GST) and maltose binding protein (MBP). Groups of pigs were immunized twice with the GST fusions of the antigens and boosted a third time with the MBP fusions prior to receiving a challenge infection with T. solium eggs. The TSOL16 antigen was found to be capable of inducing high levels of immunity in pig

Immunity to Trichinella spiralis infection in vitamin A-deficient mice.

Vitamin A-deficient (A-) mice make strikingly poor IgG responses when they are immunized with purified protein antigens. Previously, we showed that A- T cells overproduce interferon gamma (IFN-gamma), which then could inhibit interleukin 4 (IL-4)-stimulated B cell IgG responses. To determine if the altered IFN-gamma regulation pattern and its immunological consequences would extend to a natural infection, we studied mice infected with the parasitic helminth Trichinella spiralis. The course of the infection was similar in A- and A-sufficient (A+) mice. These mice did not differ with respect to

Self-reactive IgG4 antibodies are associated with blocking of pathology in human lymphatic filariasis.

Lymphatic filariasis, a chronic disfiguring disease exhibits complex pathology. Based on different clinical manifestations, infected individuals are categorized into asymptomatic-carriers and chronic-patients. The mechanism behind differential clinical outcomes remains unclear. Roles of filaria-specific B cell responses in filariasis have been documented, whereas the contribution of B1 cell response and poly-specific IgG and IgA in the context of clinical filariasis is not deciphered. In this study, we measured the poly-specific IgG and IgA levels in different clinical categories of filariasis

Immunological cross-reaction between an Onchocerca paramyosin-like molecule and a microfilaria surface antigen.

A monoclonal antibody (2A5B9), previously shown to be reactive with a 14 kD surface associated antigen of Onchocerca microfilariae, was found to recognise a 92 kD molecule present in an adult worm extract. The antibody was used to select cDNA clones with a coding capacity larger than 14 kD, from a lambda gt11 library of O. volvulus. Nucleotide sequencing of the cDNA of one such clone revealed extensive homology to the myosin (unc-54) and paramyosin (unc-15) genes of Caenorhabditis elegans, similarly to myosin and paramyosin genes of Onchocerca volvulus, Brugia malayi, Dirofilaria immitis and S

[Preliminary evaluation of promoter regions of four Schistosoma japonicum genes for expressing a luciferase reporter].

OBJECTIVE: To evaluate the relevant promoter regions of four Schistosoma japonicum genes for expressing a luciferase reporter. METHODS: The polymerase chain reaction (PCR) was used to amplify the promoter regions of four S. japonicum genes and then each PCR product was cloned into a pGlu-Basic vector according to the standard molecular procedures. These recombinant plasmids were either transfected into human HEK293 cells by using lipofectamine or introduced into cultured schistosomes by electroporation. Then, the luciferase activities were measured by using a dual luciferase reporter system i

Cloning, expression, and preliminary characterization of the dysferlin tegument protein in Schistosoma japonicum.

The schistosomal tegument is a dynamic host-interactive layer. Proteins exposed to the host on the tegumental surface are important for completion of the parasitic lifecycle. Dysferlin is a member of the ferlin family and is involved in plasma membrane repair. Based on the results of a proteomics study of tegument surface proteins of Schistosoma japonicum in our laboratory, dysferlin was identified as a tegumental protein of S. japonicum. The gene encoding S. japonicum dysferlin (SjDF), which codes for several Ca(2+) binding sites, was cloned, expressed in Escherichia coli, and characterized.

Dominance of P-glycoprotein 12 in phenotypic resistance conversion against ivermectin in Caenorhabditis elegans.

While diseases caused by nematodes remains a considerable drawback for the livestock, agriculture and public health, anthelmintics drug resistance has been observed over the past years and is a major concern for parasite control. Ivermectin, initially considered as a highly potent drug, currently presents a reduced anti-helminthic efficacy, which is influenced by expression of several ATP-binding cassette transporters (ABC), among them the P-glycoproteins (Pgps). Here we present some evidences of Pgps dominance during Ivermectin resistance/susceptibility using Pgps double silencing in C. elega

Gene expression of ABC transporters in Cooperia oncophora after field and laboratory selection with macrocyclic lactones.

The most widespread helminth parasites of grazing cattle in northern Europe are the gastrointestinal nematodes Ostertagia ostertagi and Cooperia oncophora. Heavy reliance on the use of macrocyclic lactone (ML) in cattle has led to world-wide emergence of resistance to this drug class in C. oncophora. There is evidence that members of the ATP-binding cassette (ABC) transporter family, such as P-glycoproteins (P-gp) and multidrug-resistant proteins (MRP), play a role in resistance to ML. In this study gene expression of Con-pgp9, Con-pgp11, Con-pgp12, Con-pgp16 and Con-mrp1 was examined in two i
