ATG5 gene regulates testosterone synthesis of testicular Leydig cells in Hezuo pig.

Autophagy-related gene 5 (ATG5) plays a crucial role in autophagosome formation. Recent studies have investigated the role of autophagy in regulating testosterone production; however, its expression in testicular tissues and Leydig cells of Hezuo pig remains less understood. In this study, we cloned the coding sequence (CDS) region of the ATG5 gene and assessed its expression using qPCR across various tissues and testes at different developmental stages in Hezuo pigs. Subsequently, we constructed silencing and overexpression vectors for ATG5 and transfected them into Leydig cells. Cell prolife

Using siRNA-Based Anti-Inflammatory Lipid Nanoparticles for Gene Regulation in Psoriasis.

BACKGROUND: Psoriasis is a chronic inflammatory autoimmune disease, yet it affects hundreds of millions of people. Long-term effective intervention of the disease by targeting the causative genes via RNAi (RNA interference) has become a reality. However, its further application is hindered by inflammatory side effects caused by delivery systems such as LNP (lipid nanoparticles). PURPOSE: This study aimed to develop a novel anti-inflammatory LNP rationally tailored for topical application in psoriasis and to validate its potential to deliver Stat3 (signal transducer and activator of transcript

Epilepsy in NHS actin remodeling regulator gene (NHS) and genotype-phenotype correlations.

BACKGROUND: NHS gene encodes a protein with four conserved nuclear localization signals that regulates actin assembly and cell spreading. NHS variants are associated with cataract-related disorders with/without seizures. However, the NHS-epilepsy association remains unclear. METHODS: Trio-based whole-exome sequencing was performed in patients with epilepsy of unknown cause. Previously reported variants were reviewed to analyze the mechanisms underlying phenotypical variations. RESULTS: Seven NHS variants were identified in one female and six male patients. All the variants were not present i

Oral supplementation of fucoxanthin regulates gene expression in the brain of middle-aged rats.

Age is the main risk factor for many neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease and frontotemporal dementia. Despite our limited understanding of cellular mechanisms of ageing-associated neuronal loss, an increasing number of studies demonstrate that oxidative stress and inflammation are key drivers. Epidemiological studies indicate that diet during middle adulthood can influence the risk of developing neurodegenerative diseases later in life, so it is important to investigate dietary interventions to combat oxidative stress and inflammation. In this study, we

Light-regulated microRNAs shape dynamic gene expression in the zebrafish circadian clock.

A key property of the circadian clock is that it is reset by light to remain synchronized with the day-night cycle. An attractive model to explore light input to the circadian clock in vertebrates is the zebrafish. Circadian clocks in zebrafish peripheral tissues and even zebrafish-derived cell lines are entrainable by direct light exposure thus providing unique insight into the function and evolution of light regulatory pathways. Our previous work has revealed that light-induced gene transcription is a key step in the entrainment of the circadian clock as well as enabling the more general ada

Antitumor Activity of Warbugia ugandensis: Methanolic Extracts and Gene Regulation in Colorectal Cancer.

A promising approach to accelerating the development of innovative anti-cancer therapies involves the evaluation of natural plant compounds. In this study, we focused on examining the effects of Warbugia ugandensis (W. ugandensis) methanolic root and stem infusions on the activity of five target genes-COX-2, CASPS-9, Bcl-xL, Bcl2, and 5-LOX-using colorectal cancer (CRC) cell lines (Caco-2). The plant extracts were prepared for testing by dissolving them in dimethyl sulfoxide (DMSO) after undergoing a step-by-step extraction process. Caco-2 cells were then treated with different concentrations

Cocaine-induced gene regulation in D1 and D2 neuronal ensembles of the nucleus accumbens.

Cocaine use disorder is characterized by persistent drug-seeking behavior and a high risk of relapse, driven in part by lasting molecular and circuit adaptations in the nucleus accumbens. To explore the transcriptomic changes underlying these alterations, we employed fluorescence-activated nucleus sorting coupled with single-nucleus RNA sequencing to analyze D1 and D2 medium spiny neurons in this brain region of male mice subjected to acute cocaine exposure or to prolonged withdrawal from repeated cocaine exposure without or with an acute cocaine rechallenge. This approach allowed us to precis

Sex chromosome aneuploidy impacts on human gene expression and regulation: a systematic review.

BACKGROUND: Sex chromosome aneuploidies (SCAs) are collectively common genetic disorders that impact diverse body systems. The molecular mechanisms by which an extra or missing sex chromosome increases clinical risk are not fully understood, but they likely involve imbalances in expression and regulation of dosage-sensitive genes. There has been a recent surge in transcriptomic and epigenomic studies on genomic effects of SCAs - making it an opportune time to: (i) map existing knowledge of SCA impacts on gene expression and regulation; (ii) resolve consensus findings on SCA dosage-sensitive ge

Programmable chronogenetic gene circuits for self-regulated circadian delivery of biologic drugs.

Cells of the body rely on the circadian clock to orchestrate daily changes in physiology that impact both homeostatic and pathological conditions, such as the inflammatory autoimmune disease rheumatoid arthritis (RA). In RA, high levels of proinflammatory cytokines peak early in the morning hours, reflected by daily changes in joint stiffness. Chronotherapy (or circadian medicine) seeks to delivery drugs at optimal times to maximize their efficacy. However, chronotherapy remains a largely unexplored approach for disease modifying, antirheumatic treatment, particularly for cell-based therapies.

Non-coding RNAs (miRNAs - circRNAs - lncRNAs) and genes interact with the regulation of vitiligo.

Vitiligo is a chronic skin disorder characterized by the loss of melanocytes, resulting in depigmented patches on the skin. The molecular mechanisms underlying vitiligo remain incompletely understood, with recent studies highlighting the role of non-coding RNAs in disease pathogenesis. To identify and analyze the roles of miRNAs, (circular RNAs) circRNAs, and (long non-coding RNAs) lncRNAs in the pathogenesis of vitiligo, focusing on their interactions with key coding genes and pathways. We utilized Microarray data from the GSE65127 and GSE75819 datasets in the Gene Expression Omnibus (GEO) da

Proneural gene Mash1 (Ascl1) is expressed in multiple lineages and regulates their differentiation and specification.

Ascl1 (Mash1), a bHLH transcription factor, is widely expressed by neuronal progenitors. The gene plays a key role in the differentiation of the autonomic nervous system, i.e., sympathetic, parasympathetic, and enteric ganglia; all of which are derived from neural crest cells. In Ascl1-null mutants, defective development of these ganglia is induced. The differentiation of neuroendocrine cells, including the carotid body, ultimobranchial body and thyroid C cells, the neuroepithelial body in the lung, and the adrenal medulla, is also controlled by Ascl1. Although the carotid body glomus cells an

Stress-Induced Changes in Nucleocytoplasmic Localization of Crucial Factors in Gene Expression Regulation.

This study investigates the toxic effect of harmful materials, unfiltered by the placenta, on neonatal umbilical cord (UC) vessels, focusing on stress-induced adaptations in transcriptional and translational processes. It aims to analyze changes in pathways related to mRNA condensate formation, transcriptional regulation, and DNA damage response under maternal smoking-induced stress. UC vessels from neonates born to smoking (Sm) and nonsmoking mothers (Ctr) were examined. Immunofluorescence staining and confocal microscopy assessed the localization of key markers, including Transcription Compl

miR-665-Mediated Regulation of AHCYL2 and BVES Genes in Recurrent Implantation Failure.

The primary goal of this investigation was to identify mRNA targets affected by dysregulated miRNAs in RIF. This was accomplished by comprehensively analyzing mRNA and miRNA expression profiles in two groups: female subjects with normal reproductive function (control, n = 5) and female subjects experiencing recurrent implantation failure (RIF, n = 5). We conducted transcriptome sequencing and small RNA sequencing on endometrial tissue samples from these cohorts. Subsequently, we validated a selection of intriguing findings using real-time PCR with samples from the same cohort. In total, our an

[Role of superoxide dismutase in acute pancreatitis: from antioxidant protection to gene regulation].

We present modern data on the role of superoxide dismutase (SOD) in antioxidant protection and gene regulation in acute pancreatitis. Antioxidant enzymes are essential in pathogenesis of numerous diseases. SOD is one of the key enzymes of antioxidant system. In this review, we analyzed activity of this enzyme depending on various factors, mechanisms and role in physiological and pathological processes, in particular, acute pancreatitis. SOD is significantly less active in patients with severe acute pancreatitis accompanied by renal failure, severe circulatory disorders and high mortality. Ther

The role of microRNAs in the regulation of critical genes and signalling pathways that determine endometrial receptivity.

Endometrial receptivity is the ability of the endometrium to accept embryos. Thus, endometrial receptivity dysfunction is an important factor leading to embryo implantation failure. A good endometrial receptivity provides a suitable environment for embryo implantation, improving the embryo implantation rate. The "implantation window" stage, or the receptive stage of the endometrium, is regulated by various hormones, genes, proteins and cytokines, among which microRNAs (miRNAs) and their target genes have a regulatory effect on endometrial receptivity. This review outlines the relationship betw

Transcriptional mechanism of E2F1/TFAP2C/NRF1 in regulating KANK2 gene in nephrotic syndrome.

The mortality rate linked with nephrotic syndrome (NS) is quite high. The renal tubular injury influences the response of NS patients to steroid treatment. KN motif and ankyrin repeat domains 2 (KANK2) regulates actin polymerization, which is required for renal tubular cells to maintain their function. In this study, we found that the levels of KANK2 in patients with NS were considerably lower than those in healthy controls, especially in NS patients with acute kidney injury (AKI). To get a deeper understanding of the KANK2 transcriptional control mechanism, the core promoter region of the KAN

Identification and Characterization of ATOH7-Regulated Target Genes and Pathways in Human Neuroretinal Development.

The proneural transcription factor atonal basic helix-loop-helix transcription factor 7 (ATOH7) is expressed in early progenitors in the developing neuroretina. In vertebrates, this is crucial for the development of retinal ganglion cells (RGCs), as mutant animals show an almost complete absence of RGCs, underdeveloped optic nerves, and aberrations in retinal vessel development. Human mutations are rare and result in autosomal recessive optic nerve hypoplasia (ONH) or severe vascular changes, diagnosed as autosomal recessive persistent hyperplasia of the primary vitreous (PHPVAR). Epididymis-specific RNase A family genes regulate fertility and small RNA processing.

Sperm small RNAs are implicated in intergenerational transmission of paternal environmental effects. Small RNAs generated by the cleavage of tRNAs, known as tRNA fragments (tRFs) or tRNA-derived RNAs (tDRs or tsRNAs), are an abundant class of RNAs in mature sperm and can be modulated by environmental conditions. The biogenesis of tRFs in the male reproductive tract remains poorly understood. Angiogenin, a member of the ribonuclease A superfamily (RNase A), cleaves tRNAs to generate tRFs in response to cellular stress. Four paralogs of Angiogenin, namely Rnase9, Rnase10, Rnase11, and Rnase12, a

Soybean gene GmMLP34 regulates Arabidopsis negative response to high temperature stress.

The functions of major latex proteins (MLPs) in plant defense and stress responses have been widely documented; however, their roles in HT stress response in soybeans have not been elucidated. This study investigated the role of GmMLP34, a member of the major latex protein (MLP) family, in the response of soybeans to HT stress. Transcriptome analysis of HT-resistant (JD21) and HT-sensitive (HD14) soybean leaves under HT stress (43.40 ± 1.70 °C) and field conditions revealed differential expression of GmMLP34. Further examination across different HT-resistant varieties showed that GmMLP34 was d
