The causal relationship between depression and frozen shoulder: A two-sample Mendelian randomization.

To investigate the causal relationship between depression and frozen shoulder using a Mendelian randomization (MR) approach. Pooled data from a large-scale genome-wide association study were used. Genetic loci that were independent of each other and associated with depression and frozen shoulder in populations of European ancestry were selected as instrumental variables. Inverse variance weighting was used as the primary analysis method. Weighted median and MR-Egger were used as complementary analysis methods to assess causal effects. To explore the causal relationship between depression and f

Causal Association Between Thyroid Function and Myeloproliferative Disease: A Two-Sample Mendelian Randomization Study.

BACKGROUND: Thyroid dysfunction has been linked to hematologic abnormalities, but whether thyroid function has a causal role in myeloproliferative diseases (MDs), including chronic myeloproliferative disease (CMD), remains unclear. We used Mendelian randomization (MR) to assess the potential causal effects of thyroid-related traits on MD risk. METHODS: A two-sample MR analysis was performed using genome-wide association study (GWAS) summary statistics. Outcome data for CMD and MD, excluding chronic myelogenous leukemia (CML), were obtained from the Finnish R12 cohort (European ancestry). Gene

Hidradenitis Suppurativa and Smoking, Obesity, Psoriasis, Inflammatory Bowel Disease, and Systemic Sclerosis: Results From A 2-Sample Mendelian Randomization Study.

IMPORTANCE: Smoking and obesity are associated with risk of hidradenitis suppurativa, and both are considered important environmental risk factors. However, a causal relationship remains unproven. OBJECTIVE: To primarily investigate the relationship between body mass index (BMI, calculated as weight in kilograms divided by height in meters squared) and smoking and HS, and secondarily to investigate potential relationships between 3 inflammatory diseases (psoriasis, inflammatory bowel disease [IBD], and systemic sclerosis [SSc]) and HS. DESIGN, SETTING, AND PARTICIPANTS: A mendelian randomiza

Neuroimmune Genetic Overlap Between Chronic Pruritic Skin Diseases and Major Depressive Disorder: Insights from Cross-Trait GWAS Analysis.

BACKGROUND: Psoriasis (PSO), atopic dermatitis (AD), and urticaria (URT) are chronic inflammatory skin diseases marked by severe pruritus. These conditions are frequently comorbid with psychiatric disorders, especially major depressive disorder (MDD). Despite this clinical overlap, the underlying shared genetic mechanisms driving these comorbidities remain largely unclear. METHODS: We integrated large-scale European-ancestry genome-wide association study (GWAS) summary statistics for PSO, AD, URT, and MDD. Genetic overlap was assessed using linkage disequilibrium score regression (LDSC), high

Shared genetic architecture between periodontal disease and type 2 diabetes: a large scale genome-wide cross-trait analysis.

PURPOSE: To investigate the relationship between abnormal glucose metabolism, type 2 diabetes (T2D), and periodontal disease (PER) independent of Body Mass Index (BMI), we employed a genome-wide cross-trait approach to clarify the association. METHODS: Our study utilized the most extensive genome-wide association studies conducted for populations of European ancestry, including PER, T2D, fasting glucose, fasting insulin, 2-hour glucose after an oral glucose challenge, HOMA-β, HOMA-IR (unadjusted or adjusted for BMI) and HbA1c. RESULTS: With this approach, we were able to identify pleiotropic

Epidemiologic and genetic associations between primary biliary cholangitis and extrahepatic rheumatic diseases.

Patients with primary biliary cholangitis (PBC) commonly experience extrahepatic rheumatic diseases. However, the epidemiologic and genetic associations as well as causal relationship between PBC and these extrahepatic conditions remain undetermined. In this study, we first conducted systematic review and meta-analyses by analyzing 73 studies comprising 334,963 participants across 17 countries and found strong phenotypic associations between PBC and rheumatic diseases. Next, we utilized large-scale genome-wide association study summary data to define the shared genetic architecture between PBC

Genetic proxies for calcium channel blockers and cancer: a Mendelian randomization study.

Calcium channel blockers (CCBs) are commonly prescribed antihypertensives. However, concerns exist about potential off-target effects on cancer. This Mendelian randomization (MR) study examined the associations of genetic proxies for CCBs with the risk of cancer. We used published genetic proxies in the target genes of CCBs as instruments, and obtained MR estimates by applying them to large studies of 17 site-specific cancers (non-Hodgkin lymphoma, melanoma, leukemia, thyroid, rectal, pancreatic, oral cavity/pharyngeal, kidney, esophagus/stomach, colon, bladder, endometrial, cervical and breas

Pleiotropic effects of DCDC2 and DYX1C1 genes on language and mathematics traits in nuclear families of developmental dyslexia.

Converging evidence indicates that developmental problems in oral language and mathematics can predate or co-occur with developmental dyslexia (DD). Substantial genetic correlations have been found between language, mathematics and reading traits, independent of the method of sampling. We tested for association of variants of two DD susceptibility genes, DCDC2 and DYX1C1, in nuclear families ascertained through a proband with DD using concurrent measurements of language and mathematics in both probands and siblings by the Quantitative Transmission Disequilibrium Test. Evidence for significant

Preeclampsia, gestational hypertension, and cardiovascular disease risk: a genetic epidemiological study.

BACKGROUND AND AIMS: Observational studies show that women with hypertensive disorders of pregnancy have greater risk of cardiovascular disease later in life. Whether these associations reflect causal pathways is uncertain. This study used genetic epidemiology to explore the causal relevance of preeclampsia and gestational hypertension on cardiovascular disease. METHODS: Two-sample Mendelian randomization (MR) analyses were conducted using summary-level data from FinnGen and from the to date largest consortia for each outcome. One-sample MR analyses were performed using individual-level data

Shared genetic factors influence amygdala volumes and risk for alcoholism.

Alcohol abuse and dependence (alcohol use disorders, AUDs) are associated with brain shrinkage. Subcortical structures including the amygdala, hippocampus, ventral striatum, dorsal striatum, and thalamus subserve reward functioning and may be particularly vulnerable to alcohol-related damage. These structures may also show pre-existing deficits impacting the development and maintenance of AUD. It remains unclear whether there are common genetic features underlying both subcortical volumes and AUD. In this study, structural brain images were acquired from 872 Mexican-American individuals from e

Genetic Architecture of Comorbidity Between Chronic Obstructive Pulmonary Disease and Cardiovascular Diseases: Exploring Shared Mechanisms and Potential Therapeutic Targets.

BACKGROUND: Chronic obstructive pulmonary disease (COPD) and cardiovascular diseases (CVDs), including hypertension (HTN), coronary heart disease (CHD), and heart failure (HF), are major global health burdens. The shared genetic mechanisms underlying the high comorbidity between COPD and CVDs remain unclear. METHODS: We integrated large-scale GWAS summary statistics for COPD and three major CVDs (HTN, CHD, HF). Several analytic approaches were applied, including linkage disequilibrium score regression (LDSC), high-definition likelihood (HDL), multi-marker analysis of genomic annotation (MAGMA

Exploring the interconnected between type 2 diabetes mellitus and nonalcoholic fatty liver disease: Genetic correlation and Mendelian randomization analysis.

Epidemiological and clinical studies have indicated a higher risk of nonalcoholic fatty liver disease (NAFLD) and type 2 diabetes mellitus (T2DM), implying a potentially shared genetic etiology, which is still less explored. Genetic links between T2DM and NAFLD were assessed using linkage disequilibrium score regression and pleiotropic analysis under composite null hypothesis. European GWAS data have identified shared genes, whereas SNP-level pleiotropic analysis under composite null hypothesis has explored pleiotropic loci. generalized gene-set analysis of GWAS data determines pleiotropic pat

Genome arrays for the detection of copy number variations in idiopathic mental retardation, idiopathic generalized epilepsy and neuropsychiatric disorders: lessons for diagnostic workflow and research.

We review the contributions and limitations of genome-wide array-based identification of copy number variants (CNVs) in the clinical diagnostic evaluation of patients with mental retardation (MR) and other brain-related disorders. In unselected MR referrals a causative genomic gain or loss is detected in 14-18% of cases. Usually, such CNVs arise de novo, are not found in healthy subjects, and have a major impact on the phenotype by altering the dosage of multiple genes. This high diagnostic yield justifies array-based segmental aneuploidy screening as the initial genetic test in these patients

Dynamic neuro-immune regulation of psychiatric risk loci in human neurons.

The immune environment influences neurodevelopment and subsequent clinical trajectories for psychiatric outcomes in childhood and adolescence. Yet it remains unclear if the impact of maternal and fetal immune activation varies with distinct polygenic risk profiles. Therefore, here we catalog genotype and environment (GxE) interactions, contrasting allele-specific regulatory activity between inflammatory cues. We report a cue-specific neuronal massively parallel reporter assay (MPRA) of 152 loci from genome-wide association studies (GWAS) of ten brain traits/disorders, empirically dissecting th

Altered neurodevelopmental trajectories of brain structure in Tourette syndrome and Chronic Tic Disorders.

Tourette syndrome (TS) is a neurodevelopmental disorder characterized by symptoms that emerge in childhood and often improve or even disappe … rbitofrontal cortex and putamen, alongside additional regions such as frontal and paralimbic areas. Genetic pleiotropy analyses identified overlap between TS/CTD-associated genetic effects on brain structure and neuroanatomical differences. Cross-disorder comparisons revealed correlations with ADHD and OCD and age-related patterns. These findings demonstrate altered …
