Evidence for overlapping genetic architecture between lifestyle factors and severe mental disorders with different patterns across diagnoses.

BACKGROUND: Severe mental disorders (SMDs) are associated with unhealthy lifestyle, contributing to increased risk of comorbid cardiovascular disease. Genetic factors influence both SMDs and lifestyle behaviours, but their genetic relationships remain unclear. Here, we aimed to unravel the shared genetic architecture of SMDs and lifestyle factors. Additionally, we assessed if genetic propensity to SMDs predicts body mass index (BMI) and lipids through lifestyle factors. METHODS: We analysed genome-wide data on major depression (MD) (N = 480,359), schizophrenia (SCZ) (N = 130,644), bipolar dis

Development and Validation of SNP Markers for Plant Genomic Studies.

One of the most useful techniques for studying genetic variations in plants is single-nucleotide polymorphisms (SNPs), which are useful in developing molecular markers for genetic research. In this chapter, the development and validation of SNP markers using targeted resequencing and in silico analysis of genomic resources for plant genomic research and breeding are included. Key steps include genomic DNA isolation, whole-genome resequencing, raw read processing, reference-based alignment, SNP detection, and stringent filtering to identify high-confidence variants. The importance of cross-plat…

A Recessive oca2 Mutation Underlies Albinism in Xiphophorus fish.

Oculocutaneous albinism (OCA) is a group of genetic disorders characterized by impaired melanin production, leading to reduced pigmentation in the skin, hair, and eyes. Xiphophorus , a genus of small freshwater fish, has been a pivotal model organism in pigmentation disorder research, providing key findings in the genetic pathways governing physiological and pathological pigment cell biology. Leveraging the well-established research framework provided by Xiphophorus , we have identified a spontaneously occurring albinism phenotype in swordtail fish Xiphophorus hellerii . Genetic mapping of alb

Mouse Models of Congenital Kidney Anomalies.

Congenital anomalies of the kidney and urinary tract (CAKUT) are common birth defects, which cause the majority of chronic kidney diseases in children. CAKUT covers a wide range of malformations that derive from deficiencies in embryonic kidney and lower urinary tract development, including renal aplasia, hypodysplasia, hypoplasia, ectopia, and different forms of ureter abnormalities. The majority of the genetic causes of CAKUT remain unknown. Research on mutant mice has identified multiple genes that critically regulate renal differentiation. The data generated from this research have served

Drosophila Model in Cancer: An Introduction.

Cancer is a cumulative manifestation of several complicated disease states that affect multiple organs. Over the last few decades, the fruit fly Drosophila melanogaster, has become a successful model for studying human cancers. The genetic simplicity and vast arsenal of genetic tools available in Drosophila provides a unique opportunity to address questions regarding cancer initiation and progression that would be extremely challenging in other model systems. In this chapter we provide a historical overview of Drosophila as a model organism for cancer research, summarize the multitude of genet

Pharmacogenomics of mood stabilizers in the treatment of bipolar disorder.

Bipolar disorder (BD) is a chronic and often severe psychiatric illness characterized by manic and depressive episodes. Among the most effective treatments, mood stabilizers represent the keystone in acute mania, depression, and maintenance treatment of BD. However, treatment response is a highly heterogeneous trait, thus emphasizing the need for a structured informational framework of phenotypic and genetic predictors. In this paper, we present the current state of pharmacogenomic research on long-term treatment in BD, specifically focusing on mood stabilizers. While the results provided so f

Generalist genes and learning disabilities.

The authors reviewed recent quantitative genetic research on learning disabilities that led to the conclusion that genetic diagnoses differ from traditional diagnoses in that the effects of relevant genes are largely general rather than specific. This research suggests that most genes associated with common learning disabilities--language impairment, reading disability, and mathematics disability--are generalists in 3 ways. First, genes that affect common learning disabilities are largely the same genes responsible for normal variation in learning abilities. Second, genes that affect any aspec…

Causal Association Between Primary Biliary Cholangitis and Osteoporosis: A Mendelian Randomization Study.

BACKGROUND: As the prevalence of osteoporotic fractures increases, impacting the health of the aging population significantly, understanding the genetic link between chronic diseases such as primary biliary cholangitis (PBC) and osteoporosis (OP) is crucial. Despite existing research, the direct genetic relationship between these conditions remains unclear. MATERIALS AND METHODS: This study used a two-sample Mendelian randomization approach, drawing on the largest available genome-wide association studies. Instrumental variables were selected based on single nucleotide polymorphisms to explor

Role of Genetics in Early-Onset Cardiovascular Disease.

Genetics increasingly comes to the front with early-onset cardiovascular disease (CVD) since researchers investigate the complex interplay of hereditary factors that promote an early manifestation of the disease. CVD is one of the most general causes of morbidity and mortality worldwide, presenting unique challenges when it arises in younger populations many times due to genetic predispositions. The various etiologies in the pathogenesis of early-onset CVD involve genetic factors, including the monogenic disorders of familial hypercholesterolemia (FH) and hypertrophic cardiomyopathy (HCM) of t

Families dealing with the uncertainty of genetic disorders: the case of Neurofibromatosis Type 1.

Some scholars contend that genetic medicine is transforming the experience of illness and the social category of the family - bringing future risks into the present, and potentially strengthening familial biological bonds in light of these shared genetic risks. However, research has shown that genetic information is interpreted and acted upon through a rich repertoire of adaptable social, cultural and familial factors which pre-exist and interact with biomedical knowledge. This paper reports research into families living with Neurofibromatosis Type 1 (NF1), a highly uncertain condition the man

A possible genetic predisposition to suspected hypoxic-ischaemic encephalopathy.

Within the last decade, several studies have explored whether there might be a genetic component in hypoxic-ischaemic encephalopathy (HIE) that influences susceptibility to or outcomes following hypoxic-ischaemic injury. This review provides a comprehensive overview of the findings to date from published studies investigating the genetics of HIE. It also highlights some of the challenges faced by researchers, as well as recommendations for future research.

Paving the way while playing catch up: mitochondrial genetics in African ancestry primary open-angle glaucoma.

Glaucoma, the leading cause of irreversible blindness worldwide, disproportionately affects individuals of African descent. Specifically, previous research has indicated that primary open-angle glaucoma (POAG), the most common form of disease, is more prevalent, severe, early-onset, and rapidly-progressive in populations of African ancestry. Recent studies have identified genetic variations that may contribute to the greater burden of disease in this population. In particular, mitochondrial genetics has emerged as a profoundly influential factor in multiple neurodegenerative diseases, includin

Environmental exposure and cancer prevention in the Czech Republic.

Cancer risk is predominantly shaped by environmental exposures interacting with genetic susceptibility, yet modifiable non-genetic determinants remain insufficiently characterised. The human exposome framework promotes a comprehensive, life-course approach to studying chemical, biological, and physical exposures alongside socioeconomic and psychosocial factors, but it is underutilised in cancer research. Systematically mapping biomarkers of exposure to each hallmark of cancer could provide a powerful strategy to identify modifiable risk factors with translational relevance. Czechia, with its p

Reproductive decision-making in young female carriers of a BRCA mutation.

STUDY QUESTION: How do young women, who were identified as carrying a BRCA gene mutation before they had children, approach reproductive decision-making and what are their attitudes towards reproductive genetic testing? SUMMARY ANSWER: Reproductive decision-making within the context of cancer risk is complex and influenced by personal experiences of cancer. Younger women were not concerned with reproductive decision-making at the time of their genetic test; however, the impact on subsequent reproductive decision-making was considerable and left them with unanticipated dilemmas, such as having

A review of genetic factors underlying craniorachischisis and omphalocele: Inspired by a unique trisomy 18 case.

Very few cases of craniorachischisis (CRN) with concomitant omphalocele (OMP) in the setting of trisomy 18 are reported in literature. Solitary midline closure defects are estimated to be more prevalent in trisomy 18 compared to the general population. Neurulation defect comparisons include anencephaly 0-2% versus 0.0206%, spina bifida 1-3% versus 0.0350%, and encephalocele 0-2% versus 0.0082% [Parker et al. (2010); Birth Defects Research. Antagonistic pleiotropy as a widespread mechanism for the maintenance of polymorphic disease alleles.

BACKGROUND: Many serious diseases have a genetic basis which, from an evolutionary point of view, should have been selected against, resulting in very low frequencies. The remarkable sustained prevalence of a number of disease-associated alleles is therefore surprising. We believe that antagonistic pleiotropy, when multiple effects of a gene have opposing effects on fitness (e.g., sickle cell disease), may be more widespread than typically considered. We hypothesize that, rather than being an exception to the rule of genetic disorders, antagonistic pleiotropy may be common. METHODS: We survey

Diving into the zebrafish brain: exploring neuroscience frontiers with genetic tools, imaging techniques, and behavioral insights.

The zebrafish (Danio rerio) is increasingly used in neuroscience research. Zebrafish are relatively easy to maintain, and their high fecundity makes them suitable for high-throughput experiments. Their small, transparent embryos and larvae allow for easy microscopic imaging of the developing brain. Zebrafish also share a high degree of genetic similarity with humans, and are amenable to genetic manipulation techniques, such as gene knockdown, knockout, or knock-in, which allows researchers to study the role of specific genes relevant to human brain development, function, and disease. Zebrafish

Postnatal Development of the Mouse Enteric Nervous System.

Owing to over three decades of research, we now have a good understanding of the genetic and molecular control of enteric nervous system (ENS) development during embryonic and prenatal stages. On the other hand, it has only just become clear that a substantial process of ENS maturation occurs after birth (Hao et al. 2013a). During postnatal stages, in addition to genetic influences, ENS development is also potentially affected by the external environment. Thus it is possible that manipulating certain environmental factors could help prevent or reduce motility disorders. However the genetic and

Progress in our understanding of the pathogenesis of ankylosing spondylitis.

AS is a common rheumatic condition characterized by inflammation and new bone formation. The pathogenesis of AS is likely multifactorial and has not been fully elucidated to date. A major genetic role has been demonstrated. The strongest genetic association is with HLA B27. Numerous other associated genetic polymorphisms have been identified, including those affecting the type 17 immune pathway, although the precise link between genetics and pathogenesis remains unexplained. Several immunological alterations, together with recent therapeutic advances, support a central role for IL-23- and IL-1
