An organometallic hybrid antibiotic of metronidazole with a Gold(I) N-Heterocyclic Carbene overcomes metronidazole resistance in Clostridioides difficile.

Antimicrobial resistance (AMR) has been emerging as a major global health threat and calls for the development of novel drug candidates. Metal complexes have been demonstrating high efficiency as antibacterial agents that differ substantially from the established types of antibiotics in their chemical structures and their mechanism of action. One strategy to exploit this potential is the design of metal-based hybrid organometallics that consist of an established antibiotic and a metal-based warhead that contributes an additional mechanism of action different from that of the parent antibiotic.

Complexes of metals other than platinum as antitumour agents.

The earliest reports on the therapeutic use of metals or metal-containing compounds in cancer and leukemia date from the sixteenth and nineteenth centuries. They were forgotten until the 1960s, when the anti-tumour activity of the inorganic complex cis-diammine-dichloroplatinum(II) (cisplatin) was discovered. This led to the development of other types of non-organic cytostatic drugs. Cisplatin has developed into one of the most frequently used and most effective cytostatic drugs for the treatment of solid carcinomas. Numerous other metal compounds containing platinum, other platinum metals, an

Sustainable Primary Cell Banking for Topical Compound Cytotoxicity Assays: Protocol Validation on Novel Biocides and Antifungals for Optimized Burn Wound Care.

Thorough biological safety testing of topical therapeutic compounds and antimicrobials is a critical prerequisite for appropriate cutaneous wound care. Increasing pathogen resistance rates to traditional antibiotics and antifungals are driving the development and registration of novel chemical entities. Although they are notably useful for animal testing reduction, the gold standard in vitro cytotoxicity assays in continuous cell lines (HaCaT keratinocytes, 3T3 fibroblasts) may be discussed from a translational relevance standpoint. The aim of this study was thus to establish and validate a su

Real-time, label-free monitoring of cell viability based on cell adhesion measurements with an atomic force microscope.

BACKGROUND: The adhesion of cells to an oscillating cantilever sensitively influences the oscillation amplitude at a given frequency. Even early stages of cytotoxicity cause a change in the viscosity of the cell membrane and morphology, both affecting their adhesion to the cantilever. We present a generally applicable method for real-time, label free monitoring and fast-screening technique to assess early stages of cytotoxicity recorded in terms of loss of cell adhesion. RESULTS: We present data taken from gold nanoparticles of different sizes and surface coatings as well as some reference su

A visual assay and spectrophotometric determination of LLM-105 explosive using detection of gold nanoparticle aggregation at two pH values.

We report a simple, rapid, and sensitive assay for visual and spectrophotometric detection of the 2,6-diamino-3,5-dinitropyrazine-1-oxide (LLM-105) explosive. The assay is based on different interactions between LLM-105 and gold nanoparticle (AuNP) dispersions at two pH values, leading to the formation of dispersed or aggregated AuNPs. Two AuNP dispersions at two pH values were applied to recognize and detect LLM-105 instead of traditional AuNP dispersion under an aptotic pH to improve the anti-interference ability. The developed assay showed excellent sensitivity with a detection limit of 3 n

Inelastic electron tunneling spectroscopy of alkane monolayers with dissimilar attachment chemistry to gold.

We compare the low-temperature electron transport properties of alkyl monolayers which utilize different attachment strategies to gold. Inelastic electron tunneling spectroscopy (IETS) and current-voltage analysis were performed on molecular junctions incorporating alkyl-dithiocarbamate and alkanethiolate self-assembled monolayers of similar length. Alkyl-dithiocarbamate monolayers were formed by the condensation of dioctylamine or didecylamine with carbon disulfide in anhydrous ethanol and compared to alkanethiolate SAMs of 1-decanethiol and 1-dodecanethiol, respectively. The electron transpo

Spectroscopic investigations of Bourdichon miniatures: masterpieces of light and color.

Metallic bismuth, pyrite (FeS2), and specular hematite (Fe2O3) were found to have been used extensively on two miniatures taken from the Book of Hours of Louis XII by Jean Bourdichon, painter at the Royal Court of France between the end of the 15th and the beginning of the 16th centuries. These unusual materials were identified, together with thirteen other more traditional pigments and dyes, by Raman microscopy and X-ray fluorescence. Pyrite was found in many areas, suggesting that it may have been deliberately added as a pigment. The luminary quality of both miniatures was enhanced by the ex

Modular Orthogonal Dual-Channel Gold Nanosensor Platform for Ratiometric Imaging of Tumor-Associated Macrophage M1/M2 Polarization.

The phenotypic balance between M1 and M2 tumor-associated macrophages (MΦs) governs tumor immune homeostasis and immunotherapeutic outcomes. However, in situ visualization of this polarization equilibrium remains challenging. Conventional histological and cytological approaches only afford static ex vivo phenotyping, while single-biomarker-responsive fluorescent probes lack the capacity to simultaneously detect M1 and M2. Herein, we fabricate a modular dual-channel ratiometric nanosensor for simultaneous imaging and assessment of M1/M2 MΦ polarization states. Two spectrally orthogonal fluoroph

Rational design of gold nanoparticle-based artificial phosphatase with high activity comparable to natural phosphatase.

An artificial phosphatase, GoldAP, is rationally designed by installation of a metal-binding site onto a natural protein fragment to form a well-defined active centre on gold nanoparticles. GoldAP efficiently catalyses the hydrolysis of p-nitrophenyl phosphate in seconds with a kcat that is higher than that of Escherichia coli alkaline phosphatase.

Hydrogen-Bond-Engineered Photoactivated Au10 Clusterzymes for Amplex Red Specificity and Highly Efficient Fluorescence Sensing.

Despite the promise of gold clusterzymes in biosensing, their applications are predominantly confined to peroxidase (POD)-mimicking systems reliant on unstable, cytotoxic hydrogen peroxide (H2O2) under acidic conditions. Therefore, constructing ultrasensitive fluorescence platforms independent of POD-H2O2 at neutral pH remains challenging. Herein, we propose a highly active, substrate-specific photoactivated oxidase-like Au10 clusterzymes functionalized with a thymine derivative (ATT) for sensitive and efficient fluorescence sensing using amplex red (AR) as the substrate.
