Ultramafic specialist lichens persist through flexible symbioses structured by climate-driven filtering and modulated by substrate effects.

Ultramafic substrates impose severe edaphic stress characterised by metal toxicity, nutrient imbalance, and extreme microclimatic conditions, creating spatially fragmented habitats that can shape evolutionary and ecological dynamics of associated organisms. Together with climatic variation, such factors may also influence the structure of holobiont communities of lichenized fungi. Here we investigated the ultramafic specialist lichen Solenopsora liparina across its entire range. We contextualized its symbiotic associations using calcicolous congeners (S. candicans and S. cesatii), as ecologica

Circular RNAs in virus-induced cancers: From mechanism to clinical implications.

Viruses, during their lifecycle, are continuously dependent on host machinery for their survival. Sometimes this dependency leads to uncontrolled cell proliferation in the host, accounting for 12% of virus-associated cancers. Since early times, viruses have been known to produce proteins and dysregulate host proteins, but what is new to the field is the addition of a circular regulatory RNA called "circRNA". circular RNAs, due to their structure-associated stability and specificity for various cancers, are key RNA molecules for researchers seeking to understand the complex outcomes of virus-as

Isolation, Characterization, and Pathogenicity of Two Pseudomonas syringae Pathovars from Populus trichocarpa Seeds.

Pseudomonas syringae is a ubiquitous plant pathogen, infecting both woody and herbaceous plants and resulting in devastating agricultural crop losses. Characterized by a remarkable specificity for plant hosts, P. syringae pathovars utilize a number of virulence factors including the type III secretion system and effector proteins to elicit disease in a particular host species. Here, two Pseudomonas syringae strains were isolated from diseased Populustrichocarpa seeds. The pathovars were capable of inhibiting poplar seed germination and were selective for the Populus genus. Sequencing of the ne

Towards minimizing second-generation mis-identification of Blastocystis.

At least 1-2% of DNA sequences annotated as Blastocystis in GenBank represent organisms other than Blastocystis or sequence artefacts. As well as being biologically incorrect, such practice can lead to overestimates of genetic diversity, underestimated host specificity, and incorrect classification of samples tested for Blastocystis using DNA-based methods.

Genetic crosses within and between species of Cryptosporidium.

Parasites and their hosts are engaged in reciprocal coevolution that balances competing mechanisms of virulence, resistance, and evasion. This often leads to host specificity, but genomic reassortment between different strains can enable parasites to jump host barriers and conquer new niches. In the apicomplexan parasite Cryptosporidium, genetic exchange has been hypothesized to play a prominent role in adaptation to humans. The sexual lifecycle of the parasite provides a potential mechanism for such exchange; however, the boundaries of Cryptosporidium sex are currently undefined. To explore t…

Prolyl Oligopeptidase from the Blood Fluke Schistosoma mansoni: From Functional Analysis to Anti-schistosomal Inhibitors.

BACKGROUND: Blood flukes of the genus Schistosoma cause schistosomiasis, a parasitic disease that infects over 240 million people worldwide, and for which there is a need to identify new targets for chemotherapeutic interventions. Our research is focused on Schistosoma mansoni prolyl oligopeptidase (SmPOP) from the serine peptidase family S9, which has not been investigated in detail in trematodes. METHODOLOGY/PRINCIPAL FINDINGS: We demonstrate that SmPOP is expressed in adult worms and schistosomula in an enzymatically active form. By immunofluorescence microscopy, SmPOP is localized in the

The triple threat of cryptococcosis: it's the body site, the strain, and/or the host.

Cryptococcosis is the leading invasive fungal infection in the world today. Over the past century, the causative agents, Cryptococcus neoformans and Cryptococcus gattii, have risen from the status of medical curiosities to common but life-threatening central nervous system pathogens. In an elegant experimental pathobiology study of these two organisms carried out by Ngamskulrungroj et al., there are three matters that merit further discussion. First is the question of whether there is a variable specific pathobiology for each yeast strain. Does it make biological and clinical sense to designat

Temporal dynamics of the S. thermophilus phage population in a yogurt production facility.

Starter cultures are essential for milk fermentation, but phage infections can disrupt production, compromising product quality and causing economic losses. In this study, we investigated the dynamics, diversity, and persistence of 25 S. thermophilus phages isolated over a 15-year period from a large yogurt manufacturing plant. Phages were classified into Moineauvirus (16) and Brussowvirus (9), comprising ten distinct restriction profiles. Host range, genomic features, and resistance to thermal and chemical treatments were analyzed to identify factors underlying their long-term persistence. No

Targeting nontuberculous mycobacteria with phages: optimization of in vitro host range assays.

The clinical implementation of mycobacteriophages is hampered by the lack of standardized in vitro methods and inconsistent host range results, limiting reliable assessment of their therapeutic potential. Nontuberculous mycobacterial (NTM) infections are frequently difficult to treat, emphasizing the urgent need for alternative or adjunctive strategies to antibiotics. To facilitate reproducible translational research, a detailed and systematic understanding of in vitro assay conditions is essential. Here, we comprehensively evaluated commonly used liquid and solid media for their suitability i

Calcium-binding motif-mediated binding of redundant calcium offers a chimeolysin enhanced bactericidal activity and extended host range under physiological conditions.

OBJECTIVES: Calcium-binding motifs are shared by multiple bacteriophage lysins; however, the influence of calcium on the enzymatic activity and host range of these enzymes is still not understood. To address this, ClyF, a chimeric lysin with a putative calcium-binding motif, was used as a model for in vitro and in vivo investigations. METHODS: The concentration of calcium bound to ClyF was determined by atomic absorption spectrometry. The influence of calcium on the structure, activity and host range of ClyF was assessed by circular dichroism and time-kill assays. The bactericidal activity of

Multilayered Networks of SalmoNet2 Enable Strain Comparisons of the Salmonella Genus on a Molecular Level.

Serovars of the genus Salmonella primarily evolved as gastrointestinal pathogens in a wide range of hosts. Some serotypes later evolved further, adopting a more invasive lifestyle in a narrower host range associated with systemic infections. A system-level knowledge of these pathogens could identify the complex adaptations associated with the evolution of serovars with distinct pathogenicity, host range, and risk to human health. This promises to aid the design of interventions and serve as a knowledge base in the Salmonella research community. Here, we present SalmoNet2, a major update to Sal

Bacteriophage Sf6 host range mutant that infects Shigella flexneri serotype 2a2 strains.

Shigella flexneri serotype 2a2 (II:9;10) is the most prevalent strain in causing bacillary dysentery in developing countries. Chemical modifications such as glucosylation, O-acetylation, and phosphoethanolamine modifications of lipopolysaccharide (LPS) O antigen (Oag) contribute to the emergence of various serotypes. Sf6 is a Shigella-specific bacteriophage that infects only a limited range of S. flexneri serotypes [X, Y]. LPS Oag is the primary receptor for bacteriophage Sf6 where it uses its tailspike protein (TSP) in binding and hydrolysing LPS Oags. Sf6TSP has recently been shown to be cap

A review on Trichinella infection in South America.

Trichinella spp. causes human trichinellosis by means of the consumption of raw or inadequately treated meat from domestic or game animals. In the Americas, as well as in other continents, Trichinella infection is a health issue for humans and has a negative impact on the pork meat market, generated by people's fear of becoming infected with the parasite. The distribution of human cases and the sources of this disease in humans and animals were analysed in this report, which summarizes the information available regarding Trichinella infection in animals and humans in South America. Within Sout

Divergent Effects of Schistosoma haematobium Exposure on Intermediate-Host Snail Species Bulinus nasutus and Bulinus globosus from Coastal Kenya.

AbstractSchistosoma haematobium infection causes urogenital schistosomiasis, a chronic inflammatory disease that is highly prevalent in many parts of sub-Saharan Africa. Bulinid snails are the obligate intermediate hosts in the transmission of this parasite. In the present study, Bulinus globosus and Bulinus nasutus snails from coastal Kenya were raised in the laboratory and exposed to miracidia derived from sympatric S. haematobium specimens to assess the species-specific impact of parasite contact and infection. The snails' subsequent patterns of survival, cercarial shedding, and reproductio
