Microporous Bio-orthogonally Annealed Particle Hydrogels for Tissue Engineering and Regenerative Medicine.

Microporous annealed particle (MAP) hydrogels are an emerging class of biomaterials with the potential to improve outcomes in tissue repair and regeneration. Here, a new MAP hydrogel platform comprising poly(ethylene) glycol (PEG) hydrogel microparticles that are annealed in situ using bio-orthogonal tetrazine click chemistry is reported (i.e., TzMAP hydrogels). Briefly, clickable PEG-peptide hydrogel microparticles with extracellular matrix mimetic peptides to permit cell adhesion and enzymatic degradation were fabricated via submerged electrospraying and stoichiometrically controlled thiol-n…

Stiff micelle-crosslinked hyaluronate hydrogels with low swelling for potential cartilage repair.

Pluronic F127 diacrylate (F127DA) nano-micelle crosslinked methacrylated hyaluronic acid (MeHA) hydrogels (NMgels) with strong compressive properties have been demonstrated in our previous study. The current study further focuses on how the F127DA micelles and long-term swelling affect the mechanical performance of hydrogels from the view of in vitro/in vivo applications. Co-contributions of the F127DA micelles and MeHA to the compression performance are first investigated through mechanical analysis and cyclic loading/unloading tests before and after swelling. The optimized NMgel with F127DA …

Photothermally triggered on-demand insulin release from reduced graphene oxide modified hydrogels.

On-demand delivery of therapeutics plays an essential role in simplifying and improving patient care. The high loading capacity of reduced graphene oxide (rGO) for drugs has made this matrix of particular interest for its hybridization with therapeutics. In this work, we describe the formulation of rGO impregnated poly(ethylene glycol) dimethacrylate based hydrogels (PEGDMA-rGO) and their efficient loading with insulin. Near-infrared (NIR) light induced heating of the PEGDMA-rGO hydrogels allows for highly efficient insulin release. Most importantly, we validate that the NIR irradiation of the…

Development of poly(ethylene glycol) hydrogels for salivary gland tissue engineering applications.

More than 40,000 patients are diagnosed with head and neck cancers annually in the United States with the vast majority receiving radiation therapy. Salivary glands are irreparably damaged by radiation therapy resulting in xerostomia, which severely affects patient quality of life. Cell-based therapies have shown some promise in mouse models of radiation-induced xerostomia, but they suffer from insufficient and inconsistent gland regeneration and accompanying secretory function. To aid in the development of regenerative therapies, poly(ethylene glycol) hydrogels were investigated for the encap…

TGFβ functionalized starPEG-heparin hydrogels modulate human dermal fibroblast growth and differentiation.

UNLABELLED: Hydrogels are promising biomaterials that can adapt easily to complex tissue entities. Furthermore, chemical modifications enable these hydrogels to become an instructive biomaterial to a variety of cell types. Human dermal fibroblasts play a pivotal role during wound healing, especially for the synthesis of novel dermal tissue replacing the primary fibrin clot. Thus, the control of growth and differentiation of dermal fibroblasts is important to modulate wound healing. In here, we utilized a versatile starPEG-heparin hydrogel platform that can be independently adjusted with respec…

Enzymatic biodegradation of hydrogels for protein delivery targeted to the small intestine.

Multiresponsive poly(methacrylic acid-co-N-vinylpyrrolidone) hydrogels were synthesized with biodegradable oligopeptide crosslinks. The oligopeptide crosslinks were incorporated using EDC-NHS zero-length links between the carboxylic acid groups of the polymer and free primary amines on the peptide. The reaction of the peptide was confirmed by primary amine assay and IR spectroscopy. The microgels exhibited pH-responsive swelling as well as enzyme-catalyzed degradation targeted by trypsin present in the small intestine, as demonstrated upon incubation with gastrointestinal fluids from rats. Rel…

Salicylic acid-based pH-sensitive hydrogels as potential oral insulin delivery systems.

Salicylic acid (SA)-based physically crosslinked pH-sensitive hydrogels were developed for oral insulin delivery using various ratios of salicylate-based poly(anhydride-ester) and poly(acrylic acid) (PAA). Pore size, swelling behavior, insulin loading, insulin and SA release rates of the gels were varied by changing PAA concentration. About 50% of insulin was incorporated within all of the hydrogels, with about 4-8% of insulin released in acidic conditions (pH 1.2) over 2 h. In pH simulating the intestine (pH 6.8), 90% of the insulin and 70% of SA were released within 24 h from the hydrogel sy…

Ultrashort peptide nanofibrous hydrogels for the acceleration of healing of burn wounds.

There is an unmet clinical need for wound dressings to treat partial thickness burns that damage the epidermis and dermis. An ideal dressing needs to prevent infection, maintain skin hydration to facilitate debridement of the necrotic tissue, and provide cues to enhance tissue regeneration. We developed a class of 'smart' peptide hydrogels, which fulfill these criteria. Our ultrashort aliphatic peptides have an innate tendency to self-assemble into helical fibers, forming biomimetic hydrogel scaffolds which are non-immunogenic and non-cytotoxic. These nanofibrous hydrogels accelerated wound cl…

Viscoelastic and mechanical properties of hyaluronan films and hydrogels modified by carbodiimide.

This study investigated an effect of different ways of the preparation of insoluble hyaluronan material on its mechanical and viscoelastic properties. Hyaluronan (NaHy) of molecular weight Mw=500,000 g mol(-1) was modified with N-(3-dimethylaminopropyl-N'-ethylcarbodiimide hydrochloride (EDC) and N-hydroxysuccinimide (NHS), to be able absorb liquid without changing its mechanical properties. The modified, water insoluble NaHy materials were prepared in different geometry; as modified films and modified cylinders with exact dimensions. The occurrence of modification was confirmed by FT-IR (Four

Clotrimazole-loaded nanostructured lipid carrier hydrogels: thermal analysis and in vitro studies.

The aim of the present work is to design a new formulation containing clotrimazole (CTZ) loaded into nanostructured lipid carriers (NLC) for the treatment of fungal vaginal infections. In order to obtain formulations with suitable viscosity for mucosal application, NLC containing CTZ produced by the ultrasonication method were viscosized by the addition of poloxamer P407 in the NLC dispersion (CTZ-NLC-gel). These systems exhibit well-known thermogelling properties. The rheological characterization of the CTZ-NLC hydrogel using a controlled stress rheometer evidenced that the presence of NLC or

Dual functionality of antimicrobial and antifouling of poly(N-hydroxyethylacrylamide)/salicylate hydrogels.

The emergence and reemergence of microbial infection demand an urgent response to develop effective biomaterials that prevent biofilm formation and associated bacterial infection. In this work, we have synthesized and characterized hybrid poly(N-hydroxyethylacrylamide) (polyHEAA)/salicylate (SA) hydrogels with integrated antifouling and antimicrobial capacities. The antifouling efficacy of polyHEAA hydrogels was examined via exposure to proteins, cells, and bacteria, while the antimicrobial activity of SA-treated polyHEAA hydrogels was investigated against both gram-negative Escherichia coli R…

Dextran-based in situ cross-linked injectable hydrogels to prevent peritoneal adhesions.

Peritoneal adhesions are serious sequelae of surgery, and can cause significant morbidity and/or mortality due to pain, infertility, and bowel obstruction. We have designed and synthesized novel dextran (DX)-based injectable hydrogels for adhesion prevention, which are formed by mixing hydrazide-modified carboxymethyldextran (CMDX-ADH) with aldehyde-modified DX (DX-CHO) or carboxymethylcellulose (CMC-CHO). At high polymer concentrations, hydrogels formed very quickly upon mixing, e.g. 5% CMDX-ADH with 6% DX-CHO (=CMDX-DX; 1.8 s) and 5% CMDX-ADH with 6% CMC-CHO (=CMDX-CMC; 5.8 s). CMDX-DX shran…

Robust Hydrogel Actuators Functioning in Multi-Environments Enabled by Thermo-Responsive Polymer Nanoparticle Coatings on Hydrogel Surfaces.

Hydrogel actuators with anisotropic structures exhibit reversible responsiveness upon the trigger of various external stimuli, rendering them promising for applications in many fields including artificial muscles and soft robotics. However, their effective operation across multiple environments remains a persistent challenge, even for widely studied thermo-responsive polymers like poly(N-isopropyl acrylamide) (PNIPAm). Current attempts to address this issue are hindered by complex synthetic procedures or specific substrates. This study introduces a straightforward methodology to grow a thin, d

Preclustering Gelatin for Faster-Forming Injectable Hydrogels: A Strategy for Fabricating 3D Hydrogel Scaffolds with Improved Cell Dispersion.

Gelatin-based injectable hydrogels capable of encapsulating cells are pivotal in tissue engineering because they can conform to any geometry and exhibit physical properties similar to those of living tissues. However, the slow gelation process observed in these cell-encapsulating hydrogels often causes an uneven dispersion of cells. This study proposes an approach for achieving fast gelation of unmodified gelatin under physiological conditions through gelatin preclustering. By using tetrafunctional succinimidyl-terminated poly(ethylene glycol) as a clustering agent, the gelation process is suc…

Injectable Shear Thinning Hydrogels from Functionalized Silk Proteins for 3D Mammalian Cell Culture.

Silk fibroin hydrogels are a promising platform for tissue engineering due to their biocompatibility and tunability, yet efficient cell recovery from gels remains a challenge. This work synthesizes reversible hydrogels by electrostatically-crosslinking carboxylated silk with linear and branched polyamine. Solvent choice (water, saline, or medium) and protein concentration impacted gel stiffness, mesh size, degradability, and function as an in vitro culture substrate. Hydrogels flowed as a liquid and recovered their viscoelastic properties when cycled under shear. Unlike enzymatically-crosslink…

Highly Deformable Organic Heterojunctions Empower Piezoelectric-Conductive Hydrogels for Functional Volumetric Muscle Regeneration.

Volumetric muscle loss (VML) results in irreversible muscle impairment, necessitating tissue-matched electrostimulation scaffolds for clinical repair. Here, we report a biointegrated hydrogel that leverages a piezoelectric-conductive organic heterojunction to deliver self-powered electrical cues directly to the injury site. The heterojunction comprises a piezoelectric metal-organic framework (MOF) coated with conductive polypyrrole (PPy), forming a stable p-n organic heterojunction with an intrinsic built-in electric field. Unlike inorganic piezoelectrics, the organic MOF-PPy features an ultra
