Varicella at "Casa Garrahan", 2008-2013: Assessment of postexposure prophylaxis measures.

INTRODUCTION: Casa Garrahan (CG) accommodates children with complex conditions referred nationwide; these children are seen in children's hospitals located in the Autonomous City of Buenos Aires. Varicella is a highly-contagious disease, with attack rates of up to 90% among susceptible individuals. In closed communities, the implementation of outbreak control measures is critical. OBJECTIVES: To describe the characteristics of children exposed to varicella at CG, the implemented prophylaxis measures and their effectiveness. METHODS: Prospective, cohort study. Children exposed to varicella at

Immunization with a multisubunit vaccine induces immunological memory and cross-protective immunity against heterologous Chlamydia serovars.

We previously reported that intramuscular (IM) and intranasal (IN) immunization with a recombinant Vibrio cholerae (rVCG)-based multiepitope subunit vaccine (rVCG-MECA) elicited robust long-lasting, cellular, and humoral immune effectors that protected mice against transcervical (TC) challenge infection with the homologous C. trachomatis (CT) serovar D. In this study, we evaluated the ability of this vaccine to confer cross-protection against primary and secondary infection with the heterologous CT serovars F and J and induce cross-reactive immunological memory following reinfection. Thus, gro

A case of vaccine-associated myocarditis following pneumococcal immunization leading to acute mitral regurgitation.

Vaccine-associated myocarditis (VAM) is a rare entity but can result in potentially serious sequelae if left untreated. However, the mechanisms of the complications of VAM and its treatment remain unclear. Herein, we report the first case of VAM related to pneumococcal immunization, presenting as a local and systemic inflammatory reaction, in which the patient developed significant secondary mitral regurgitation, resulting in acute heart failure. Finally, the patient recovered completely following corticosteroid treatment. This case highlights the value of cardiac magnetic resonance and the pi

[Meningococcal vaccines. Global epidemiological situation and strategies for prevention by vaccination].

N. meningitidis is a major cause of meningitis and septicemia and a major public health problem in many countries. The disease, that can be fulminant, has a high mortality and may cause serious sequelae, even in cases of apparently optimal medical treatment. Chemoprophylaxis may prevent secondary cases among those in close contact with the ill, but, since secondary cases represent only 1%-2% of all meningococcal disease, chemoprophylaxis has a small impact when fighting most of endemic and epidemic forms. Given that al least 5% -15% of children and young adults are carriers, the fight against

Porphyrin metabolism in hepatitis C infection.

Hepatitis C virus has been implicated as a major precipitating factor in porphyria cutanea tarda (PCT). To determine whether hepatitis C infection alone is sufficient to induce PCT, we screened two groups of patients with hepatitis C infection. The first group comprised women who had become HCV positive secondary to immunization with anti-D immunoglobulin (group 1). Group 2 included males and females who were HCV positive but HIV negative secondary to intravenous drug abuse. Though both groups had very abnormal liver function tests, we found no significant abnormalities in porphyrin metabolism

WHO global vaccine safety multi-country collaboration project on safety in pregnancy: Assessing the level of diagnostic certainty using standardized case definitions for perinatal and neonatal outcomes and maternal immunization.

Standardized case definitions strengthen post-marketing safety surveillance of new vaccines by improving generated data, interpretation and comparability across surveillance systems. The Global Alignment of Immunization Safety Assessment in Pregnancy (GAIA) project developed standardized case definitions for 21 key obstetric and neonatal terms following the Brighton Collaboration (BC) methodology. In this prospective cohort study, we assessed the applicability of GAIA definitions for maternal immunization exposure and for low birth weight (LBW), preterm birth, small for gestational age (SGA),

Unadjuvanted antenatal pertussis OMV vaccination drives enhanced and durable humoral immunity in offspring.

BACKGROUND: Antenatal-immunization is the primary strategy to protect neonates from pertussis. However, the global shift to acellular vaccines (aP) has raised concerns regarding the duration of transferred immunity, largely due to the Th2-skewed profile they induce. This study evaluates the immunogenicity and protective efficacy transferred to offspring by a novel, unadjuvanted Bordetella pertussis Outer Membrane Vesicle-based vaccine (OMVPreg), which induces a mixed Th1/Th2/Th17 response, compared to the commercial aP vaccine (aPPreg). METHODS: Female mice received a primary two-dose series

Persistence of immunity and impact of third dose of inactivated COVID-19 vaccine against emerging variants.

This is a comprehensive report on immunogenicity of COVAXIN® booster dose against ancestral and Variants of Concern (VOCs) up to 12 months. It is well known that neutralizing antibodies induced by COVID-19 vaccines wane within 6 months of vaccination leading to questions on the effectiveness of two-dose vaccination against breakthrough infections. Therefore, we assessed the persistence of immunogenicity up to 6 months after a two or three-dose with BBV152 and the safety of a booster dose in an ongoing phase 2, double-blind, randomized controlled trial (ClinicalTrials.gov: NCT04471519). A Prime/Boost Vaccine Regimen Alters the Rectal Microbiome and Impacts Immune Responses and Viremia Control Post-Simian Immunodeficiency Virus Infection in Male and Female Rhesus Macaques.

An efficacious human immunodeficiency virus (HIV) vaccine will likely require induction of both mucosal and systemic immune responses. We compared the immunogenicity and protective efficacy of two mucosal/systemic vaccine regimens and investigated their effects on the rectal microbiome. Rhesus macaques were primed twice mucosally with replication-competent adenovirus type 5 host range mutant (Ad5hr)-simian immunodeficiency virus (SIV) recombinants and boosted twice intramuscularly with ALVAC-SIV recombinant plus SIV gp120 protein or with DNA for SIV genes and rhesus interleukin-12 plus SIV gp1

The immunization delivery effectiveness assessment score: a better immunization measure?

BACKGROUND: Childhood immunization measures, such as the Health Employer Data Information Set (HEDIS) or the National Immunization Survey, assess the percentage of children up-to-date for a specified series of vaccinations. In particular, the HEDIS assesses immunization delivery to children enrolled in managed care organizations (MCO). Such measures do not assess the timeliness of immunization delivery with reference to recommended age standards. To achieve maximal protection against vaccine-preventable diseases, children should receive all immunizations within recommended age intervals-fully

Heterologous DNA prime-protein boost immunization with RecA and FliD offers cross-clade protection against leptospiral infection.

The emergence of >300 serovars of Leptospira confounded the use of generalized bacterin, the whole cell lysate, as vaccines to control leptospirosis. Because of substantial genetic and geographic heterogeneity among circulating serovars, one vaccine strain per serovar cannot be efficacious against all the serovars. We have performed heterologous DNA prime-protein boost vaccination challenge studies in hamsters using in vivo expressed, leptospiral recombinase A (RecA) and flagellar hook associated protein (FliD). We prepared the monovalent recombinant protein, plasmid DNA, and DNA prime protein

Rationale for use of Tdap booster vaccines for adolescent immunization: overview of efficacy, safety, and clinical use.

Pertussis is one of the only vaccine-preventable diseases with increasing incidence in the United States. The incidence of pertussis infection in adolescents is a growing concern; it can be a severe disease that may lead to significant morbidity and mortality when transmitted to susceptible populations (eg, infants). Experts have conceded that booster vaccination of adolescents may considerably decrease the incidence of pertussis infections in previously immunized, partially immunized, and nonimmunized populations. Studies in 2 tetanus, diphtheria, and acellular pertussis (Tdap) booster vaccin

Single-Cell Sequencing Reveals Varying Cell-Mediated Immunity Profiles in Individuals With Distinct Antibody Responses Following Ad5-nCoV Booster.

BACKGROUND: Vaccination is critical for controlling infectious diseases; however, assessing efficacy solely through neutralizing antibody titers oversimplifies immune protection. The cellular and molecular mechanisms driving variable vaccine-induced immune responses remain underexplored, limiting comprehensive vaccine evaluation. METHODS: Using single-cell sequencing, we profiled and compared cellular dynamic, transcriptomic profiles, the immune repertoire, and cellular communication in vaccine recipients with high and low antibody titers after Ad5-nCoV booster vaccination, using peripheral b

Vaccination with monovalent Leptospira interrogans serovar Copenhageni and its implication for the diagnosis of natural infection in dogs.

Using microscopic agglutination test (MAT), a single titre ≥ 1/800 in dogs unvaccinated against leptospirosis is considered suggestive of infection. The emergence of canine leptospirosis in Sydney, Australia, led to considerable vaccination uptake with monovalent Leptospira interrogans serovar Copenhageni, yet the humoral response post-vaccination has not been described. Aims were to characterise magnitude and duration of humoral response post-vaccination with serovar Copenhageni (Protech®C2i, Boehringer Ingelheim, Australia) in healthy dogs to assess potential effects on diagnosis. Serial MAT

Immunisation of chickens with live Salmonella vaccines - Role of booster vaccination.

It is accepted that booster vaccinations of chickens with live Salmonella vaccines are essential part of vaccinations schemes to induce an effective adaptive immune response. As manufacturer of registered live Salmonella vaccines recommend different times of booster the question raises whether the duration between the first and second immunisation might influence the protective effect against Salmonella exposure. Chickens were immunised with a live Salmonella Enteritidis vaccine on day 1 of age followed by a booster vaccination at different intervals (day 28, 35 or 42 of age) to study the effe
