The prognosis for relapsed Hodgkin lymphoma after allogeneic hematopoietic cell transplantation (HSCT) is poor, partly because of limited treatment options. Here we present a case of a Hodgkin lymphoma patient who relapsed after allogeneic HSCT but remains in complete remission (CR) at 38 months from the start of extended brentuximab vedotin (BV) dosing. A 33-year-old man with refractory and relapsed nodular sclerosis classical Hodgkin lymphoma who underwent previous treatments, including adriamycin, bleomycin, vinblastine, and dacarbazine (ABVD) ; seven combination regimens; and autologous HS

The calcineurin inhibitors (CNIs) remain the standard of care for maintenance immunosuppression following renal transplantation. CNIs have demonstrated their effectiveness in reducing acute cellular rejection; however, some evidence suggests that these compounds negatively affect native renal function and are associated with allograft injury in renal transplant recipients. CNIs have also been linked with hypertension, new-onset diabetes after transplantation, tremor, and thrombotic microangiopathy, which have significant consequences for long-term allograft function and patient health overall.

BACKGROUND: Rheumatoid arthritis (RA) is an autoimmune disease characterized by chronic joint inflammation, which if left untreated leads to progressive disability and joint destruction. A combination of antiinflammatory agents, steroids, disease-modifying antirheumatic drugs, and biological agents are used to treat RA. Beyond the use of conventional measures of disease activity, such as American College of Rheumatology (ACR) response rates, the importance of patient-reported outcomes (PROs) in assessing therapeutic benefits is gaining increasing emphasis in clinical trials of RA and other chr

Humanized monoclonal antibodies are dramatically improving the therapy of lymphomas. The anti-CD20 monoclonal antibody rituximab is active in follicular lymphoma, with an objective response rate varying from 48% to 73%, but also in others under subtypes of B lymphoma. The association of the antibody to CHOP (R-CHOP) chemotherapy increases in diffuse large cells lymphomas by approximately 15% complete remission rate (63% vs 76%) and survival. The rituximab allows to eliminate lymphoma cells of the bone marrow and it is integrated in the strategies of autograft. These monoclonal antibody can be

Small cell lung cancer (SCLC) is a highly aggressive neuroendocrine tumour with a poor prognosis. Although treatment regimens such as first-line immunochemotherapy have shown clinical benefits in extensive stage SCLC (ES-SCLC), patients still face disease recurrence and drug resistance. Antibody-drug conjugates (ADCs) are new therapeutic compounds made up of a monoclonal antibody (mAb) attached to a cytotoxic drug (payload) using a linker. They achieve precise killing of tumour cells by conjugating highly cytotoxic drugs with monoclonal antibodies targeting specific tumour antigens. In recent

The malignant progression of cancer depends not only on oncogenic driver mutations but also on the adaptive rewiring of organelle stress responses that sustain cell survival under hostile tumor microenvironment (TME) conditions. Among these, the hijacking of lysosomal homeostasis has emerged as a critical vulnerability and a driver of therapeutic resistance. Glycoprotein nonmetastatic B (GPNMB), a highly glycosylated type I transmembrane protein predominantly localized to lysosomes, is robustly upregulated across multiple cancer types as an adaptive responder to lysosomal stress. In tumors, GP

BACKGROUND: Sacituzumab govitecan (SG) is an antibody-drug conjugate that has significantly improved survival outcomes versus standard of care across multiple studies in patients with previously treated metastatic breast cancer (mBC). Cancer treatment is associated with differing rates of treatment-emergent adverse events (TEAEs) based on patient race/location. We evaluated consistency and manageability of SG safety in mBC across multiple clinical studies and regions in this pooled analysis.
MATERIALS AND METHODS: Safety data for patients who received SG treatment (10 mg/kg, days 1 and 8 ever

PURPOSE: Rituximab resistance in B-cell non-Hodgkin lymphoma (RR B-NHL) involves decreased CD20 expression, limiting the efficacy of CD20-targeting radioimmunotherapy (RIT). Since the histone deacetylase inhibitor Chidamide enhances CD20 expression, we investigated its potential to improve the effectiveness of rituximab and a novel rituximab-based RIT in RR Raji cells.
METHODS: RR Raji cells were generated via one-week rituximab exposure, then treated with Chidamide for 24 h to upregulate CD20. Cell death was measured following exposure to a rituximab-serum mixture. Additionally, a radioimmun

Antibody-Drug Conjugates (ADCs) have become well-established as an important class of oncology therapeutics with 15 FDA-approved ADCs for various cancer types and hundreds more ADCs in clinical development. This chapter highlights the unique regulatory considerations for the development of ADCs in oncology given their dual nature as biologics and small-molecule drugs. Topics covered include the organization of applications in eCTD format, meeting types with FDA, and key considerations for a successful regulatory application with a particular emphasis on the Chemistry, Manufacturing, and Contro

Bladder cancer, particularly in its muscle-invasive and advanced urothelial carcinoma forms, has long been constrained by the therapeutic ceiling of platinum-based chemotherapy, with a substantial proportion of patients deemed ineligible for cisplatin due to comorbidities, underscoring an urgent unmet need for novel treatment modalities. The introduction of immune checkpoint inhibitors targeting the PD-1/PD-L1 axis marked a paradigm shift in the therapeutic landscape, solidifying immunotherapy(IO) as a foundational strategy. More recently, antibody-drug conjugates(ADCs) have emerged as transfo

Marginal zone lymphomas (MZL) are indolent lymphomas that may undergo transformation into aggressive lymphomas, most frequently diffuse large B-cell lymphoma (DLBCL). Loncastuximab tesirine (LONCA) is an antibody-drug conjugate used in the treatment of relapsed/refractory large B-cell lymphomas. Here we present the clinical case of a patient with nodal transformed DLBCL and concomitant bone marrow involvement by indolent lymphoma successfully treated with LONCA. However the therapy was discontinued after six cycles as the patient developed treatment-related bone marrow aplasia. This case raise

Abdominal aortic aneurysm (AAA) tends to occur in the lower segment of the abdominal aorta in the elderly population. Its continuously growth and rupture risk seriously threaten the life and health of patients. AAA molecular pathomechanism is complex, its core mechanism is a IL-6 mediated immune response regulation and inflammatory mediators release, and b) oxidative stress triggered the programmed death of endothelial cells and vascular smooth muscle cells (VSMCs). Thereby, the damage to the blood vessel walls continues to worsen. The treatment of AAA with traditional drugs often has off-targ

The expanding clinical use of Antibody-Drug Conjugates (ADCs) necessitates a clearer understanding of their real-world toxicity profiles across diverse clinical settings. This retrospective pharmacovigilance study analyzed 19,697 adverse event (AE) reports for seven approved ADCs from the FDA Adverse Event Reporting System (FAERS) database (Q1 2004 to Q1 2025). Using disproportionality analysis validated by multivariable logistic regression, we identified safety signals for both monotherapy and combination regimens. The analysis revealed that toxicity profiles are strongly driven by an ADC's m

Esophageal cancer remains one of the most lethal malignancies worldwide, with particularly poor outcomes following disease progression after first-line chemoimmunotherapy. Antibody-drug conjugates (ADCs) have emerged as a transformative therapeutic class that combines the targeting precision of monoclonal antibodies with potent cytotoxic payloads, enabling selective tumor cell killing while minimizing off-target toxicity. In the management of advanced esophageal cancer, I advocate for the integration of ADCs as a therapeutic option following progression on first-line chemoimmunotherapy. For pa

PURPOSE OF REVIEW: B-cell acute lymphoblastic leukemia (B-ALL) is a highly aggressive hematologic malignancy, with particularly poor outcomes in relapsed or refractory cases. Relapses remain a major challenge, often resulting in short remission durations and limited survival. This review summarizes emerging therapeutic strategies for B-ALL, focusing on novel immunotherapies and targeted approaches beyond the relapsed/refractory setting.
RECENT FINDINGS: Transformative therapies such as chimeric antigen receptor (CAR) T-cell therapy, antibody-drug conjugates (ADCs), and bispecific antibodies h
