Beyond Antiretroviral Therapy: Molecular and Immunological Innovations in HIV Treatment.

Despite prolonged viral inhibition with combination antiretroviral therapy (ART), HIV-1 survives as genetically intact, replication-capable proviruses within durable CD4+ T-cell fractions, involving central memory, transitional memory, and stem cell-like memory populations, as well as within tissue-resident compartments including lymphoid follicles and gut-associated lymphoid tissue. Reservoir stability is preserved via clonal growth of infected cells and epigenetic processes that impose proviral transcriptional silencing. As a result, current therapeutic approaches seek to either directly alt

Forging resilient warriors within: exercise's epic role in training innate immunity and taming inflammation's storm.

The concept of "trained immunity," which refers to a type of long-term immunological memory within innate cells, has significantly challenged the traditional division between the non-specific innate and antigen-specific adaptive immune systems. Simultaneously, it is now understood that the resolution of inflammation is not just a passive return to homeostasis but rather an active, strictly regulated biological process. This comprehensive review synthesizes these evolving concepts by hypothesizing that exercise, through its pleiotropic effects on cellular metabolism, may strongly stimulate both

OMIP-121: Immune Phenotyping of Canine Peripheral Leukocytes by Mass Cytometry.

Dogs are a critical non-rodent species used in preclinical safety studies, particularly, in the pharmaceutical field, due to their physiological, metabolic, and immunological similarities to humans. As such, immunophenotyping of canine peripheral blood mononuclear cells (PBMCs) plays a crucial role in translational research, immune monitoring, and safety evaluations in drug development. However, the limited availability of canine-specific antibodies restricts detailed and accurate immune profiling, which is essential for advancing safety evaluations in drug development. To address this challen

Immunological characteristics of children with autism spectrum disorder and comorbid atopic dermatitis.

OBJECTIVE: Current studies on immune dysregulation in autism spectrum disorder (ASD) have not adequately accounted for the influence of common comorbidities like atopic dermatitis (AD). To address this gap, the objective of this study was to characterize the peripheral immune profile in children with both ASD and AD, and to assess whether AD comorbidity modulates the link between immune dysregulation and ASD symptoms. METHODS: This single-center cross-sectional study recruited children with ASD who attended the outpatient clinic of Peking Union Medical Hospital (PUMCH) from April 2019 to Sept

Immunological characteristics of a recombinant alphaherpesvirus with an envelope-embedded Cap protein of circovirus.

INTRODUCTION: Variant pseudorabies virus (PRV) is a newly emerged zoonotic pathogen that can cause human blindness. PRV can take advantage of its large genome and multiple non-essential genes to construct recombinant attenuated vaccines carrying foreign genes. However, a major problem is that the foreign genes in recombinant PRV are only integrated into the genome for independent expression, rather than assembled on the surface of virion. METHODS: We reported a recombinant PRV with deleted gE/TK genes and an inserted porcine circovirus virus 2 (PCV2) Cap gene into the extracellular domain of

Feasibility and safety profile of intranasal third-party allogeneic CD45RO+/RA- memory T cells: a phase I clinical trial (vRELEASE-I NCT06699758).

BACKGROUND: Respiratory viral infections remain a major global health challenge, particularly in immunocompromised populations. Although adoptive cell therapies using virus-experienced memory T lymphocytes have demonstrated safety and effectiveness when administered intravenously, the safety profile and feasibility of intranasal delivery of immune effector cells in humans was not previously assessed. METHODS: vRELEASE I (NCT06699758) represents a phase I, single-center, open-label, dose-escalation study designed to evaluate the safety profile and feasibility of intranasal administration of of

Immunogenetics and immunology of transplantation in Leiden.

The historical observation of Jon van Rood that pregnancy can lead to the induction of HLA antibodies (1) was the start of a long history of research on immunogenetics and immunology of transplantation in Leiden. Some of the current research topics are presented in this mini-review. These include amongst others the differential immunogenicity of HLA mismatches in clinical transplantation, a special strategy to transplant highly sensitized patients, new tools to monitor donor specific memory B cells, new parameters for risk assessment in transplant recipients and the importance of the innate im

Regulatory Τ-cell differentiation between maternal and cord blood samples in pregnancies with spontaneous vaginal delivery and with elective cesarian section.

PROBLEM: The immunological mechanisms preventing fetal antigenic rejection during normal pregnancy and the extent to which the type of delivery influences lymphocyte reactions are elusive. METHOD OF STUDY: Maternal peripheral blood and neonatal umbilical cord blood (CB) was collected upon labor after vaginal delivery or cesarian section. Leukocytes were analyzed with flow cytometry, focusing on regulatory and γ/δ T-cells. RESULTS: In CB from neonates delivered by vaginal delivery, natural killer cells were increased. On the other hand, in maternal blood, γ/δ T-cells were increased, and activ

RES blockade: effects on immunity and tolerance.

Reticuloendothelial cell blockade has been studied for decades in regards to physiological and immunological effects. "Overloading" of RE cells with inert colloidal particles, such as carbon or other particulate substances, has often been used to analyze the role of phagocytic activities in antibody formation, often with contrasting results. In the present studies the effects of colloidal carbon treatment of mice on immunologic responsiveness to sheep erythrocytes was investigated. Pre-treatment of mice with carbon shortly before either primary or secondary immunization with SRBCs markedly sup

Role of mucosal chemokines in the development of tissue-resident CD4+ and CD8+ TRM cells to fend off herpes simplex infections.

The mucosal immune system represents the largest and most significant component of the immune network, providing critical defense against infectious pathogens at mucosal surfaces. Mucosal surfaces include the oronasal cavities, ocular surface, gastrointestinal tract, respiratory tract, and reproductive tract. Mucosal tissue-resident memory (TRM) CD4+ and CD8+ TRM cells serve as sentinels and critical mediators of adaptive mucosal immunity, continuously trafficking to mucosal tissues to surveil and clear invading pathogens. The development of mucosal CD4+ and CD8+ TRM cells is regulated by mech

CXCL16 as a potential therapeutic target to limit the activity of asthmatic CD4 memory T cells.

BACKGROUND: The C-X-C motif chemokine receptor CXCR6 is considered a marker of some tissue-resident memory T (Trm) cells, but its importance in controlling lung CD4 Trm cells in asthma remains unclear. OBJECTIVE: We explored the involvement of CXCL16-CXCR6 interactions in the activity and accumulation of CD4 Trm cells relevant to lung inflammatory disease. METHODS: Human and murine asthmatic lung CD4 T cells were analyzed by single-cell transcriptomics to assess CXCR6 expression, and the effects of blocking the CXCL16-CXCR6 pathway were then evaluated using a disease model. RESULTS: Single-

The Dual Role of Preexisting Immunity in Influenza: Protective Recall Versus Constraint of Novel Responses.

Preexisting immunity to influenza confers rapid protection against antigenically matched viruses but can also constrain responses to drifted variants. This review asks when and why prior infection or vaccination is protective versus detrimental, and which biological mechanisms-epitope masking by circulating antibody, inhibitory FcγRIIb signaling, and competitive dominance by affinity-matured memory B cells-drive these outcomes. We synthesize human cohort, serological, and vaccine-effectiveness data with mechanistic mouse and fate-mapping studies to explore how vaccine formulation, antigen dose

Basophil-specific deletion of interleukin 15 impairs memory CD8+ T cell homeostasis in the bone marrow.

Basophils develop in the bone marrow and are associated with allergic inflammation but also contribute to protective immune responses against pathogens. Upon activation, they produce large amounts of cytokines, including interleukin (IL)-4 and IL-13. However, their physiological functions during homeostasis remain poorly defined. Here, using single-cell RNA sequencing and cytokine knock-in reporter mice, we identify basophils as the immune cell population with the highest IL-15 expression among immature and mature bone marrow leukocytes. Conditional deletion of IL-15 in basophils using Mcpt8-C

Elevated memory T-cell conversion in a preclinical mouse model of hemophilia A.

One of the major challenges in the choice of the best therapeutic approach for the treatment of patients affected by hemophilia A (HA) is the definition of criteria predicting the formation of factor VIII (FVIII) neutralizing antibodies, called inhibitors. Both genetic and environmental elements influencing the immune response toward FVIII have been identified but still not all the factors causing the pathological rejection of FVIII have been identified. Since there is a connection between coagulation and inflammation, here we assessed the role played by the FVIII deficiency in shaping the hum

Chronic HCV infection promotes cytotoxicity in antigen-specific CD8+ T cells regardless of virus specificity.

INTRODUCTION: Despite advancements in hepatitis C virus (HCV) infection treatment, HCV still represents a significant public health burden. Besides progressive hepatic damage, viral persistence has lasting effects on innate and adaptive immune responses. Lack of a complete understanding of the factors driving an effective HCV response contributes to the failure to develop a vaccine for prevention. This study advances the existing knowledge on HCV-specific CD8+ T cells and describes the impact of current or past HCV infection on CD8+ T cells specific for other viruses. METHODS: We used barcode

Lymphangiogenesis-inducing vaccines elicit potent and long-lasting T cell immunity against melanomas.

In melanoma, the induction of lymphatic growth (lymphangiogenesis) has long been correlated with metastasis and poor prognosis, but we recently showed it can synergistically enhance cancer immunotherapy and boost T cell immunity. Here, we develop a translational approach for exploiting this "lymphangiogenic potentiation" of immunotherapy in a cancer vaccine using lethally irradiated tumor cells overexpressing vascular endothelial growth factor C (VEGF-C) and topical adjuvants. Our "VEGFC vax" induced extensive local lymphangiogenesis and promoted stronger T cell activation in both the intrader

Intraepithelial CD15 infiltration identifies high-grade anal dysplasia in people with HIV.

Men who have sex with men (MSM) with HIV are at high risk for squamous intraepithelial lesion (SIL) and anal cancer. Identifying local immunological mechanisms involved in the development of anal dysplasia could aid treatment and diagnostics. Here, we studied 111 anal biopsies obtained from 101 MSM with HIV, who participated in an anal screening program. We first assessed multiple immune subsets by flow cytometry, in addition to histological examination, in a discovery cohort. Selected molecules were further evaluated by immunohistochemistry in a validation cohort. Pathological samples were ch

Long-Lasting Antibody and CD8+ Memory T Cell Responses Induced by N-Tc52/TSKb20 Vaccination upon Trypanosoma cruzi Antigen Re-Encounter.

Background: Chagas disease, caused by Trypanosoma cruzi, remains a major public health problem in Latin America and an emerging concern worldwide. Current chemotherapies show limited efficacy during chronic infection, and no licensed vaccine is currently available. We previously developed the chimeric antigen N-Tc52/TSKb20 as a vaccine candidate against T. cruzi infection. In a murine model, this vaccine induced robust antigen-specific immune response associated with protection shortly after vaccination. Objectives: Here, we investigated the long-term persistence and effector functions of the
