COX-2-mediated inflammation in fat is crucial for obesity-linked insulin resistance and fatty liver.

The aim was to examine the role of cyclooxygenase (COX)-2-mediated inflammation in the development of obese linked insulin resistance and fatty liver. The rats were fed separately regular diet (CONT), high-fat diet (HFD) ad libitum, or energy restrictedly for 12 weeks. Rats fed HFD ad libitum were further divided into three subgroups co-treated with vehicle (HFa), or a selective COX-2 inhibitor celecoxib (HFa-Cel) or mesulid (HFa-Mes). Euglycemic hyperinsulinemic clamp (EHC) experiment was performed at the end of study. Another set of rats with similar grouping was further divided into those w

Nimodipine protects dopaminergic neurons against inflammation-mediated degeneration through inhibition of microglial activation.

Nimodipine, a calcium channel blocker, has been used mainly in the therapy of cardiovascular diseases. Recently, its indications have been extended experimentally to a wider range of disorders especially some central nervous system (CNS) disorders. In this study, we investigated whether nimodipine is neuroprotective to inflammation-mediated neurodegenerative diseases. Pretreatment with nimodipine reduced the degeneration of dopaminergic (DA) neurons induced by LPS in mesencephalic neuron-glia cultures in a dose-dependent manner. The neuroprotective effect of nimodipine was attributed to the in

CpG-ODN-induced inflammation is sufficient to cause T-cell-mediated autoaggression against hepatocytes.

Autoimmune diseases are often associated with microbial infections. Molecular mimicry between microbial antigens and self-epitopes has been suggested as a mechanism for breaking self-tolerance and induction of autoimmunity. Since infections also cause inflammatory responses we explored the role of local inflammation in organ-specific autoimmunity. For this purpose, transgenic mice were used expressing the MHC class I molecule Kb exclusively on hepatocytes. These mice exhibit Kb-specific tolerance as exemplified by the acceptance of Kb+ grafts. Inflammatory reactions were induced by injection o

Spino-bulbo-spinal pathway mediating vagal modulation of nociceptive-neuroendocrine control of inflammation in the rat.

Stimulation of nociceptors by intradermal capsaicin produces depression of bradykinin (BK)-induced synovial plasma extravasation (PE) that is markedly enhanced by subdiaphragmatic vagotomy. This depression is mediated by the adrenal medullae, a propriospinal pathway between the afferent nociceptive input and preganglionic neurones projecting to the adrenal medullae, and a spino-bulbo-spinal pathway. Here we investigated the role of spinal ascending and descending pathways in the interaction between noxious and vagal afferent inputs, leading to inhibition of BK-induced PE mediated by the adrena

Immune-mediated inflammation directly impairs pulmonary function, contributing to the pathogenesis of Pneumocystis carinii pneumonia.

The clinical severity of Pneumocystis carinii pneumonia (PCP) correlates closely with the appearance of pulmonary markers of inflammation. Therefore, a model system was developed whereby physiological studies could be performed on live mice to determine the extent to which pulmonary inflammation contributes to respiratory impairment during PCP. P. carinii-infected severe combined immunodeficient mice displayed little evidence of pulmonary inflammation and exhibited normal oxygenation and dynamic lung compliance. When comparably infected littermates were immunologically reconstituted, however,

Vitamin D status as a moderator of inflammation-mediated associations between adiposity and major adverse cardiovascular events: a cohort-based moderated mediation analysis.

BACKGROUND AND AIM: This study aimed to quantify the mediation of the adiposity-cardiovascular risk association by chronic low-grade inflammation and to test whether vitamin D status modulates this mediation. METHODS AND RESULTS: This prospective cohort study included 1828 adults without prior cardiovascular disease, recruited in 2016 and followed for 54 months. Baseline measurements included body fat percentage (BF%), serum 25-hydroxyvitamin D (25OHD), and high-sensitivity C-reactive protein (hsCRP). The primary endpoint was incidence of major adverse cardiovascular events (MACE). Associatio

PHB/ErGO electrochemical immunosensor for H5N1 serology: Insights into selectivity and inflammation-mediated matrix effects.

Reliable electrochemical immunosensing in biological samples requires not only selective molecular recognition but also preservation of analytical performance in complex matrices. Here, a label-free electrochemical immunosensor based on electrochemically reduced graphene oxide (ErGO) and polyhydroxybutyrate (PHB) nanoparticles was developed for the detection of anti-H5N1 antibodies in chicken serum. PHB nanoparticles were pre-functionalized with H5N1 hemagglutinin and deposited onto disposable ErGO-modified screen-printed electrodes, providing a simplified strategy for biointerface assembly. S

REV-ERBα deficiency exacerbates IL-33-mediated airway inflammation and airway hyperresponsiveness in mice.

Asthma involves type 2 airway inflammation driven by epithelial-derived interleukin-33 (IL-33), a key mediator of allergic responses. REV-ERBα, a circadian transcriptional repressor, modulates pulmonary inflammation, but its role in IL-33-induced airway pathology remains unclear. Here we investigated IL-33-mediated pulmonary inflammation in wild-type and global Rev-erbα knockout mice exposed intranasally to recombinant IL-33. Loss of REV-ERBα markedly increased recruitment of eosinophils, neutrophils, innate lymphoid cells, and Th2 lymphocytes, exacerbated airway hyperresponsiveness, and eleva

The dual role of capsaicin in inflammatory bowel disease: from TRPV1-mediated protection to neurogenic inflammation.

Capsaicin, the pungent vanilloid of chili pepper, is a widely consumed dietary constituent that exerts pleiotropic actions on physiological and pathophysiological processes, including anti-inflammatory, antioxidant, antiproliferative, metabolic and cardioprotective effects. Capsaicin is a small, highly lipophilic molecule that is not appreciably hydrolyzed by digestive enzymes in the gut lumen; it is absorbed across the gastrointestinal epithelium largely in its intact form, mainly by passive diffusion, and enters the portal circulation, where it undergoes extensive first-pass metabolism in th

Persistent Systemic Inflammation Mediates the Impact of Postoperative Complications on Survival After Gastric Cancer Surgery.

BACKGROUND: Postoperative complications (POCs) are associated with poor long-term outcomes in gastric cancer. However, the underlying biologic mechanisms remain unclear, and systemic inflammation may mediate this association. METHODS: This study retrospectively analyzed 4177 gastric cancer patients who underwent curative gastrectomy between 2013 and 2018. The neutrophil-to-lymphocyte ratio (NLR) was assessed preoperatively (pre-NLR), on postoperative day 2 (early NLR), and 3 months postoperatively (late NLR). Multivariable Cox and logistic regression models assessed the associations of NLR an

Selective Modulation of Th1-Mediated Inflammation and T Cell Metabolism via Shikonin-Loaded Nanogels.

Chronic inflammatory and autoimmune diseases are characterised by dysregulated Th1 and Th17 immune responses, leading to excessive production of proinflammatory cytokines. Shikonin (SK) is a natural anti-inflammatory compound whose therapeutic use is limited by poor solubility and bioavailability. Here, we evaluated SK-loaded hyaluronic acid-zein (HA-Zein) nanogels as a targeted strategy to modulate inflammatory CD4+ T-cell responses in vitro and in vivo. HA-Zein nanogels were designed to preferentially target CD44+ activated/effector T cells, key mediators of inflammation in Th1- or Th17-asso

Central Adiposity and Inflammation Mediate the Association Between Ultraprocessed Food Intake and Cardiovascular Disease Risk.

BACKGROUND: Higher consumption of ultraprocessed foods (UPFs) has been consistently associated with increased cardiovascular disease (CVD) risk, but the biological pathways underlying this relationship remain incompletely understood. OBJECTIVES: The study aims to examine whether body composition, systemic inflammation, and metabolic biomarkers mediate the association between UPF consumption and incident CVD. METHODS: MESA (Multi-Ethnic Study of Atherosclerosis) is a population-based, prospective cohort study of men and women aged 45 to 84 years who were free of clinically apparent CVD at bas

Second-order regulation: IFN-γ suppresses IL-17A-mediated type 3 inflammation.

BACKGROUND: T helper 1 (TH1) cells often accompany TH17 cells across diverse tissues in health and disease, including the lungs. However, roles for the TH1 effector cytokine, IFN-γ, in TH17-driven type 3 inflammation is unclear. METHODS: We devised a simplified reductionistic model to determine the role of IFN-γ in IL-17A-driven inflammation during Streptococcus pneumoniae (Spn) infection in vivo. Briefly, intratracheal instillation of Spn along with recombinant TNF-α and IL-17A was used to mimic rapid Spn-specific, TH17-driven, type 3 inflammation seen in lungs on memory recall infection wit

PFDoA induces developmental and immune toxicity in zebrafish through ROS-mediated inflammation-apoptosis signaling.

Perfluorododecanoic acid (PFDoA) is a persistent and bioaccumulative long-chain perfluoroalkyl acid, yet the mechanisms underlying its immunotoxicity remain poorly understood. This study investigated PFDoA-induced immunotoxicity in zebrafish embryos, demonstrating that 72 h exposure caused dose-dependent developmental toxicity (reduced survival, bradycardia, and shortened body length), markedly decreased neutrophil and macrophage populations, and induced thymic atrophy, indicating impairment of both innate and adaptive immunity. Transcriptomic analysis revealed enrichment of PPAR signaling, ap

Inhibition of calpain-mediated HMGB1 alleviates cardiac inflammation and dysfunction induced by ultra-processed foods.

Increased consumption of ultra-processed foods (UPFs) is a risk factor for metabolic disorder-associated heart failure (HF). Here, we demonstrate that UPF-induced calpain-1 aggravated oxidative stress, thereby increasing high mobility group box 1-mediated (HMGB1-mediated) myocardial inflammation, which contributes to cardiac dysfunction. After illustrating the dysregulated inflammatory pathways in human and murine hearts upon metabolic stress, we revealed an increase in calpain-1 alongside profound oxidative stress and inflammation in the failing myocardium. Mechanistically, in neonatal rat ca

Artemisinin derivative SM934 targets α-enolase to inhibit PEP-mediated TAK1 stabilization and inflammation.

TGF-β-activated kinase 1 (TAK1) is a key signaling hub and drug target in inflammatory responses. Although metabolism has been critically linked to immune cell function and inflammation, the metabolic control of TAK1 activation and intervention strategy remains to be explored. Here, we show that SM934, a derivative of the traditional Chinese medicine artemisinin, inhibits inflammatory responses via targeting α-enolase and inhibiting metabolite phosphoenolpyruvate (PEP) production. PEP directly binds TAK1 and inhibits its ubiquitination at the lysine-72 site, which promotes NF-κB activation and

Sophoridine alleviates spleen injury in sepsis by inhibiting ASK1-mediated inflammation, oxidative stress, and apoptosis.

Sepsis is a severe systemic inflammatory condition characterized by infection, and marked by high morbidity and mortality. Injury to the spleen resulting from sepsis disrupts immune defense functions, exacerbates systemic inflammatory dysregulation, and contributes to multiple organ failure, thereby substantially increasing patient mortality. Sophoridine (SR), a naturally occurring alkaloid derived from Sophora flavescens, exhibits anti-inflammatory, antioxidant and neuroprotective effects. This study investigated the protective effects of SR against sepsis-induced splenic damage and elucidate

The SP-MrgprX2 Axis in Allergic Rhinitis: a Neuro-immune Paradigm Beyond IgE-mediated Inflammation.

Allergic rhinitis (AR) is traditionally defined as an IgE-mediated hypersensitivity disorder; however, this paradigm fails to explain the clinical heterogeneity observed in patients with persistent symptoms and poor therapeutic responsiveness. Emerging evidence indicates that neuro-immune interactions constitute a critical, yet underappreciated, layer of AR pathogenesis. Substance P (SP)-Mas-related G protein-coupled receptor X2 (MrgprX2) axis has recently been recognized as a central mediator of IgE-independent inflammation. Upon sensory neuron activation, SP is rapidly released and directly

Berberine Stabilizes the Arrhythmogenic Substrate in Obese Rats by Klotho-Mediated Attenuation of Oxidative Stress and Inflammation.

Obesity increases susceptibility to ventricular arrhythmias due to an arrhythmogenic substrate by promoting oxidative stress and inflammation-driven cardiac remodeling. Klotho, an anti-aging protein that is reduced in obesity-related cardiovascular disease, protects against oxidative injury and inflammation. Berberine (BBR) has been demonstrated to have antiarrhythmic properties, but Klotho mediates these effects in obesity remains unclear. Here, high-fat diet (HFD)-induced obese rats were treated with BBR for 8 weeks. Surface electrocardiography showed BBR shortened prolonged QT, QTc, and Tp-

SIRT1/NRF2 axis regulates NLRP3-mediated macrophage inflammation and renal injury in hypertension with hyperuricemia.

BACKGROUND: Hypertension and hyperuricemia commonly coexist and are associated with persistent inflammation and renal injury. However, the mechanism linking these two conditions remains unclear. This study investigated whether the SIRT1/NRF2 axis regulates NLRP3-mediated macrophage inflammation in hypertension complicated by hyperuricemia. METHODS: Clinical data from 84 patients with hypertension and hyperuricemia and 84 patients with hypertension alone were analyzed. Circulating NLRP3 and inflammatory cytokines were measured, and the value of NLRP3 in identifying hyperuricemia occurrence and
