Paper-Based Analytical Biosensor Chip Designed from Graphene-Nanoplatelet-Amphiphilic-diblock-co-Polymer Composite for Cortisol Detection in Human Saliva.

Cortisol has been identified as a biomarker in saliva to monitor psychological stress. In this work, we report a label-free paper-based electrical biosensor chip to quantify salivary cortisol at a point-of-care (POC) level. A high specificity of the sensor chip to detect cortisol with a detection limit of 3 pg/mL was achieved by conjugating anticortisol antibody (anti-CAB) on top of gold (Au) microelectrodes using 3,3'-dithiodipropionic acid di(N-hydroxysuccinimide ester (DTSP) as a self-assembled monolayer (SAM) agent. The electrode design utilized poly(styrene)-block-poly(acrylic acid) (PS67

Transcriptional profiling reveals the role of Candida albicans Rap1 in oxidative stress response.

Candida albicans is a member of the human commensal microbiota but can also cause opportunistic infections, including life-threatening invasive candidiasis, particularly in immunocompromised patients. One of the important features of C. albicans commensalism and virulence is its ability to adapt to diverse environmental stress conditions within the host. Rap1 is a DNA-binding protein identified in yeasts, protozoa, and mammalian cells, and it plays multiple functions, including telomere regulation. Intriguingly, our previous study showed that Rap1 is also involved in cell wall integrity, biofi

Integration of Metabolomics and Transcriptomics Reveals Ketone Body and Lipid Metabolism Disturbance Related to ER Stress in the Liver.

Once protein synthesis is excessive or misfolded protein becomes aggregated, which eventually overwhelms the capacity of the endoplasmic reticulum (ER), a state named ER stress would be reached. ER stress could affect many tissues, especially the liver, in which nonalcoholic fatty liver disease, liver steatosis, etc. have been reported relative. However, there is still a lack of systematic insight into ER stress in the liver, which can be obtained by integrating metabolomics and transcriptomics of the tissue. Here, tunicamycin was utilized to induce ER stress in C57BL/6N mice. Microarray and u

Glucosamine and selenium differentially regulate glutathione, lipid peroxidation, and mitochondrial function during glutamate stress in retinal neural cells.

Glutamate can disrupt cystine-dependent glutathione metabolism and promote oxidative and mitochondrial dysfunction in neural cells. We integrated human multiple-sclerosis optic-chiasm transcriptomics (GSE100297), glutamate-treated primary-neuron RNA sequencing (GSE255041), R28 differential-expression summaries, a normal human optic-nerve single-nucleus atlas (GSE236566), and R28 retinal precursor-cell experiments to characterize glucosamine (GlcN) and selenium responses. Human tissue showed enriched unfolded-protein-response, oxidative-stress, apoptosis, and ferroptosis programs, whereas gluta

Alcohol binge drinking in adolescence and psychological profile: Can the preclinical model crack the chicken-or-egg question?

During adolescence, internal and external factors contribute to engaging with alcohol binge drinking (ABD), putting at risk the neurodevelopment of brain regions crucial for emotional control and stress coping. This research assessed the prevalence of ABD in late adolescent students of Southern Italy and characterized their psychological profile and drinking motives. Translational effects of alcohol binge drinking in the animal model were also studied. Seven hundred and fifty-nine high school students of both sexes (aged 18-20) were recruited. Alcohol Use Disorder Identification Test-Consumpti

Eradication of Helicobacter pylori infection in patients with intractable gastric ulcer.

BACKGROUND: Chromochloris zofingiensis, a freshwater alga capable of synthesizing both triacylglycerol (TAG) and astaxanthin, has been receiving increasing attention as a leading candidate producer. While the mechanism of oleaginousness and/or carotenogenesis has been studied under such induction conditions as nitrogen deprivation, high light and glucose feeding, it remains to be elucidated in response to salt stress, a condition critical for reducing freshwater footprint during algal production processes. RESULTS: Firstly, the effect of salt concentrations on growth, lipids and carotenoids w

Cyclin-dependent kinase 9-cyclin K functions in the replication stress response.

Cyclin-dependent kinase 9 (CDK9) is a well-characterized subunit of the positive transcription elongation factor b complex in which it regulates transcription elongation in cooperation with cyclin T. However, CDK9 also forms a complex with cyclin K, the function of which is less clear. Using a synthetic lethal RNA interference screen in human cells, we identified CDK9 as a component of the replication stress response. Loss of CDK9 activity causes an increase in spontaneous levels of DNA damage signalling in replicating cells and a decreased ability to recover from a transient replication arres

UVB-aged polystyrene microplastics induce enhanced stress responses in human proximal tubular cells.

Microplastics (MPs) are increasingly detected in human biological matrices, raising concerns about their potential systemic effects, including on the kidney. However, the cellular responses of renal tubular epithelium to MPs and the role of environmental aging processes in modulating their biological activity remain poorly defined. Under environmental conditions, MPs undergo photo-oxidative transformations that alter their surface chemistry and may influence their interactions with biological systems. In this study, we investigated the effects of 1 µm polystyrene MPs in virgin (MPsV) and UVB-o

Stress-related skin disorders.

Emerging research has suggested that the skin is not only a target of organismal distress but also an active participant of the stress response through production of local "HPA axis" components, peripheral nerve endings, and resident skin cells, including keratinocytes, mast cells, and immune/immune accessory cells. There are also bidirectional communication pathways between the brain and the skin, which play significant roles in integrating these interactions. In this review, we summarize the intricate relations between stress and several skin conditions. We have tried to identify the underly

Transcriptome and lipidome analyses reveal membrane lipid remodeling in root tissues of Cannabis sativa L. exposed to cold stress.

To construct a regulatory network model of the cold stress response in hemp roots, we used "Longdama 9" as the experimental material and integrated phenotypic, physiological, biochemical, lipidomic, and transcriptomic analyses to examine root responses to cold stress at 4°C. After 7 d of cold treatment, root growth was significantly inhibited, with root volume and fresh weight decreasing by 24% and 28%, respectively. Cold stress also induced membrane damage, as indicated by a 2.9-fold increase in relative electrical conductivity and a 3.8-fold increase in malondialdehyde (MDA) content. In resp

ATF4-dependent increase in mitochondrial-endoplasmic reticulum tethering following OPA1 deletion in skeletal muscle.

Mitochondria and endoplasmic reticulum (ER) contact sites (MERCs) are protein- and lipid-enriched hubs that mediate interorganellar communic … Additionally, loss of OPA1 increased the expression of activating transcription factor 4 (ATF4), an integrated stress response (ISR) pathway effector. Reducing Atf4 expression prevented the OPA1-loss-induced tightening of MERC structures. OPA1 reduction was associated with decreased mitochondrial and sarcoplasmic reticulum, a specialized form of ER, calcium, which was rever …

Deletion of the GntR8 transcriptional regulator impairs Brucella abortus intracellular survival and virulence by modulating stress response genes.

GntR transcription factors are emerging as critical regulators of bacterial metabolism, stress responses, and pathogenicity, however, their roles in the virulence mechanisms of Brucella abortus remain poorly understood. In this study, we generated a gntR8 (BAB_RS24500) deletion strain (ΔgntR8) in B. abortus 2308 and systematically investigated its role in virulence. The results demonstrate that deletion of gntR8 markedly impairs intracellular survival of B. abortus in RAW264.7 cells and significantly reduces virulence in a mouse infection model. Moreover, the ΔgntR8 strain exhibited increased

Crosstalk of the Caspase Family and Mammalian Target of Rapamycin Signaling.

Cell can integrate the caspase family and mammalian target of rapamycin (mTOR) signaling in response to cellular stress triggered by environment. It is necessary here to elucidate the direct response and interaction mechanism between the two signaling pathways in regulating cell survival and determining cell fate under cellular stress. Members of the caspase family are crucial regulators of inflammation, endoplasmic reticulum stress response and apoptosis. mTOR signaling is known to mediate cell growth, nutrition and metabolism. For instance, over-nutrition can cause the hyperactivation of mTO

Hypoxic and Thermal Stress: Many Ways Leading to the NOS/NO System in the Fish Heart.

Teleost fish are often regarded with interest for the remarkable ability of several species to tolerate even dramatic stresses, either internal or external, as in the case of fluctuations in O2 availability and temperature regimes. These events are naturally experienced by many fish species under different time scales, but they are now exacerbated by growing environmental changes. This further challenges the intrinsic ability of animals to cope with stress. The heart is crucial for the stress response, since a proper modulation of the cardiac function allows blood perfusion to the whole organi

Oxytocin, Vasopressin and Stress: A Hormetic Perspective.

The purpose of this article is to examine a previously unrecognized role for the vasopressin-oxytocin (VP-OT) system in mammalian "stress-response hormesis." The current review adds hormesis to the long list of beneficial effects of OT. Hormesis, a biphasic adaptive response to low-level stressors, is introduced here to contextualize the dynamic roles of oxytocin and vasopressin. As with hormesis, the properties of the VP-OT system are context-, time-, and dose-sensitive. Here we suggest that one key to understanding hormesis is the fact that VP and OT and their receptors function as an integr

Redox homeostasis and cellular stress response in aging and neurodegeneration.

Decreased expression and/or activity of antioxidant proteins leads to oxidative stress, accelerated aging, and neurodegeneration. While overwhelming levels and uncontrolled/dysregulated actions of reactive oxygen species (ROS) lead to deleterious effects, tighter regulation of those plays an important role in cell signaling. Mutations causing protein misfolding and the overload of toxic products derived from the free radical oxidation of polyunsaturated fatty acids, cholesterol, and glucose contribute to the disruption of the cellular redox homeostasis. Collectively or individually, these effe

Role of corticotropin-releasing factor pathways in stress-related alterations of colonic motor function and viscerosensibility in female rodents.

BACKGROUND: Clinical reports have shown that irritable bowel syndrome (IBS) is comorbid with anxiety/depression and stress-related events, and that the disorder is more prevalent among women than among men. In rodents, colorectal distention (CRD) induces abdominal contractions, and this visceromotor response is used to assess visceral pain. The activation of brain corticotropin-releasing factor (CRF) pathways has a key role in the behavioral and visceral responses to stress. OBJECTIVE: In this review of experimental studies that delineate the underlying mechanisms of the stress response, we f

The Alleviating Effect of Arginine on Ethanol Stress in Wickerhamomyces anomalus.

Yeast cells are passively exposed to ethanol stress during alcoholic fermentation, ultimately impairing cell viability and reducing fermentation efficiency. Arginine, a versatile amino acid, plays a crucial role in regulating cellular responses to various stresses. This study aimed to explore the underlying mechanism by which arginine protects Wickerhamomyces anomalus against ethanol stress. The effects of arginine supplementation (5 mM) under ethanol stress (9% v/v) on cell survival, reactive oxygen species (ROS) production, cellular and mitochondrial membrane integrity, and nitric oxide synt
