Two types of exocytosis evoked by ACh in salivary acinar cells: a morphological observation in perfused submandibular gland of rats.

Exocytosis in the perfused rat submandibular gland was studied using electron microscopy. Stimulation with 1 microM acetylcholine (ACh) evoked exocytosis; i.e., the secretory granules fused with the luminal membrane and their contents were extruded into the luminal space, and watery vacuoles were observed in some acinar cells. Stimulation with 1 mM ACh also induced exocytosis, and led to a marked increase in the number of watery vacuoles. Some watery vacuoles were connected to the lumen and were thus suggested to be formed by the fusion of the granules. On the other hand, no watery vacuoles we

Fibers, crystals, and other forms of HbS polymers in deoxygenated sickle erythrocytes.

We have examined by electron microscopy the formation of fibers and crystals from sickle hemoglobin within sickle erythrocytes following deoxygenation during capillary storage from 1 to 132 days. Intracellular fibers were found on the first day and throughout the period of study. The fibers exhibited a diameter (mean +/- SD) of 17.4 +/- 0.62 nm and were aligned in the cell with a fiber-to-fiber spacing of 18.6 nm (x-axis) by 22.7 nm (y-axis). Between 65 and 132 days, extracellular hemoglobin crystals developed, with a lattice periodicity of 9.63 +/- 0.6 nm. Fibers and crystals coexist as separ

Effect of PHA on viability and stress resilience in Rhodospirillum rubrum.

AIMS: Purple bacteria possess considerable biotechnological potential due to their versatile metabolism, enabling the production of valuable biopolymers such as polyhydroxyalkanoates (PHAs). PHAs are biodegradable biopolyesters that serve in microorganisms as intracellular carbon and energy sources and can also function as protective compounds against environmental stress. This study focuses on Rhodospirillum rubrum, a purple non-sulfur bacterium known for its metabolic flexibility and ability to accumulate PHAs. METHODS AND RESULTS: A wild-type strain (WT) Rhodospirillum rubrum (DSM 467) and

Potential roles of extracellular granzyme B in corneal and conjunctival diseases: A narrative review.

The cornea and conjunctiva are critical structures of the ocular surface that are prone to a range of inflammatory diseases impacting the vision of millions globally. Moreover, due to their complex etiology and pathophysiology, these conditions remain a significant clinical challenge. Granzyme B (GrB), a serine protease traditionally recognized for its intracellular role in targeting tumor or pathogen-infected cells, has been implicated in numerous pathogenic mechanisms observed in inflammatory corneal and conjunctival diseases, such as dysregulated inflammation and tissue remodeling, barrier

B cell receptor-mediated calcium signaling is impaired in B lymphocytes of type Ia patients with common variable immunodeficiency.

Several lines of evidence have demonstrated B cell intrinsic activation defects in patients with common variable immunodeficiency (CVID). The rapid increase of intracellular free calcium concentrations after engagement of the BCR represents one crucial element in this activation process. The analysis of 53 patients with CVID for BCR-induced calcium flux identified a subgroup of patients with significantly reduced Ca2+ signals in primary B cells. This subgroup strongly corresponded to the class Ia of the Freiburg classification. Comparison at the level of defined B cell subpopulations revealed

The Role of Extracellular Heat Shock Proteins in Cardiovascular Diseases.

In the early 1960s, heat shock proteins (HSPs) were first identified as vital intracellular proteinaceous components that help in stress physiology and reprogram the cellular responses to enable the organism's survival. By the early 1990s, HSPs were detected in extracellular spaces and found to activate gamma-delta T-lymphocytes. Subsequent investigations identified their association with varied disease conditions, including autoimmune disorders, diabetes, cancer, hepatic, pancreatic, and renal disorders, and cachexia. In cardiology, extracellular HSPs play a definite, but still unclear, role

Carbon nanotubes as nanovectors for intracellular delivery of laronidase in Mucopolysaccharidosis type I.

The immobilization of proteins on carbon nanotubes (CNTs) has been widely reported mainly for the preparation of sensors while the conjugation of enzymes for therapeutic purposes has scarcely been considered. Herein we report, to the best of our knowledge, the first example of intracellular delivery of a therapeutic enzyme by means of CNTs, retaining its activity. Mucopolysaccharidosis I is a rare genetic disease characterized by the deficiency or absence of the activity of the α-l-iduronidase (IDUA) enzyme. We evaluated the capacity of the recombinant form of the human IDUA enzyme, laronidase

[Gouty arthritis].

Gouty arthritis is the most common form of arthritis in men. As a consequence of persisting hyperuricemia, uric acid crystals are deposited in the intra-articular and periarticular spaces and activate the innate immune system. The clinically impressive abrupt onset monoarticular arthritis in the lower extremities is highly suggestive of a gout attack. Arthrosonography can be used for early detection of crystal deposition on joint cartilage. In synovial fluid the detection of uric acid crystals in polarization microscopy is proof of gout even without the detection of intracellular uric acid cry

Live-cell imaging of neurofilament transport in cultured neurons.

Neurofilaments, which are the intermediate filaments of nerve cells, are space-filling cytoskeletal polymers that contribute to the growth of axonal caliber. In addition to their structural role, neurofilaments are cargos of axonal transport that move along microtubule tracks in a rapid, intermittent, and bidirectional manner. Though they measure just 10nm in diameter, which is well below the diffraction limit of optical microscopes, these polymers can reach 100 μm or more in length and are often packed densely, just tens of nanometers apart. These properties of neurofilaments present unique c

Three-dimensional architecture of actin filaments in Listeria monocytogenes comet tails.

The intracellular bacterial pathogen Listeria monocytogenes is capable of remodelling the actin cytoskeleton of its host cells such that "comet tails" are assembled powering its movement within cells and enabling cell-to-cell spread. We used cryo-electron tomography to visualize the 3D structure of the comet tails in situ at the level of individual filaments. We have performed a quantitative analysis of their supramolecular architecture revealing the existence of bundles of nearly parallel hexagonally packed filaments with spacings of 12-13 nm. Similar configurations were observed in stress fi

Genetic variation in inflammasome genes is associated with outcome in bacterial meningitis.

Bacterial meningitis is a severe and deadly disease, most commonly caused by Streptococcus pneumoniae. Disease outcome has been related to severity of the inflammatory response in the subarachnoid space. Inflammasomes are intracellular signaling complexes contributing to this inflammatory response. The role of genetic variation in inflammasome genes in bacterial meningitis is largely unknown. In a prospective nationwide cohort of patients with pneumococcal meningitis, we performed a genetic association study and found that single-nucleotide polymorphisms in the inflammasome genes CARD8 (rs2043

Immunohistochemical study of dopamine in rat gastric mucosa with acute gastric ulcer.

Recent studies have shown the presence of dopamine (DA) in gastric and duodenal mucosa, and changes in gastric mucosal DA content have been observed in patient with acute ulcers. Immunohistochemical demonstration of the distribution of DA in gastric mucosa under stress was studied by light and electron microscopy. In the control group, DA was present in the gastric gland proper in the gastric corpus and antrum on light microscopy, and on the surface of mucous granules in chief cells, mucous neck cells, and surface epithelium on electron microscopy. In the stress group, DA in gastric mucosa was

Potassium and the heart.

The electrical stability of the heart is more sensitive to the extracellular than to the intracellular potassium concentration. During exercise, extracellular potassium varies rapidly. Catecholamines also modulate the plasma potassium concentration. Hypokalaemia of any cause can precipitate arrhythmias. Ischaemic myocardium loses potassium into the extracellular space within seconds and the cell becomes depolarized. The rise of the extracellular potassium ion concentration accounts for many of the early electrophysiological changes. Abrupt changes of plasma potassium concentration in normal my

Adrenergic control of Na+-K+-homoeostasis.

Catecholamines induce hypokalaemia via stimulation of beta-adrenoceptors, primarily in skeletal muscle but also in other tissues. This is the result of increased active Na+-K+-transport, leading to a rise in the intracellular K+/Na+-ratio and hyperpolarisation in muscle cells. These effects are mediated by 3', 5'-cyclic adenosine monophosphate, can be detected down to physiological concentrations of adrenaline and noradrenaline, and are seen both in vitro and in vivo. Catecholamines released from the adrenal medulla as well as sympathetic nerve endings are of importance in clearing K+ from the

Cell-Size Confinement Drives Size-Dependent Scaling of Intracellular Reaction-Diffusion Waves for Robust Patterning.

Intracellular reaction-diffusion (iRD) waves are a mechanism to set the positions of molecules within cells for their proper biological function. Despite their biological importance, the physical characteristics of iRD waves were still elusive due to the specific constraints of cells, where spatial confinement and surface-bulk coupling govern pattern formation. Here, by an artificial cell experiment of iRD wave for cell division (Min wave) using defined factors, we showed that the wavelength of iRD wave is selected by the space size. This wavelength selection adjusted the wave shape and speed

cAMP imaging of cells treated with pertussis toxin, cholera toxin, and anthrax edema toxin.

The enzymatic activity of the three most studied bacterial toxins that increase the cytosolic cAMP level: pertussis toxin (PT), cholera toxin (CT), and anthrax edema toxin (ET), was imaged by fluorescence videomicroscopy. Three different cell lines were transfected with a fluorescence resonance energy transfer biosensor based on the PKA regulatory and catalytic subunits fused to CFP and YFP, respectively. Real-time imaging of cells expressing this cAMP biosensor provided time and space resolved pictures of the toxins action. The time course of the PT-induced cAMP increase suggests that its act
