Use of D-STAT to prevent bleeding following percutaneous transhepatic intraportal islet transplantation.

An infrequent but nevertheless concerning complication associated with percutaneous transhepatic islet transplantation is bleeding. Historically in 61 procedures at this institution, we experienced four bleeding complications in three patients (6.6%), two requiring blood transfusion (3.3%) and two asymptomatic intraperitoneal bleeds detected sonographically at 24 h postprocedure (3.3%). It is suggested that the source of the majority of these bleeds is the liver parenchymal tract following removal of the infusion catheter combined with a significant dose of heparin administered to prevent port

The canine sympathetic neuropeptide galanin: a neurotransmitter in pancreas, a neuromodulator in liver.

Our laboratory has investigated the role of the neuropeptide galanin in the sympathetic neural control of both the canine endocrine pancreas and liver. Galanin mRNA and peptide were found in the neuronal cell bodies of the celiac ganglion, which projects fibers to both organs. Galanin fibers formed dense networks around the islets. Galanin was released from these nerves and the amount released appeared sufficient to markedly inhibit basal insulin secretion. We therefore propose that galanin is a sympathetic neurotransmitter in canine endocrine pancreas. Galanin was also found in hepatic nerves

An investigation of the relationship between duct system and A cell-rich and PP cell-rich pancreatic islets in the canine pancreas.

The pancreata of four six-month-old dogs of the same mother, two with both the pancreatic and accessory pancreatic ducts (X-type) and two with only the accessory pancreatic duct (Y-type), were examined in this study. To clarify the relationships between the type of pancreatic duct system and the composition of pancreatic endocrine cells, the pancreata were examined immunohistochemically using antiserum against four types of pancreatic hormones (glucagon, insulin, somatostatin and pancreatic polypeptide). In all areas of the X- and Y-type duct system pancreata, B cells accounted for 52-82% of t

The key role of islet dysfunction in type II diabetes mellitus.

Fasting plasma glucose levels are constant from day to day in normal individuals. This constancy is due to a close co-ordination between glucose production by the liver and glucose uptake in peripheral tissues. This review focusses on the key role of the endocrine pancreas alpha- and beta-cells to provide this co-ordination. Non-insulin-dependent diabetes mellitus (NIDDM) is characterized by fasting hyperglycemia. The degree of fasting hyperglycemia, in turn, is correlated with the basal rate of hepatic glucose production. This increased rate of glucose release by the liver results in part fro

[Some characteristics of the histological structure of the fetal bovine pancreas].

Some properties of histological structure of fetal bovine pancreas were demonstrated using light microscopic methods. The different forms of acino-insular complexes were described: 1) acino-insular complexes with single B-cells including epithelial layer of acini; 2) acino-insular complexes with segmental (sector) localization of insular cell groups; 3) acino-insular complexes with small and more large groups of endocrine cells timely contacted with acini; 4) acino-insular complexes at the stage of separation of endocrine cell groups (microislets) from acini. The consideration of acino-insular

Immunohistochemical study of the developing endocrine pancreas of the opossum (Didelphis virginiana).

Cells immunoreactive for insulin, glucagon, somatostatin, bovine pancreatic polypeptide and 5-hydroxytryptamine are found in the pancreas of the newborn opossum and of all later stages examined. All immunoreactive cell types are present in primary and secondary islets and within elements of the exocrine pancreas. Cells immunoreactive for glucagon, bovine pancreatic polypeptide, somatostatin and 5-hydroxytryptamine generally are confined to the periphery of secondary (intralobular) islets, whereas insulin-immunoreactive cells occupy the central region. Endocrine cells within primary (interlobul

Epidemiology of islet cell antibodies.

In spite of the potential assay problems, a relatively clear general picture concerning islet cell antibodies has emerged. They are undoubtedly the most potent indicator of risk for insulin-dependent diabetes, at least among high-risk persons (that is, first-degree relatives of insulin-dependent diabetics). Islet cell antibodies have been the focus of intense research scrutiny over the last decade, yet their etiologic role remains unclear. In patients with insulin-dependent diabetes, there is no striking relation at onset between islet cell antibodies and determinants such as age and HLA type.

[Ultrastructure of cells from the liver-endocrine pancreas system in hibernating animals].

In hibernated ground squirrel (Citellus erythrogenys) during various periods of hibernation morphological changes in organs participating in insurance of energoplastic homeostasis (liver, endocrine part of the pancreas) have been followed. At the beginning of hibernation certain signs of functional strain in the liver are observed-shortening and swelling of the endoplasmic reticulum (ER) canaliculi, decreases of glycogen resources and activation of the lysosomal apparatus, as well as certain features demonstrating its decreased activity--shortening cisterns in the Golgi complex, poorly manifes

An immunocytochemical and ultrastructural study of the endocrine pancreas of Sparus auratus L. (Teleostei).

The pancreatic endocrine cells of Sparus auratus (gilthead sea bream) are concentrated in two or three principal islets, or Brockmann bodies, and numerous smaller islets embedded in the exocrine tissue. Insulin-, glucagon-, somatostatin-, and pancreatic polypeptide (PP)-immunoreactive cells were identified in all pancreatic islets of S. auratus using an indirect immunocytochemical (PAP) method. Insulin-immunoreactive cells were found in the central region of the islets. Glucagon-immunoreactive cells could be seen at the periphery of the islets and isolated in the exocrine tissue surrounding th

Fetal endocrine pancreas.

The developing fetal endocrine pancrease produces both insulin and glucagon from an early stage in human development. These hormones do not cross the placenta and must be metabolized within the fetal-placental unit. Altered substrate metabolism in the gestational diabetic of the insulin dependent diabetic patient can pertubate normal fetal hormone homeostasis. Maternal hyperglycemia produces fetal hyperglycemia, hyperinsulinemia, and macrosomia while meticulous regulation of maternal glycemia abolishes the fetal macrosomia. Conversely, low levels of insulin or absence of fetal insulin results

[Immunocytochemical anatomy of alpha granules in the human endocrine pancreas].

Using the sensitive protein A-gold technique for the demonstration of antigenic sites in thin section for electron microscopy it was found that in the alpha granules of human pancreas, glucagon immunoreactivity (specific, C-terminal) is restricted to the dense granule core while glicentin immunoreactivity predominates on the peripheral halo surrounding the dense core.
