Models are developed that allow the effect of beta-lactam antibiotics on the level of apoptosis in human lymphoid cells to be evaluated in vitro. The antibiotics produced a dose-dependent decrease in apoptosis of mature lymphocytes both under normal conditions and in the state of dexamethasone-induced immunodepression. The beta-lactam antibiotics also affect the apoptosis in a pre-B-lymphoblast model. The dexamethasone-induced apoptosis is inhibited by beta-lactam antibiotics only in high doses.

Bacterial resistance to antibiotic is the inevitable consequence of the utilization of antimicrobial agents all over the world, particularly in developed countries. It is particularly evident with beta-lactam agents because they are among most frequently prescribed drugs. The resistance is mainly attributable to production of various types of beta-lactamases but other mechanisms like alterations in PBP molecules or in outer membrane proteins can play a significant role. Increased resistance can be seen among fastidious gram-negative bacteria like Haemophilus influenzae or Moraxella catarrhalis

Resistance to beta-lactam antibiotics of 183 clinical isolates belonging to Bacteroides, Porphyromonas and Prevotella was tested by a micro-broth dilution MIC method. Beta-lactamase production was screened by using nitrocefin sticks. Prevalence of resistance to different beta-lactam antibiotics was higher among the Bacteroides strains other than B. fragilis parallel with a beta-lactamase production (88% and 96%, respectively). Resistance was observed less frequently among Porphyromonas and Prevotella strains. Cefoxitin resistance was 11.5% and amoxicillin/clavulanic acid 3.5% among Bacteroides

The binding of five structurally diverse beta-lactam antibiotics to penicillin-binding proteins (PBPs) of two clinical isolates of Streptococcus pneumoniae resistant to penicillin G was compared with that of a susceptible strain. A common feature of the PBP patterns of the resistant strains was the absence of PBP 1a detected in the susceptible strain. For each beta-lactam antibiotic tested, there appeared to be significant decreases in the affinity for BPB 1b, 2a and 2b of the resistant strains. We attempted to evaluate a quantitative correlation between the antibacterial activity of the drugs

We have recently found a factor (Factor T) in aged mixtures of tungstate and phosphate which greatly enhances the antibacterial effects of beta-lactams on methicillin-resistant strains of staphylococcal species such as methicillin-resistant Staphylococcus aureus (MRSA), but shows only weak effects on methicillin-susceptible S. aureus and bacterial strains other than staphylococci. Factor T alone did not strongly inhibit cell metabolism and bacterial growth unless an excess amount was added. When Factor T was added to the culture medium beforehand, the growth of MRSA cells was rapidly suppresse…

An intestinal active transport system specific to small peptides and peptide-like drugs such as beta-lactam antibiotics was functionally expressed in Xenopus laevis oocytes after microinjection of messenger RNA (mRNA) derived from rat intestinal mucosal cells. The transport activity was evaluated by measuring the uptake of a tripeptide-like cephalosporin antibiotic, ceftibuten, which has high affinity for the intestinal peptide/H+ co-transporter and is resistant to peptidases. Ceftibuten transport in mRNA-injected oocytes was pH dependent (a proton gradient is the driving force), stereo select

Bacterial resistance to the beta-lactam drugs is extremely widespread, as a result of extensive drug use. Loss of susceptibility is primarily attributable to hydrolysis by inactivating enzymes, namely the beta-lactamases. While the number of characterised beta-lactamases may exceed 100, only a few are a problem in the treatment of community-acquired infections (TEM-1, TEM-2, SHV-1, BRO-1). Chromosomally mediated and extended-spectrum beta-lactamases are usually dominant in nosocomial pathogens where oral antibiotic therapy is seldom used. Therefore, the threat posed by beta-lactamases must be

We have studied the susceptibility to ampicillin and the characteristics of the beta-lactamase activity of the 169 Haemophilus spp. strains (128 H. influenzae, 40 H. parainfluenzae and one H. paraphrophilus) isolated during 12 months, years 1988-1989, in the Hospital del Mar clinical microbiology laboratory. Our objective was to know in the strains of our center the frequency of those resistant or slightly susceptible to ampicillin, the role of beta-lactamases in the loss of susceptibility and the type of enzymes involved. Susceptibility was studied by diffusion for all the antibiotics tested

The means by which gram-negative bacilli can resist newer beta-lactam antibiotics are reviewed. A first situation is generated by mutants which produce great amounts of a chromosomal cephalosporinase. These cells are present (frequency: 10(-4) to 10(-7] within bacterial populations such as Enterobacter cloacae, and multiply after selection during therapy by third-generation cephalosporins or monobactams. A second problem arose by 1985, with the sudden development of plasmid-mediated beta-lactamases markedly active against third-generation cephalosporins. Some recent molecules, such as penems,

Streptococcus agalactiae displaying resistance to penicillins, cephalosporins, and several other antibiotics have been isolated from the udders of dairy cattle receiving antibiotic exposure in the form of routine antibiotic infusions between lactations (dry cow treatment). Expression of penicillin resistance was induced by subinhibitory concentrations of penicillin, and growth in penicillin broth increased the ability of resistant strains to form colonies on penicillin agar. beta-Lactamase activity was not detected by standard assays of either resistant cells or cell lysates. However, resistan

The in vitro activity of cefpiramide, a new ureido cephalosporin antibiotic, was determined against 1128 gram-positive and gram-negative aerobic and anaerobic bacteria selected for resistance to ampicillin and cefazolin. Cefpiramide was less active on a milligram basis than cefotaxime, ceftazidime, moxalactam, and aztreonam against all of the members of the Enterobacteriaceae. Cefpiramide inhibited many Pseudomonas aeruginosa resistant to carbenicillin, piperacillin, and cefotaxime, but it was less active than ceftazidime and cefsulodin. Cefpiramide inhibited staphylococci and streptococci and

Four beta-lactam-resistant permeability mutants of Pseudomonas aeruginosa PAO503 were studied. The resistance phenotypes were correlated to changes within the lipopolysaccharide. Two of the mutants, PCC1 and PCC19, were shown to differentiate between beta-lactams on the basis of relative hydrophobicity. The more hydrophilic antibiotics were less effective at inhibiting these strains. This phenotype was correlated to the presence of mannose, in measurable quantities, in lipopolysaccharide isolated from these strains. The other two strains, PCC23 and PCC100, differentiated between cephem antibio…

Of three patients with Enterobacter mediastinitis treated with cefamandole, two failed initial treatment due to emergence of strains resistant to cefamandole and third generation cephalosporins. Extracts of the sensitive strains showed inducible hydrolysis of cephalothin; resistant strains (wild and laboratory derived) showed constitutive hydrolysis of cephalothin and cefamandole and bound but did not hydrolyze cefotaxime and latamoxef (moxalactam). In one mutant, increased latamoxef resistance appeared due to non-enzymatic mechanism(s). The Enterobacter strain from the patient who was treated

Reactivating legacy antibiotics offers a practical route to combat drug-resistant infections, but requires delivery systems that coordinate bacterial engagement with pathogen-triggered drug release. Here, we report DFC, a deferoxamine (DFO)-reprogrammed Fe-carbenicillin (Car) coordination nanoassembly that restores the activity of the legacy β-lactam antibiotic Car against MRSA. DFC was fabricated via DFO-mediated competitive coordination, transforming homogeneous Fe-Car networks into heterogeneous Car-Fe-DFO structures. Computational modeling and spectroscopic analyses showed stronger Fe3+-DF

Antibiotic tolerance enables bacterial survival during antibiotic exposure and can promote the evolution of resistance, thereby threatening therapeutic efficacy. However, the molecular mechanisms underlying tolerance and its relationship with resistance remain incompletely understood. In this study, we show that a class D β-lactamase-deficient strain of Shewanella oneidensis (∆blaA) is highly susceptible to β-lactams, but retains the ability to survive lethal antibiotic concentrations. Under β-lactam stress, ∆blaA cells form reversible spheroplasts that revert to rod-shaped morphology and resu…

Collective antibiotic resistance occurs when populations of bacteria survive antibiotic treatments that are lethal to individual bacteria, which affects the efficacy of drug therapies. An important mechanism of collective resistance against widely used [Formula: see text]-lactams is the production of drug-degrading [Formula: see text]-lactamases. Here, we integrate experiments with mathematical modeling to understand the collective survival of Escherichia coli challenged with cefotaxime (CTX). At near-lethal CTX concentrations, we observe complex dynamics, involving initial biomass growth due …
