Magnesium is an important, predominantly intracellular cation that is required for a wide variety of cellular processes. The mammalian kidney plays a key role in whole-body magnesium homeostasis, but the molecular and cellular mechanisms that underlie renal epithelial magnesium reabsorption are poorly understood. Traditional physiologic approaches have been severely hampered by the lack of a useful radioisotope of magnesium that can be used for tracer flux studies. The present review discusses physiologic insights gained from recent reverse-genetic studies that have identified a plethora of ge…

BACKGROUND: Eclampsia, the occurrence of a convulsion (fit) in association with pre-eclampsia, remains a rare but serious complication of pregnancy. A number of different anticonvulsants are used to control eclamptic fits and to prevent further convulsions.
OBJECTIVES: The objective of this review was to assess the effects of magnesium sulphate compared with phenytoin when used for the care of women with eclampsia. Magnesium sulphate is compared with diazepam and with lytic cocktail in other Cochrane reviews.
SEARCH STRATEGY: We searched the Cochrane Pregnancy and Childbirth trials register …

Lipid and protein peroxidations were investigated in female patients with magnesium deficit (MD), fibromyalgia (FM) and age matched controls: malondialdehyde and protein carbonyls (PC), as well as serum, leucocyte and erythrocyte magnesium (EMg) were assessed in 20 controls, 25 FM and 16 MD patients. MDA are unchanged in MD and FM. PC are significantly increased (P < 0.01) in FM. EMg is significantly decreased in MD. There is a slight, but not significant, negative correlation between PC and EMg, in controls and MD. Protein peroxidations are demonstrated in FM. Further studies are needed in MD…

Fifty cases of idiopathic epilepsy and 25 age and sex matched normal healthy controls were studied. There was a significant fall in serum magnesium levels in idiopathic epilepsy on day one and 4 of the seizures (P < 0.01) as compared to controls, and was not decreased significantly on day 7 of the seizures (P < 0.05). The fall was maximum in status epilepticus and severe epilepsy than in mild and moderate epilepsy. Serum magnesium took less time to come back to normal in mild and moderate epilepsy than in other types. This shows that serum magnesium does have a prognostic significance. Lesser …

Magnesium is an essential cation, involved in many enzymatic reactions, as a cofactor to adenosine triphosphatases. It is critical in energy-requiring metabolic processes, as well as protein synthesis and anaerobic phosphorylation. Serum Mg concentration is maintained within a narrow range by the kidney and small intestine since under conditions of Mg deprivation both organs increase their fractional absorption of Mg. If Mg depletion continues, the bone store contributes by exchanging part of its content with extracellular fluid (ECF). The serum Mg can be normal in the presence of intracellula…

Magnesium plays a critical role in numerous metabolic functions, including all reactions involving adenosine triphosphate, and is thus essential for the production and use of energy. Magnesium imbalances are common in hospitalized patients, with magnesium deficiency occurring in 20% to 65% of critically ill patients. This article details the homeostatic mechanisms regulating magnesium, the functions of magnesium, and the causes, manifestations, and treatment of both hyper- and hypomagnesemia. Indications and guidelines for the therapeutic uses of magnesium are also reviewed.

Male weanling rats were maintained on magnesium-deficient diet for 30 d and compared with pair-fed control rats fed magnesium-supplemented diet. Magnesium deficiency led to slow growth and finally to a significant decrease in body weight (P < 0.001) accompanied by a significant hypomagnesaemia, hypomagnesuria and hyperoxaluria (P < 0.001 in each case) in experimental rats as compared to the control rats. Magnesium deficiency altered the glyoxylate metabolism in the liver and kidney mitochondria by significantly decreasing glyoxylate oxidation (by 26 per cent in liver and 17 per cent in kidney)…

The serum level of magnesium and calcium was systematically measured in patients with gallstones before and after cholecystectomy. It was found that 60 percent of the operated patients suffered of different digestive syndromes in association with magnesium deficiency, while 40 percent of patients had the same complaints in association of magnesium and calcium deficiency. When magnesium and/or magnesium plus calcium was supplemented these syndromes could be decreased significantly. In the latter case, an optimal ratio of magnesium/calcium is needed in the supplementary therapy.

The author presents an account of results of the magnesium loading test and its method, from a group of 50 patients with cardiovascular diseases. The magnesium loading test reveals shortage of bodily reserves of magnesium (mg) by calculating the percentage of Mg retention. In a group of examined subjects the author found a high percentage of Mg retention (on average 68%) which suggests an unequivocal deficiency of bodily Mg reserves in this group of patients. At the same time changes of serum concentrations and urinary excretion of other investigated minerals are discussed. As compared with ot…

In 19 patients receiving a cytostatic therapy with epirubicine and cisplatinum, 31 times in total the renal loss of magnesium was determined using the colorimetric xylidile-blue-method. 16 patients, whose magnesium input postoperatively was equally well known, served as a control. In all these patients a negative magnesium balance could be demonstrated being significantly higher (p less than 0.01) in those receiving cytostatic therapy.

Magnesium (Mg) deficiency is a common finding in critically ill patients. Mg deficiency results primarily from gastrointestinal or urinary Mg losses, but malnutrition and decreased dietary Mg intake may hasten the development of Mg depletion. In our medical intensive-care unit, we have found hypomagnesemia in 65% of patients with normal serum creatinine concentrations. The prevalence of normomagnesemic Mg deficiency in critically ill patients may be even higher and may contribute to the pathogenesis of hypocalcemia, cardiac arrhythmias and other symptoms of Mg deficiency.

Thiazides and loop diuretics, facilitate the loss of Mg and K resulting in increased excretion in the urine. Although serum-K and serum-Mg values in patients receiving long-term treatment for hypertension or incompensated heart disease usually are normal, muscle-Mg and muscle-K contents are reduced in around 50% of these patients. Mg deficiency increases K loss and K/Mg deficiencies are frequently observed simultaneously. K repletion is often difficult if the accompanying Mg deficiency is not corrected simultaneously. The K/Mg loss from the muscles is accompanied by reduced concentration of Na

Glucose homeostasis was studied in rats fed diets containing 750,200, or 100 mg/kg Mg for 6 to 8 weeks, from the age of 4 weeks. Weight gain of the rats receiving 200 and 100 mg/kg diets was decreased. This resulted from both a lower food intake and reduced effectiveness of the ingested food. Fed or fasting plasma glucose levels were similar in the three groups. During an intravenous glucose tolerance test, the rate of glucose disappearance was higher in Mg 100 rats than in controls. By contrast, during an oral glucose tolerance test, the rise in plasma glucose was greater and more sustained i

Magnesium (Mg) is a vasodilator which may play a role in the regulation of blood pressure. The present study measured tissue Mg and calcium (Ca) levels in a hypertension model, the spontaneously hypertensive (SHR) rat, using the Wistar-Kyoto rat as a control. Mean serum Mg levels were normal in both types of rats. In SHR rats given H2O ad libitum, four of nine tissues (kidney, heart, lung, bone) had significant 6-16% decreases in Mg content (less than 0.025-0.0005). In SHR rats undergoing chronic saline diuresis, seven of nine tissues (liver, kidney, testis, heart, lung, spleen, bone) had sign…
