Impairment of object recognition, object location and spatial working memory performance in inbred RHA vs. RLA rats.

The inbred Roman rat strains, which have been selected and bred for their very high (RHA) or extremely poor (RLA) ability to acquire the two-way active avoidance task, constitute a bidirectional genetic model in which each strain differs from the other and/or from "standard" laboratory rat strains in a large number of behavioral and neurobiological phenotypes. Relative to Roman low-avoidance (RLA) rats, the Roman High-avoidance (RHA) rats exhibit increased mesolimbic dopamine function, social deficits, and impaired attention and cognitive flexibility, besides many other neurobehavioral phenoty

Concurrent Presentation of Memory-Related Odours and Sounds Nullified Sleep Reactivation Benefits.

Reactivation of recently acquired memories during sleep supports their longevity. Reactivation can be altered during sleep using odours or s … hat continued at least 10 s after odour offset. Overall, these findings highlight the complexity of memory consolidation during sleep when multiple memories and multiple cues are involved.

Dentate Gyrus Engrams in Fear and Reward: Mechanistic Principles, Critical Gaps, and Paths to Translation.

The hippocampal dentate gyrus (DG) has emerged as a cornerstone of engram research. While DG fear-based engrams have been extensively studied, revealing principles of allocation, consolidation, retrieval, and valence switching, engrams encoding context-reward associations, particularly those involving drugs of abuse, remain comparatively underexplored. This knowledge gap has critical implications for understanding addiction, depression, and other disorders involving dysfunctional reward processing. In this review, we first establish the DG's unique anatomical and functional properties that pos

Beneficial Effect of Minimal Interference on Item Memory But Not on Source Memory in Alzheimer's Disease.

OBJECTIVES: Research suggests beneficial effects of minimal interference on item memory in mild Alzheimer's disease (AD). We conducted a study to test whether these beneficial effects extend to source memory, i.e., the ability to remember the episodic context in which an information was previously acquired. METHOD: Twenty-six mild AD participants and 28 controls manipulated six objects or watched the experimenter manipulating six other objects. After immediate item recall ("what were the items?") and source recall ("by whom the items were previously manipulated?"), an interference or a minima

Targeting neuronal nitric oxide synthase and the nitrergic system in post-traumatic stress disorder.

RATIONALE: Current pharmacological approaches to treatment of post-traumatic stress disorder (PTSD) lack adequate effectiveness. As a result, identifying new molecular targets for drug development is necessary. Furthermore, fear learning and memory in PTSD can undergo different phases, such as fear acquisition, consolidation, and extinction. Each phase may involve different cellular pathways and brain regions. As a result, effective management of PTSD requires mindfulness of the timing of drug administration. One of the molecular targets currently under intense investigation is the N-methyl-D-

Deep brain stimulation, histone deacetylase inhibitors and glutamatergic drugs rescue resistance to fear extinction in a genetic mouse model.

Anxiety disorders are characterized by persistent, excessive fear. Therapeutic interventions that reverse deficits in fear extinction represent a tractable approach to treating these disorders. We previously reported that 129S1/SvImJ (S1) mice show no extinction learning following normal fear conditioning. We now demonstrate that weak fear conditioning does permit fear reduction during massed extinction training in S1 mice, but reveals specific deficiency in extinction memory consolidation/retrieval. Rescue of this impaired extinction consolidation/retrieval was achieved with d-cycloserine (N-…

Role of the hippocampus and amygdala in the extinction of fear-motivated learning.

Fear-motivated learning is at the root of phobias, panic, generalized anxiety and the posttraumatic stress disorder. This makes the inhibition of fear-motivated behavior a therapeutic desideratum in these diseases. The simplest way to accomplish this is by extinction, a procedure by which a given association between a conditioned stimulus or context (CS) and a fearsome event is replaced by a new association between the CS and the lack of the fearsome stimulus. This is a new learning for the subject and, in rats, it requires gene expression and protein synthesis both in the hippocampus and the

Intrinsic and synaptic plasticity in the vestibular system.

The vestibular system provides an attractive model for understanding how changes in cellular and synaptic activity influence learning and memory in a quantifiable behavior, the vestibulo-ocular reflex. The vestibulo-ocular reflex produces eye movements that compensate for head motion; simple yet powerful forms of motor learning calibrate the circuit throughout life. Learning in the vestibulo-ocular reflex depends initially on the activity of Purkinje cells in the cerebellar flocculus, but consolidated memories appear to be stored downstream of Purkinje cells, probably in the vestibular nuclei.

An Abrupt Transformation of Phobic Behavior After a Post-Retrieval Amnesic Agent.

BACKGROUND: Although disrupting the process of memory reconsolidation has a great potential for clinical practice, the fear-amnesic effects are typically demonstrated through Pavlovian conditioning. Given that older and stronger memories are generally more resistant to change, we tested whether disrupting reconsolidation would also diminish fear in individuals who had developed a persistent spider fear outside the laboratory. METHODS: Spider-fearful participants received a single dose of 40 mg of the noradrenergic β-blocker propranolol (n = 15), double-blind and placebo-controlled (n = 15), a

Motor Sequence Learning Under Sleep Deprivation.

Sleep deprivation (SD) compromises declarative memory encoding and subsequent consolidation, yet its consequences for the acquisition of new motor skill learning remain unknown. Forty-eight healthy adults underwent either one night of total SD or normal sleep and were tested the next morning using a multimodal approach combining psychomotor vigilance, subjective sleepiness, cortisol, brain-derived neurotrophic factor (BDNF), and corticospinal excitability, alongside a standardised explicit motor sequence learning task dissociating goal-based accuracy from movement-based execution speed. Recovery as a function of the degree of amnesia due to protein synthesis inhibition.

Retrograde amnesia following inhibition of cerebral protein synthesis has generally been explained as either a failure of consolidation or impairment of a retrieval mechanism. Major evidence for the retrieval hypothesis is provided by studies which utilize a reminder (usually footshock) to attenuate the effect of the protein inhibitor. To examine this question, mice were injected subcutaneously with anisomycin (1 mg/animal, 7 mg/animal, or 1 mg/animal every 2 hr x 7) and given one training trial in a passive avoidance box. All animals received a single retention test on each of four consecutiv

Trauma therapy: risks, side effects, and the forensic implications of treatment history.

Many victims of sexual violence develop trauma-related disorders, often requiring early trauma-focused psychotherapeutic treatment. Guideline-compliant treatments can effectively alleviate PTSD symptoms; the positive effects of recommended trauma-focused techniques are undisputed. Nevertheless, it is important to critically discuss potential risks. Several recent publications on effects of trauma therapy report no cause for concern, emphasising that trauma-specific treatment has no negative effects on the memories of patients who have experienced trauma. This line of reasoning overlooks the fa

Hippocampus muscarinic M4 receptor mRNA expression may influence central cholinergic activity, causing fear memory strengthening by peripheral adrenaline.

Adrenaline (Ad) strengthens contextual fear memory by increasing blood glucose, possibly enhancing hippocampus acetylcholine synthesis. Neve … subtype receptor down expression, which was reversed by Ad. This may be related to contextual fear memory consolidation or retrieval induced by peripheral Ad. Furthermore, the effect of Ad contextual fear memory might be due to increased hippocampus muscarinic subtype M4 expression, which may contribute to increased cholinergic activity in the central nervous system.

Impairments in Fear Extinction Memory and Basolateral Amygdala Plasticity in the TgF344-AD Rat Model of Alzheimer's Disease Are Distinct from Nonpathological Aging.

Fear-based disorders such as post-traumatic stress disorder (PTSD) steepen age-related cognitive decline and double the risk for developing Alzheimer's disease (AD). Because of the seemingly hyperactive properties of fear memories, PTSD symptoms can worsen with age. Perturbations in the synaptic circuitry supporting fear memory extinction are key neural substrates of PTSD. The basolateral amygdala (BLA) is a medial temporal lobe structure that is critical in the encoding, consolidation, and retrieval of fear memories. As little is known about fear extinction memory and BLA synaptic dysfunction

The Role of Sleep Quality, Trait Anxiety and Hypothalamic-Pituitary-Adrenal Axis Measures in Cognitive Abilities of Healthy Individuals.

Sleep plays a crucial role in cognitive processes. Sleep and wake memory consolidation seem to be regulated by glucocorticoids, pointing out the potential role of the hypothalamic-pituitary-adrenal (HPA) axis in the relationship between sleep quality and cognitive abilities. Trait anxiety is another factor that is likely to moderate the relationship between sleep and cognition, because poorer sleep quality and subtle HPA axis abnormalities have been reported in people with high trait anxiety. The current study aimed to explore whether HPA axis activity or trait anxiety moderate the relationshi…

Investigations on Rho/ROCK signaling in post-traumatic stress disorder-like behavior in mice.

Post-Traumatic Stress Disorder (PTSD) is a chronic condition that occurs in response to a traumatic event, and consequently, enhances the threat sensitivity. Rho/ROCK signaling has been implicated in the consolidation of fear memory, stress, depression, anxiety, and traumatic brain injury. However, its role in post-traumatic stress disorder remains elusive. Therefore, the present study was designed to explore the role of fasudil, a Rho/ROCK inhibitor, a mouse model of PTSD. Mice were subjected to underwater trauma stress followed by three situational reminders. Underwater trauma (UWT) signific

Sex-specific exploration patterns and noradrenergic modulation in object recognition memory: Role of the perirhinal cortex.

INTRODUCTION: Object recognition memory (ORM) relies on two processes: familiarity, mediated by the perirhinal cortex (PRH), and recollection, supported by the hippocampus (HIP). These processes are recruited depending on memory strength: brief exploration forms weak traces relying on PRH familiarity, whereas extended exploration produces strong traces requiring HIP recollection. Noradrenaline (NA) modulates these mechanisms via β2-adrenergic receptors (β2-AR), influencing memory strength and consolidation. However, whether this modulation differs between sexes under conditions generating weak

A Lack of Practice Effects on Memory Tasks Predicts Conversion to Alzheimer Disease in Patients With Amnestic Mild Cognitive Impairment.

The aim of the current study was to test the accuracy of practice effects, that is, improvement in test performance due to repeated neuropsychological evaluations, in identifying patients with amnestic mild cognitive impairment (a-MCI) at greater risk of conversion to Alzheimer disease (AD). For this purpose, we conducted a longitudinal study of 54 patients diagnosed with a-MCI at the first assessment and followed-up for 4 years. During this time, 18 patients converted to AD. Baseline and 6- to 12-month follow-up performances on a large set of neuropsychological tests were analyzed to determin

Autobiographical memory in children with Idiopathic Generalised Epilepsy.

Autobiographical memory involves the recall of both personal facts (semantic memory) and the re-experiencing of past personal events (episodic memory). The recall of autobiographical episodic details has been associated with a specific network, which involves the prefrontal and medial temporal lobes, in addition to posterior regions of the brain. Seizure activity has been previously shown to disrupt the consolidation of newly-learned information into long-term memory, but it is not yet known whether primary generalised seizures alone are also associated with deficits in the recall of autobiogr
