Metabolomics and proteomics reveal blocking argininosuccinate synthetase 1 alleviates colitis in mice.

To date, treating ulcerative colitis (UC) remains a significant challenge due to its complex etiology. In this study, metabolomics and proteomics analysis for multi-center cohorts reveal that both serum arginine levels and the rate-limiting enzyme argininosuccinate synthetase 1 (ASS1) are significantly elevated in UC patients. Exogenous arginine infusion and ASS1 overexpression exacerbate the pathological features of colitis in mice, while inhibiting or silencing ASS1 offers protection against experimental colitis. The induction of ASS1 is accompanied by increased levels of acetylated H3 and t…

Metabolomics analysis of acute lung injury induced by aortic dissection in mice.

Thoracic aortic aneurysm/dissection (TAA/D) is a complicated vascular disorder with galloping development and high mortality. Phenotype switching plays an important role in the pathological process of TAA/D. Previous studies indicated the potential correlation between the expression level of lncRNA RP11-465L10.10 and MMP9 involved in the development of TAA/D. Here, our results showed that RP11-465L10.10 and MMP9 were highly increased in TAD patient tissues, which was consistent in Ang II-induced vascular smooth muscle cell (VSMC) phenotype switching. However, the effects and underlying mechani

Stepping out of the dark: how metabolomics shed light on fungal biology.

Metabolomics, a critical tool for analyzing small-molecule metabolites, integrates with genomics, transcriptomics, and proteomics to provide a systems-level understanding of fungal biology. By mapping metabolic networks, it elucidates regulatory mechanisms driving physiological and ecological adaptations. In fungal pathogenesis, metabolomics reveals host-pathogen dynamics, identifying virulence factors like gliotoxin in Aspergillus fumigatus and metabolic shifts, such as glyoxylate cycle upregulation in Candida albicans. Ecologically, it highlights fungal responses to abiotic stressors, includ…

Native metabolomics identifies pteridines as CutA ligands and modulators of copper binding.

CutA, a conserved protein across all domains of life, has been suggested to be involved in copper tolerance in bacteria, though recent studies have questioned this association, leaving its biological role unknown. To clarify its function, we studied cutA knockout mutants from two phylogenetically distant species, Synechococcus elongatus PCC 7942 and Escherichia coli, using phenotyping combined with metabolomics. To investigate the interaction of CutA with potential substrates and cofactors, we performed a series of native metabolomics experiments with CutA and cell extracts from which we ident…

Integrative AI-metabolomics: a new frontier in diagnosing pulmonary tumor thrombotic microangiopathy.

Pulmonary tumor thrombotic microangiopathy (PTTM) is a rare, rapidly progressive malignancy-related pulmonary vascular condition that remains difficult to diagnose before death due to its nonspecific clinical and imaging features. Recent advances in metabolomics, particularly when powered by artificial intelligence (AI), offer promising opportunities for early detection by identifying distinct metabolic signatures. While AI-driven metabolomics has shown success in various cancers, its application to PTTM is challenged by the rarity of the disease, limited datasets, and privacy concerns. Innova…
